    Notes: Abstract First-motion analyses of digital seismic records and an assessment of damage provided conclusive evidence that a fault-slip source mechanism was responsible for a Richter magnitude 2 that occurred at the Lucky Friday Mine on April 13, 1990. This event was the first time that movement along an argillite bed had been observed after the macroseismic monitoring system had been installed. The physical evidence of a shear-slip type failure established confidence in using double-couple, first-motion analyses for the macroseismic system. The U.S. Bureau of Mines is conducting this research as part of its mission to enhance safety by reducing rockburst hazards in mines.

    Notes: Abstract The performance of an underground excavation at the Lucky Friday Mine that was subjected to repetitive episodes of mining-induced seismic activities is assessed. Field measurements indicate that an underground excavation may respond to mining-induced seismicity with step changes in displacement of the rock mass around the excavation or step changes in excavation closures. However, step changes in displacement or closure were also observed without the presence of mine seismic events. This behavior may be explained using the concept of stick-slip on joints or bedding planes within the rock mass. The stick-slip mechanism is believed to be controlled by the state of stresses at a joint or bedding plane. Through this mechanism, joint displacement is accumulated in a progressive fashion leading to a weakening of rock mass around an excavation. As a result, the excavation becomes less stable and therefore is more susceptible to seismic impacts. The concept of using peak particle velocity to assess damage threshold for underground excavations may not be conservative when considering the effect of rock mass fatigue.

    Notes: Abstract Stimulation of human neutrophils with zymosan particles resulted in a dose-response release of IL-8 in the 24 h culture supernatant, measured using a specific radioimmunoassay. IL-8 was detected after 8 h (0.11±0.08 nM) and peaked at 24 h (2.81±0.85 nM). Two distinct PAF receptor antagonists, WEB 2086 and UK 74505 produced a dose-dependent inhibition of zymosan-induced IL-8 release. This inhibitory action was maximal (68% 10−6 M WEB 2086; 71% 10−8 M UK 74505) when the drug was co-administered with zymosan, becoming progressively less effective when administered at increasing intervals after addition of zymosan. In contrast, a specific LTB4 antagonist, LY 255283, an IL-1 receptor antagonist and an anti-human TNFα antibody were unable to modulate the response indicating that these mediators are not involved. This study provides the first direct evidence of a central role for the phospholipid mediator PAF in the pathway of IL-8 release, following neutrophil activation by a particulate stimulusin vitro.

    Notes: Abstract Exudate fluids were removed from the rabbit peritoneal cavity 2 h after the injection of zymosan or the initiation of a reversed passive Arthus-type reaction. Inflammatory oedema was measured using Evans blue dye extravasation and the concentrations of C5a and the PGI2 metabolite, 6-oxo-PGF1α, were measured by radioimmunoassay. High levels of C5a were found in these inflammatory exudates and we suggest that the extravascular generation of C5a is important in the induction of increased microvascular permeability. The presence of PGI2 may have an enhancing effect on plasma protein leakage.

    Notes: Background Cytokines and chemokines produced by allergen-reactive T-helper type 2 (Th2) cells may be pivotal to the pathophysiology of allergic disorders.Objective This study was performed to assess the effect of 7 days of topical corticosteroid on nasal allergen challenge (NAC) in terms of eosinophils, cytokines and chemokines obtained by nasal lavage and filter paper methods.Methods Patients with grass pollen seasonal-allergic rhinitis (n=13) out of season received nasal challenge following matched placebo (twice daily into each nostril for 7 days) and fluticasone propionate (100 μg twice daily into each nostril for 7 days). Chemokine and cytokine levels were analysed using a sensitive automated bead immunoassay system at intervals up to 8 h after NAC.Results Levels of cytokines and chemokines from filter paper were generally higher than from nasal lavage. Fluticasone propionate caused a reduction in symptoms, total leukocyte counts and eosinophils, and abrogation of IL-4, IL-5, IL-6 and IL-13 responses in the filter paper taken in the late phase (P〈0.05 for IL-4 and IL-13, P〈0.01 for IL-5 and IL-6). Levels of chemokines (eotaxin, RANTES, MCP-1, MIP-1α, IL-8 and IP-10) were also reduced in the late phase (P〈0.01 at 8 h). However, levels of IL-2, IL-3, IL-7, IL-12 (p40 and p70), -15, TNF-α, IFN-γ and GM-CSF were not affected.Conclusion Fluticasone propionate has selective inhibitory effects on Th2 cytokine synthesis following nasal challenge, while also decreasing release of chemokines, but not affecting levels of Th1 cytokines.

    Notes: In previous studies we found that guinea pigs demonstrate an increase in airway reactivity and eosinophil numbers 4 days after a respiratory infection with parainfluenza-3 (PI3) virus. Clinical data support the possible involvement of eosinophils in virus-induced airway hyperresponsiveness. Eotaxin, a newly discovered chemokine, could be involved in eosinophil migration to the airways. In this study, eosinophil numbers were counted in blood and bronchoalveolar lavage (BAL) fluid and related with eotaxin concentrations in BAL fluid 1, 2, 3, and 4 days after intratracheal PI3 virus administration. On day 1, blood eosinophils increased by more than 200% (P 〈 0.01). The number of eosinophils were only slightly enhanced from day 2 to day 4 (40%–70%). BAL fluid eosinophils were not increased on day 1 but were significantly elevated on day 2 (180%) and remained high on days 3–4 (〉300%, P 〈 0.05). This increase in lung eosinophils correlated well with eotaxin levels measured in BAL fluid. There was no significant increase in eotaxin on day 1 following PI3 infection; however, on days 2–4 eotaxin levels in BAL fluid were significantly elevated (four–sixfold increase) when compared with medium inoculated controls. Eotaxin appears to play an important role in eosinophil accumulation in guinea pig lung following PI3 infection.
