    Notizen: Abstract Carbon monoxide adsorption has been investigated on Pt particles supported on a high surface area zirconia and sulfated zirconias. The accessibility of the Pt surface determined from the comparison of H2 chemisorption and transmission electron microscopy depends on two parameters: the temperature of treatment in air used to dehydroxylate sulfated zirconia, and the temperature of reduction. An oxidative pretreatment at 823 K yields a poor accessibility of Pt (0.03 〈 H/Pt 〈 0.05) whatever the temperature of reduction, whereas a Pt dispersion of 0.6 can be obtained by oxidation at 673 K followed by a mild reduction at 473 K. FTIR spectroscopy of adsorbed CO on Pt/ZrO2 shows besides the normal linear species at 2065 cm−1, a band at 1650 cm−1 which is attributed to CO bridged between Pt and Zr atoms. On Pt/ZrO2-SO 4 2− , all bridged species tend to disappear, as well as the dipole-dipole coupling andv CO is shifted by 57 cm−1 to higher frequencies. These results are attributed to sulfur adsorption on Pt which decreases the electron back-donation from Pt to the 2π * antibonding orbital of CO. The lower initial heat of CO adsorption observed on Pt/ZrO2-SO4/2− supports this proposal.

    Notizen: Summary The biolistic® particle delivery system was used for the delivery of DNA into embryogenic tissue culture cells of Pinus radiata D. Don. Several experiments with varying parameters were performed to increase the delivery efficiency. Six different controlling elements were cloned upstream of the ß-glucuronidase coding sequence (gusA reporter gene) and transient expression of the gusA reporter gene was compared three days after bombardment. The results clearly indicate a decrease in transient expression as follows: pEmu-derivatives with the ocs-enhancer-element 〉 2x CaMV 35S (with Kozak consensus-sequence) 〉 2x CaMV 35S (without Kozak consensus sequence) 〉 CaMV 35S (with Kozak consensus-sequence) 〉 CaMV 35S (without Kozak consensus sequence). Time course experiments monitoring gusA expression showed a significant decrease in the number of blue spots 10–14 days after bombardment. A few blue clumps however, were still detected 35 days after shooting. Embryo initials expressing the gusA gene in all cells were also detected. The results suggest that it will be possible to develop a reliable biolistic protocol for stable integration of genes into Pinus radiata embryogenic cultures which are capable of plant regeneration.

In angiosperms, the formation of the flower meristem is controlled by partially redundant flower meristem identity genes of which FLORICAULA (FLO)/LEAFY (LFY ) plays a central role. It is not known whether formation of reproductive organs of pre-angiosperm species is similarly regulated. Recently, a FLO/LFY-like cDNA, NEEDLY (NLY ), has been cloned in a conifer species Pinus radiata (D. Don). Here we report cloning of a different P inus radiata F LO/L FY-l ike cDNA, PRFLL. PRFLL had two large regions of high similarity to angiosperm FLO/LFY orthologues: amino acids 61–126 and 247–406 (50% and 81% identity, and 75% and 88% similarity, respectively, to LFY) and shorter regions of local similarity. Overall identity was 53% to LFY and 61% to NLY. Phylogenetic analysis of deduced protein sequences including partial LFY-like sequences from Pseudotsuga menziesii indicated that conifer proteins constituted a separate clade that could be divided into two groups represented by NLY and PRFLL. In contrast to angiosperms, both conifers had two paralogous proteins resembling LFY. Northern hybridisation analysis revealed expression of PRFLL in vegetative buds of juvenile, adolescent and mature trees. The transcript was not detected in vascular cambium, roots or secondary needles. To follow PRFLL expression during the early stages of cone development we analysed a temporal series of buds containing cone primordia, and developing cones, using Northern hybridisation and confocal microscopy in parallel. PRFLL mRNA was detected in buds from dominant and subordinate branches, in which cone and shoot primordia develop, and in developing male cones but not in developing female cones. Expression was particularly high in buds containing axillary primordia prior to their differentiation as male cone primordia. This is consistent with PRFLL being involved in determination of the male cone primordium identity.

    Notizen: Summary One descendant of the Medicago sativa Ra-3 transformant T304 was analysed with respect to the somatic stability of the synthetic phosphinothricin-N-acetyltransferase (pat) gene which was used as a selective marker and was under the control of the 5′/3′ expression signals of the cauliflower mosaic virus (CaMV) gene VI. In order to quantify gene instability, we developed a system for culturing and regenerating individual cells. Single cell suspension cultures derived from T304 and the ancestral non-transgenic M. sativa cultivar Ra-3, were established. The cells were regenerated into monoclonal calli. In transgenic calli, the phosphinothricin (Pt)-resistance phenotype was retained after more than 2 months of non-selective growth. In contrast, up to 12% of the suspension culture cells grown under non-selective conditions and at constant temperature (25° C) lost the herbicide-resistance phenotype within 150 days. Surprisingly, a heat treatment (37° C), lasting for 10 days, during the culture period resulted in an almost complete (95%) loss of the Pt resistance of the suspension culture cells. However, the frequency of cell division was identical in cultures grown under normal and heat treatment conditions. A biochemical test revealed that no phosphinothricin-N-acetyltransferase activity was present in heat treated, Pt-sensitive cells. The resistance level of the Pt-sensitive transgenic cells was equivalent to that of the wild-type cells. A PCR analysis confirmed the presence of the pat gene in heat treated, Pt-sensitive cells. From these results it is concluded that the Pt resistance gene was heat-inactivated at a high frequency in the M. sativa suspension cultures.
