Zusammenfassung Es wurde ein dünnschichtchromatographischer und UV-spektrophotometrischer Nachweis des Thiopentals im biologischen Material ausgearbeitet. Die Dünnschichtchromatographie der Organextrakte erfolgte mit drei Systemen von unterschiedlichem Typ, die sich wegen ihrer guten Trenneffekte bewährten: Formamid/Benzol (I), Chloroform-Aceton 10∶1 (II), Äthylacetat-Benzol-Ammoniak 10∶10∶0,1 (III). Bei der Entwicklung im System I wurde zum Verankern des Formamides die Celluloseschicht Lucefol Quick® benutzt, für die Systeme II und III diente als Adsorbens die Silikagelschicht Silufol UV 254®. Das UV-Spektrum des aus dem Blut, Gewebe und Magen-Darminhalt isolierten Thiopentals wurde nach der papierchromatographischen Trennung im System Formamid/Benzol im neutralen und alkalischen Milieu aufgenommen und mit der auf gleiche Weise behandelten Testsubstanz verglichen. In allen Proben des vorgelegten biologischen Materials wurde neben dem Thiopental in unveränderter Form auch sein Abbauprodukt Pentobarbital nachgewiesen.

Abstract An estimation of Thiopental in biological material was established by thin-layer chromatography and UV spectrophotometry. Thin-layer chromatography of tissue extracts presents three different systems that proved to have good partition effects: formamide/benzene (I), chloroformacetone 10∶1 (II), ethyl acetate-benzene-ammonia 10∶10∶0,1 (III). In system I, formamide was used as a stationary phase for the development on cellulose-layer Lucefol Quick®; in systems II and III silica gel-layer Silufol UV 254® was used as adsorbent. UV spectrum of Thiopental isolated from blood, tissue, and gastrointestinal content was, after the paper-chromatographic separation in system formamide/benzene, registered in neutral and alkaline mediums and compared with that of standards similarly obtained. In tests of available biological materials Pentobarbital, a metabolite of Thiopental, was also found.
