    Notes: Cells of Pseudomonas JM127 were grown in a medium containing mesquite wood as the sole source of carbon. The nutritional value of the microbial protein thus produced was determined. The protein contained more lysine and methionine than the FAO/WHO amino acid pattern and the requirement of infant, child or adult. The net protein utilization measurements indicated that the biological value of protein in the intact single cells was inferior to that of casein. The net protein utilization and nitrogen digestability were significantly improved when the cells were mechanically homogenized.

    Notes: Abstract A fluorescence quenching method was developed for the rapid determination of DNA and RNA using magdala red as fluorescence probe. In weakly acidic ¶medium, the fluorescence of magdala red (λex〉lem = 54055 nm) can be largely quenched by DNA or RNA. The calibration graphs are linear over the range 0.01–¶1.2 μg/mL for both calf thymus DNA (CT DNA) and salmon DNA (SM DNA), and 0.015–1.0 μg/mL for yeast RNA, respectively. The corresponding detection limits are ¶6.0 ng/mL for CT DNA, 7.0 ng/mL for SM DNA and ¶15.0 ng/mL for yeast RNA, respectively. CT DNA could be determined in the presence of 20% (w/w) yeast RNA, and the relative standard deviation of six replicate measurements is 3.18% for 400 ng/mL of CT DNA. Interference from coexisting substances in the determination of DNA was also examined. Real samples were determined with satisfactory results.

    Notes: Abstract A novel quantitative method for the determination of proteins in aqueous solutions has been based on the quenching of the resonance scattering light of colloidal silver chloride in the presence of proteins. The detection limits for eight kinds of proteins (BSA, HSA, egg albumin, human γ-IgG,α-chymotrypsin, E. Coli. alpsase, myoglobin, α-casein) were at about 8 ng/mL; the linear ranges of the calibration curves were 10–400 ng/mL ¶under optimal conditions,except for human γ-IgG (20–¶400 ng/mL), myoglobin (10–300 ng/mL), and α-casein (10–300 ng/mL). Three wavelengths (398 nm, 475 nm, 499 nm) were all suitable for the determination and any acidity from pH 3.0 to pH 9.0 could be chosen. A few non-protein substances at high concentration levels interfered with this method, but this problem could simply be overcome by diluting the samples before the assay. Mechanism studies showed that the quenching effect of proteins on the scattering light of colloidal silver chloride was mainly due to the coagulation of AgCl particles retarded by protein. The method was employed for the determination of total protein in human serum with sactifactory results.

    Notes: This research examined culture parameters influencing the rate of degradation of lignin in lignocellulosic substrates by the Basidiomycete Phanerochaete chrysosporium. Thermomechanical pulps prepared from western hemlock (Tsuga heterophylla) and red alder (Alnus rubra) were chosen as model substrates. Degradation of lignin in shallow, liquid-phase, stationary cultures was 10 times as rapid as in agitated cultures. Lignin degradation was at least 50% more rapid in cultures under 100% O2 than in those under air. Addition of 0.12% nutrient N (dry pulp basis) increased the rate of lignin degradation two- to fivefold; 1.2% added N at first suppressed, then stimulated, lignin degradation. Lignin in the alder pulp was degraded over five times as rapidly as in the hemlock pulp. Addition of glucose (35% of dry pulp) to the pulps containing 0.12% added N completely suppressed polysaccharide depletion during two weeks, but did not influence lignin degradation. The maximum rate of lignin degradation was 3%/day over a two-week incubation, or approximately 2.9 mg/mg fungal cell protein/day. The influence of the examined parameters was in complete accord with those found earlier for synthetic 14C-lignin metabolism by P. chrysosporium.

    Notes: We have measured the strain birefringence of Kevlar aramid fibers under static loading via a spectrophotometric method. At very low strain rates, the birefringence of Kevlar fibers increases markedly with increasing strain. On initial straining, the birefringence increases even at relatively constant modulus. This is attributed to the orientation of macroscopic species in the fiber which were observed by visible light microscopy. Such behavior is supported by the experimental observation of laser diffraction patterns and optical transmission images of Kevlar fibers under load. In the final stage of straining where the fiber modulus increases rapidly, the birefringence increase is attributable to crystalline orientation. The spectrophotometric method is useful for the simultaneous measurement of stress, strain, and birefringence of highly oriented, highly crystalline fibers such as Kevlar aramid. It is particularly useful to study the morphological inhomogeneity of a fiber which is undetectable by the conventional tensile test.
