    Notes: Summary Total immunoreactive glucagon (IRG) and immunoreactive glucagon of A cell origin (IRGa) were measured in the serum of normal, sham-operated and depancreatized rats, after the administration of three glucagon antagonists: insulin (5–200 mU/rat/h), somatostatin (SRIF; 100 μg/kg/h) and antiglucagon serum (AGS, enough to bind three times the calculated total amount of circulating IRG). Since no differences were noted between the responses of normal and sham-operated animals, the values were pooled and used as controls. Pancreatectomy caused a significant increase in serum glucose, IRGa and total IRG and a significant decrease in serum insulin. AGS and SRIF significantly decreased serum glucose in control, but not in depancreatized rats, even though SRIF caused a significant decrease of IRGa in all animals. SRIF significantly decreased plasma insulin in control rats, but did not modify total IRG secretion in either group. In control rats the minimum effective hypoglycaemic dose of insulin (5 mU/rat/h) may have decreased serum IRGa, but not total IRG. At higher doses (20 mU/rat/h) insulin stimulated glucagon secretion. In depancreatized animals, higher doses of insulin (200 mU/rat/h) were needed to lower serum glucose. On the other hand, a dose of 100 μU/rat/h was sufficient to lower the serum IRG. We conclude that although hyperglucagonaemia may contribute to the hyperglycaemia of the untreated depancreatized rats, the excessive secretion of glucagon is secondary to insulin insufficiency and that, at least in this animal model, the hypoglycaemic action of insulin is only minimally dependent upon its ability to suppress glucagon secretion.

    Notes: A vast array of cosmetic products are used routinely by consumers to alleviate a range of skin care problems. Consumer concern ranges from age-associated skin changes to complexion (e.g., spots and pimples) and dryness. However, a major cause for concern amongst European female consumers is skin dryness, which spans all age ranges and skin types. Utilizing instrumental procedures (Squametry and Image Analysis) the severity of skin dryness has been correlated with expert assessment and consumer perception of dryness. The sensitivity of these techniques enables one to follow the progression of dryness alleviation for a range of product forms (e.g., creams and lotions) as well as the purported mechanism of action of various ingredient systems (e.g., occlusivity, moisturization). While conventional cosmetic products are shown to be effective, these techniques can be used to aid in the assessment and formulation of more effective products.     Notes: Abstract: Intraspecific hybrid production from the polyethylene glycol induced fusion of yeast protoplasts was greatly increased when calcium propionate was included as the source of the requisite Ca2+. The use of calcium propionate, as opposed to the more commonly employed calcium chloride, resulted in substantially greater yields of hybrids from intraspecific fusions of protoplasts of Saccharomyces cerevisiae and Candida albicans. It is postulated that the ability of calcium propionate to enhance the fusion frequency is due to the anion binding to the etheric oxygen of PEG and potentiating the fusogenicity of the polymer.

    Notes: Electron-transferring flavoprotein from the rumen bacterium Megasphaera elsdenii is a heterodimer (Mr = 75 kDa) containing FAD as cofactor and functioning solely to mediate electron transfer between the prosthetic groups of other proteins. The enzyme was crystallized by the hanging-drop vapour-diffusion method using polyethylene glycol 4000 as precipitant. The crystals obtained belong to the space group P212121 with unit-cell dimensions of a = 58.75, b = 61.77 and c = 122.27 Å. Interestingly the crystals exhibit a low solvent content. Crystals diffracted to beyond 2.5 Å using synchrotron radiation.

    Notes: The keratinocyte expression of class II MHC antigens HLA-DR, DP and DQ was studied in 20 long-lasting allergic patch test reactions and 17 normal allergic patch tests reactions. No significicant difference in the expression of these antigens in the 2 groups was detected. No 1 allergen was responsible for the majority of long-lasting allergic patch test reactions. The immunological mechanisms which may contribute to long-lasting allergic patch test reactions are discussed with reference to HLA-DR Keratinocytes.

    Notes: : :We extruded a dry mix of 2 parts whey protein concentrate (80% protein) and 1 part cornstarch using water, 0.1 N HCl, or 0.2 M NaOH as the liquid, or adding calcium (1.69% w/w) to the dry mix with water as the liquid. The functionality and acceptability of textured whey protein (TWP) extrudates was determined. Ground beef patties containing up to 40% (w/w) of hydrated TWP extrudates made with base (TWPbase) were as acceptable to a consumer panel as all beef patties. Additionally, patties containing 40% (w/w) TWPbase incurred less cook loss and less diameter reduction after cooking, but were less cohesive than all beef patties.

    Notes: We previously showed the median duration of positive patch test reactions to nickel sulfate(5% pet) was 9 days, and defined as long-lasting (LLAPTR) the 14.3% of reactions that persisted for 17 Days or longer. The pathomechanisms of LLAPTR are unclear, but may involve either localized antigen persistence or abnormal down regulation of the cellular immune response. In this study, we compared (a) the nickel concentration and (b) the immunocytochemical nature of the local immune reaction, between biopsies from LLAPTR (n= 8) and normally resolving allergic patch lest reactions (NRAPTR) (n= 8) to nickel sulfate. The concentration of nickel in LLAPTR (median 0.8μg/g, μg/g, range 0.25–3.87 μg/g, mean 0.83μg/g, 95% CI 0 35–1.31) and NRAPTR (median 0.58 μg/g, range 0.2 1.85 μg/g, mean 0.88 μg/g, 95% CI 0.02 1.74) was similar. Activated T lymphocytes, expressing surface IL-2 receptor, HLA DR, DR alpha 1, DP, DQ, and CD2〉CD8〉CD4 antigens, were seen throughout the dermis and occasionally infiltrating the suprabasal layer of the epidermis in all biopsies. CDI and HLA DR, DR alpha 1, DP, and DQ-expressing Langerhans cells were present throughout the epidermis and occasionally seen in the papillary dermis. HLA DR, DR alpha 1, DP, and DQ antigen expression were also seen on the surface of non-dendritic cells in the epidermis (probably either keratinocytes or T lymphocytes) and vascular endothelial cells in the papillary dermis. There were no significant qualitative or quantitative differences in the immuno-cytochemical nature of the localized immune reaction between LLAPTR and NRAPTR. These findings suggest that the pathomechanism of LLAPTR to nickel sulfate is unlikely to be explained simply on the basis of nickel concentration or the nature of the localized immune reaction at the patch test site.
