    Notes: Abstract  The general characteristics of several graft-induced changes in pepper were investigated in a cross experiment. F1, F2, and BC1 progenies derived from crosses of the original stock and scion cultivars ‘Spanish Paprika’ and ‘Yatsubusa’, respectively, as well as their graft-induced variant strain G5S25 were analyzed for inheritance of the most conspicuous graft-induced variant traits. As part of a research program with the aim of revealing the mechanism of graft induction, the present study was carried out to examine the stability of the phenotypic changes and the characteristics of the graft-induced variants. For the fruit apex, a two-gene system was suggested, with other factors having a modifying influence. One of the two apex genes acted for pointed fruits and the other for inverted-blunt fruits. The inverted-blunt gene, the apex gene of the stock, was unambiguously present in the graft-induced variants, while the pointed gene that acted in the dominant mode in the original scion was inactive and expressed only under certain conditions in a mosaic state. The stable inverted-blunt cultivar used for the stock maintained certain factor(s) for pointed fruit, but the presence of that factor(s) could not be detected in graft-induced variants. The results of pungency analysis suggested a gene for non-pungency that appeared to be introduced in the graft-induced variants. The fruiting habit and fruiting direction that appeared in a mosaic state in graft-induced variants were found to combine factors of the stock with the appropriate characteristics of the scion asymmetrically. The bushy plant type appeared in a transgressive state in the variants, showing a definitely higher number of branches on the main stem and more frequent ramifications on the complete plant than on either the stock and scion cultivars or the progency derived from sexual crosses. A change in mature fruit color from red to yellow occurred in an early generation of graft-induced variants. Our results demonstrate that some of the characteristics of the stock were introduced into the progeny obtained from selfed seeds of the scion and that novel characteristics appeared as a result of graft induction.

    Notes: Summary An efficient chimera formation method by tissue culture combined with grafting was studied in Brassica. Cabbage cultivars “Ruby-ball” and “YR-ranpou” (Brassica oleracea) were used for intervarietal chimera formation. Seven-day-old seedlings leaving one of two cotyledons were approach-grafted and cultured in vitro (AGSC method). Chimeric shoots were obtained by the subculture of directly growing chimeric leaves (DGCL) from grafted part and cross-cut section of the united part after the graft culture. These were rooted and grown to complete chimera. An approach-grafted culture method was also available for interspecific chimera formation between “Komatsuna” (B. campestris) and “Ruby-ball” (B. oleracea), resulting in 20% formation per culture. Interspecific chimeras were identified as complex and peripheral-sectorial type by microscopic observation and the electrophoretic analysis of acid phosphatase isozyme pattern. The AGSC method was more effective than the usual Winkler's graft method and a simple mixed culture of heterogeneous cells or tissues.

    Notes: Summary To elucidate the genetic nature of bushy plant type in the variant strains derived from grafting, testcrosses were made on the one hand between one of the variant strains, G5S17, and both Yatsubusa and Spanish Paprika, and on the other hand between the two cultivars. The bushy strain has more nodes and branches on the main stem than the two cultivars. This bushy plant character was recessive to cv. Yatsubusa used as a scion and over-recessive to cv. Spanish Paprika used as a root stock. Such graft-induced change from recessive to over-recessive as bushy plant type was quite different from other varied characters such as, fruit shape, capsaicin content and guard cell size, which were generally intermediate between the two cultivars.

    Notes: Summary Capsaicin contents were measured in the progenies from the test crosses between the strain, G5S17 derived from fruit with the graft-induced change in fruit shape and the original two cultivars, Yatsubusa (Capsicum annuum L. var fasciculatum Ilish) and Spanish Paprika (C. annuum L. var. grossum Sendt) used for grafting. The strain, G5S17 contained a lower amount of capsaicin than the original pungent cultivar Yatsubusa used as scion. The characteristic of the lowered capsaicin content in the strain has been stably inherited through seed propagation, and has also been transmitted to the progenies from the crosses with the original two cultivars used for scion or stock. These features in the capsaicin content agreed with those in fruit shape in the graft-induced change.

    Notes: Summary Genetic changes induced by grafting were observed in soybean storage proteins, involving the 11S acidic subunit composition and Kunitz trypsin inhibitor. Changes from Gly 1 Gly 1to Gly 1 gly 1involved the 11S acidic subunit, and those from Ti 0Ti0 to Ti 3Ti0 involved the KTI in the first progeny (G1S1) in the scion (Gly 1 Gly 1 Ti 0Ti0, var. Kinzu) grafted onto var. Raiden (gly 1 gly 1 Ti 3 Ti 3). The progenies G1S2 and G2S1 from the seeds that had been changed segregated for both proteins. However, the segregation ratio in the progenies were different from those of the F2's from the sexual crosses between Kinzu and Raiden used for grafting. These findings show that the changes (changed genes) were transmitted to the progenies in an unstable manner.
