    Notes: Abstract At the end of heat activation the distribution of spore plasma membrane particles between the two fracture faces (PF and EF) is drastically changed. While in dormant spores the particle number ratio of PF/EF was about 1:1, it increased up to 9:1 in heat activated spores, indicating a subtle change in plasma membrane properties. The permeability of spores increased within 30 min following heat activation as determined by efflux measurements of radioactively labelled spores. At the onset of swelling this efflux was accelerated. During germination the osmotically active material within the spores increased, part of which could be recovered from the supernatant. The combined experiments point to the plasma membrane as possible target site of heat activation in this system.

    Notes: Abstract Purpose. Cytidine-5′-diphosphate choline (CDPc) was encapsulated in long-circulating unilamellar vesicles (SUVs) to improve the drug's biological effectiveness. Methods. SUVs made up of diaplmitoylphosphatidylcholine/diaplmitoylphosphatidylserine/ cholesterol (7:4:7 molar ratio) and 8 mol % of ganglioside GMl were prepared by extrusion through polycarbonate filters (mean diameter 50 nm). The formulation effectiveness was evaluated by an in vivo model of cerebral ischemia on Wistar rats. Results. The enhanced delivery of CDPc into the brain improved the therapeutic effectiveness of the drug. CDPc-loaded SUVs improved the survival rate of ischemized and reperfused Wistar rats (320-350 g) by -66% compared with the free drug. Liposome formulation was also able to effectively protect the brain against peroxidative damage caused by post-ischemic reperfusion. SUVs lowered the conjugated diene levels of the cerebral cortex. The liposomal delivery system did not alter the distribution patterns in the various cerebral lipid fractions of the drug, radiolabeled with 14C-CDPc. Conclusions. CDPc-loaded SUVs were able to protect the brain against damage induced by ischemia. A possible clinical application is envisaged.

    Notes: Abstract. Our previous work has shown that localised activity of the cell-wall-loosening protein expansin is sufficient to induce primordia on the apical meristem of tomato, consistent with the hypothesis that tissue expansion plays a key role in leaf initiation. In this paper we describe the earliest morphogenic events visible on the surface of the apical meristem of tomato (Lycopersicon esculentum Mill.) following treatment with expansin and report on the spectrum of final structures formed. Our observations are consistent with a proposed primary function of expansin effecting morphogenesis via altered biophysical stress patterns in the meristem. The primordia induced by expansin do not complete the full program of leaf development. We present data indicating that one reason for this might be the inability of exogenous expansin to mimic the endogenous pattern of expansin activity in the meristem. These data provide the first detailed analysis at the cellular level of expansin action on living tissue, the first description of the spectrum of structures induced by expansin on the apical meristem, and give an insight into a potentially fundamental mechanism in plant development.

    Notes: Bilayers of human erythrocyte apoprotein-lipid complexes were made by dipping a mica plate through monolayers of the complex formed at the air-water interface. Stearic acid and erythrocyte lipid alone served as controls. Freeze-fracture images of the complex at high lipid surface pressures (30 dynes/cm) showed particles (average diameter, 109 Å ± 18 Å) similar to those of erythrocyte ghosts (average diameter, 102 Å ± 19 Å). Control surfaces were smooth. We conclude that part or all of the protein molecule penetrated into the lipid bilayer and that erythrocyte apoprotein-lipid complexes yield fracture faces similar to the native erythrocyte membrane.
