    Notes: The high-resolution two-dimensional protein gel electrophoresis technique combined with matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used to analyse the oxidative stress response in Staphylococcus aureus COL. Exponentially growing cells were supplemented with 100 mM H2O2 leading to a growth arrest lasting 30 min. The comparison of the two-dimensional pattern of cytoplasmic protein extracts of stressed and unstressed cells revealed only a few changes in the protein synthesis profile. However, the isoelectric points of Gap (glyceraldehyde-3-phosphate dehydrogenase), AhpC (alkylhydroperoxide reductase) and MvaS (HMG-CoA-synthase) changed strikingly. For analysis of the modification of Gap, tandem hybrid mass spectrometry (Q-Star) was used. The observed pI shift resulted from the oxidation to sulphonic acid of cysteine 151, which is crucial for catalytic activity. A drop in ATP and a complete inactivation of Gap was accompanied by the growth arrest. About 30 min after the addition of H2O2, the damaged Gap was still present, but a new protein spot at the original location became visible, representing the newly synthesized enzyme that is active again. This is accompanied by the restoration of Gap enzyme activity, ATP levels and recovery of growth. There is a strong correlation between growth, ATP level and Gap activity under oxidative stress conditions, indicating that the H2O2-triggered Gap inactivation might be one reason for growth arrest under these conditions. Our data indicate that the damaged Gap protein was not repaired.

    Notes: Zusammenfassung Es wird über Capillarresistenzuntersuchungen mittels eines modifizierten Sautestes im Zusammenhang mit der Chemotherapie der Lungentuberkulose berichtet. Als Ergänzung zu einigen in letzter Zeit veröffentlichten Beobachtungen über Resistenzabnahmen unter INH-Therapie werden Zahlen wiedergegeben, die sowohl diese Nebenerscheinung der Chemotherapie mit INH bestätigt, auf der anderen Seite jedoch in der überwiegenden Mehrzahl der Fälle von einer Resistenzzunahme unter INH berichtet. Auf die unterschiedliche Bedeutung der Untersuchung einer Arm- und einer Rückenteststelle, wird hingewiesen. Die Arbeit verfolgt den Zweck, vor einer einseitigen Betrachtungsweise zu warnen und die Aufmerksamkeit auf das noch wenig untersuchte Gebiet der Capillarreaktionen bei Lungentuberkulose, insbesondere unter Chemotherapie, zu lenken.

    Notes: Abstract The structures of the title compounds4 and5 have been determined by single crystal X-ray methods. Both molecules contain the approximately planar C4N2S2 group with 10 π-electrons which are mainly delocalized. The mean C—C bond distances in the four membered ring system are 146.1 (4) for4 and 143.8 (7) pm for5, with small individual variations either. In both molecules the C—S and C—N bond distances are 165 and 132 pm, respectively.4 has a crystallographic two fold axes,5 an inversion center. The packing mode of both molecules is very similar.

In alcaline solutions neutral complexes 5, 6 with the deprotonated ligand [NH2NCS2CH3]- or with its related Schiffbase anion [(CH3)2CNNCS2CH3]- (7) have been obtained. The structures of the two title compounds 3a and 7 have been determined by single crystal X-ray methods. The [Mn(CO)3 (NH2NHCS2CH3)2]+ ion contains one ligand coordinated as a chelate through N and S, in addition to a monodentate ligand, acting as N donor. The centrosymmetric dimer [Mn(CO)3 ((CH3)2CNNCS2CH3)]2 (7) contains a four-membered Mn2S2 core.
    Notes: Bei der Reaktion von Dithiocarbazinsäure-S-methylester (2) mit M(CO)5Br, M=Mn, Re, bilden sich in saurer oder neutraler Lösung ionische Carbonylkomplexe 3a, b, 4a, b. In alkalischem Medium entstehen Komplexe 5, 6 des deprotonierten Liganden [NH2NCS2CH3]-, oder nach Reaktion des Liganden mit Aceton der Komplex 7 der daraus abgeleiteten Schiffschen Base. Die Strukturen der Titelverbindungen 3a und 7 wurden röntgenographisch bestimmt. Das [Mn(CO)3(NH2NHCS2CH3)2]+-Ion enthält gleichzeitig einen chelatartig über N und S sowie einen monodentat über N gebundenen neutralen Liganden. [Mn(CO)3((CH3)2CNNCS2CH3)]2 (7) ist dimer und enthält einen Mn2S2-Vierring.

They crystallize in the form of hydrogen bridged dimers with the imino nitrogen as donor atom: 3 through NH … N, 4 through NH … O, and 2 through NH … S bonds. The hydrazide group in 3 displays E-conformation with respect to the C=S bond, in 4 with respect to the C=O bond. The ester groups are in the usual Z-conformation. In the solid state and in solution identical conformers are present as shown by spectroscopy.
    Notes: Die Thiocarbazinsäure-Derivate Thiocarbazinsäure-O-methylester (3), Thiocarbazinsäure-S-methylester (4) und das Thiocarbazinat-Ion (2) sind im festen Zustand planar. Sie bilden über das Wasserstoffatom der Iminogruppe zentrosymmetrische Dimere: 3 durch NH … N-, 4 durch NH … O- und 2 durch NH … S-Bindungen. Die Hydrazidgruppe besitzt E-Konformation in Bezug auf die C=S-Bindung von 3 bzw. auf die C=O-Bindung von 4. Die Estergruppen liegen in der üblichen Z-Konformation vor. Wie die Spektren zeigen, wird die Konformation des festen Zustands in Lösung beibehalten.
