    Notes: Summary A study was performed in 6 healthy volunteers to determine whether 50 mg intramuscular doses of meptazinol repeated four hourly for 24 h would lead to accumulation of the drug. The first dose of meptazinol was rapidly absorbed with a subsequent mean (± SD) elimination half-life of 1.6±0.33 h. This value was unaffected by the administration of intermediate doses. There was no significant accumulation of the drug; 95% of the steady-state plasma concentration was achieved after a mean (± SD) of 1.9±0.39 and 99% after a mean (± SD) of 2.9±0.63 doses. These findings are consistent with the theoretical prediction for a rapidly absorbed drug with a plasma elimination half-life of 1.6 h. No clinically relevant changes were seen in pulse blood pressure, haematological or biochemical screens. Reported side effects were of a subjective and minor nature.

    Notes: Summary The pharmacokinetics of indoramin (Baratol®) have been studied in five male and five female healthy, middle-aged volunteers after intravenous administration (0.14 mg/kg). Elimination occurred in an apparently biexponential fashion with a mean elimination half-life of 4.0h (±0.25 SEM). The mean plasma clearance was 19.9 ml/min/kg (±1.32 SEM) and the mean volume of distribution 7.4 l/kg (±0.81 SEM). There were no significant differences in these parameters between male and female volunteers. Protein binding of indoramin ranged from 85.6% at 81 ng/ml to 72.2% at 129 µg/ml. Two classes of binding site were evident, with affinity constants of 6.85×104M−1 and 4.30×103M−1.

    Notes: Summary Indoramin was administered as a single 50 mg oral tablet to 5 elderly (aged 68–71 years) and 6 middle-aged (aged 46–55 years) healthy female volunteers. The mean half life of indoramin in elderly subjects (14.7±4.8 h, mean±SEM) was statistically significantly longer than that seen in middle-aged subjects (5.1±1.0 h). The mean plasma concentrations of indoramin and mean area under the plasma concentration/time curve were also greater in elderly than in middle-aged subjects, although this did not achieve statistical significance. In many elderly patients, therefore, a reduction in dosage may be required due to the apparent reduction in clearance of indoramin. However, because of the wide inter-subject variability observed in the oral pharmacokinetics of indoramin, individual titration of indoramin dosage in all patients may be desirable.

    Notes: Aim : To compare the antisecretory effects of rabeprazole and esomeprazole in an open, randomized, two-way crossover, clinical pharmacology study.Methods : Twenty-four healthy subjects (14 men, 10 women; mean age 26.8 years) received rabeprazole 20 mg or esomeprazole 20 mg daily on days 1–5, witha 14-day ‘wash-out’. Intragastric pHwasrecordedcontinuously, and serum gastrin measured, on days 0, 1 and 5.Results : On day 0, mean intragastric pH AUC was significantly higher before the esomeprazole than before the rabeprazole treatment in four of the five time intervals analysed. On days 1 and 5, mean intragastric pH AUC was higher after rabeprazole than esomeprazole during 5–11, 14–24 and 0–24 h after dosing. Mean pH AUC in the first 5 h after dosing on day 5 was higher after esomeprazole than rabeprazole (P=0.012). On day 1, mean per cent times pH 〉 3 and 〉 4 were significantly greater after rabeprazole than esomeprazole during 0–14, 14–24 and 0–24 h. On day 5, mean serum gastrin AUC0-4 was higher (P = 0.017) after rabeprazole than esomeprazole (335 vs. 316 pg/mL.h).Conclusion : In this clinical pharmacology study, rabeprazole 20 mg daily was more effective than esomeprazole 20 mg daily in increasing intragastric pH and maintaining pH 〉 3 and 〉 4. On day 5, mean pH AUC was higher after esomeprazole than rabeprazole.

    Notes: Summary This study investigated the effects of damage by the typhlocybine leafhopperOssiannilssonola callosa on the physiology of sycamore seedlings. Feeding by 20 adult leafhoppers for four days produced stippling damage of about 10% of total leaf area. Measurements immediately following removal of the leafhoppers showed a 22% decrease in photosynthesis, 25% decrease in daytime rate of water loss and a 34% increase in night-time rate of water loss, probably due to impaired stomatal function. Sixteen days later control of stomatal function was largely regained with rates of photosynthesis similar to the control plants and slightly higher rates of water loss (6.6% day, 18.0% night) attributable to the still-open feeding punctures of the leafhoppers. Daytime SO2 uptake was similar in damaged and control plants but at night the damaged plants showed a 35% increase in uptake. The hypothesis that leafhopper damage is responsible for this increase is supported by a positive relationship between uptake rate and area of leaf stippled. The physiological effects of leafhopper feeding are greater than the visible damage suggests and the effects are particularly serious when the damage is fresh. Increased rate of air pollutant entry to damaged plants is also a problem, again most marked when damage is fresh.

    Notes: Summary Four species of Geometridae showed similar consumption rates and approximate digestibilities (100 x ingestion-egestion/ingestion) of sycamore leaves (Acer pseudoplatanus L.). A significant decline in E.C.I. (100 x growth/ingestion) and E.C.D. (100x growth/ingestion-egestion) with increasing larval weight is reported. The earlier descent from the sycamore canopy ofErannis defoliaria Cl. andOperophtera brumata L. larvae, to pupate in the soil, in comparison withAlsophila aescularia D. and S. andAgriopis aurantiaria Hb. larvae is discussed in relation to small differences in the utilization efficiencies of final instar larvae. Later descending species suffered greater predation by wood-ants in the field site studied.
