    Notes: Abstract An engineered yeast with emission of fluorescence from the cell surface was constructed. Cell surface engineering was applied to display a visible reporter molecule, green fluorescent protein (GFP). A glucose-inducible promoter GAPDH as a model promoter was selected to control the expression of the reporter gene in response to environmental changes. The GFP gene was fused with the gene encoding the C-terminal half of α-agglutinin of Saccharomyces cerevisiae having a glycosylphosphatidylinositol anchor attachment signal sequence. A secretion signal sequence of the fungal glucoamylase precursor protein was connected to the N-terminal of GFP. This designed gene was integrated into the TRP1 locus of the chromosome of S. cerevisiae with homologous recombination. Fluorescence microscopy demonstrated that the transformant cells emitted green fluorescence derived from functionally expressed GFP involved in the fusion molecule. The surface display of GFP was further verified by immunofluorescence labeling with a polyclonal antibody (raised in rabbits) against GFP as the first antibody and Rhodamine Red-X-conjugated goat anti-rabbit IgG as the second antibody which cannot penetrate into the cell membrane. The display of GFP on the cell surface was confirmed using a confocal laser scanning microscope and by measuring fluorescence in each cell fraction obtained after the subcellular fractionation. As GFP was proved to be displayed as an active form on the cell surface, selection of promoters will endow yeast cells with abilities to respond to changes in environmental conditions, including nutrient concentrations in the media, through the emission of fluorescence.

    Notes: Abstract.  Laser-induced etching of polycrystalline Al2O3TiC material by a tightly-focused cw Ar ion laser has been investigated in a KOH solution with different concentrations. It is found that the KOH concentration can strongly affect the etching quality where low KOH concentration can result in rough and irregular patterns. Laser-induced etching of polycrystalline Al2O3TiC in a KOH solution is found to be a photothermal reaction in which a threshold laser power exists. With an appropriate set of etching parameters, well-defined grooves can be obtained with clean side walls and with an etching rate up to several hundred micrometers per second. The etching behavior is also found to depend on laser scanning direction. It is also found that the grains in the polycrystalline Al2O3TiC material play an important role in the etching dynamics and etching quality. This etching process is believed to be applicable to the formation of a slider surface of magnetic heads in the future.

    Notes: Omphacite and garnet coronas around amphibole occur in amphibolites in the Hong'an area, western Dabie Mountains, China. These amphibolites consist of an epidote–amphibolite facies assemblage of amphibole, garnet, albite, clinozoisite, paragonite, ilmenite and quartz, which is incompletely overprinted by an eclogite facies assemblage of garnet, omphacite and rutile. Coronas around amphibole can be divided into three types: an omphacite corona; a garnet–omphacite–rutile corona; and, a garnet–omphacite corona with less rutile. Chemographic analysis for local reaction domains in combination with petrographical observations show that reactions Amp + Ab + Pg = Omp +Czo + Qtz + H2O, and Amp + Ab = Omp ± Czo + Qtz + H2O may lead to the development of omphacite coronas. The garnet–omphacite–rutile corona was formed from the reaction Amp + Ab + Czo + Ilm ± Qtz = Omp + Grt + Rt + H2O. In garnet–omphacite coronas, the garnet corona grew during an early stage of epidote amphibolite facies metamorphism, whereas omphacite probably formed by the reactions forming the omphacite corona during the eclogite facies stage. It is estimated that these reactions occurred at 0.8–1.4 GPa and 480–610 °C using the garnet–clinopyroxene thermometer and omphacite barometer in the presence of albite.

    Notes: The Xugou garnet peridotite body of the southern Sulu ultrahigh-pressure (UHP) terrane is enclosed in felsic gneiss, bounded by faults, and consists of harzburgite and lenses of garnet clinopyroxenite and eclogite. The peridotite is composed of variable amounts of olivine (Fo91), enstatite (En92−93), garnet (Alm20−23Prp53−58Knr6−9Grs12−18), diopside and rare chromite. The ultramafic protolith has a depleted residual mantle composition, indicated by a high-Mg number, very low CaO, Al2O3 and total REE contents compared to primary mantle and other Sulu peridotites. Most garnet (Prp44−58) clinopyroxenites are foliated. Except for rare kyanite-bearing eclogitic bands, most eclogites contain a simple assemblage of garnet (Alm29−34Prp32−50Grs15−39) + omphacite (Jd24−36) + minor rutile. Clinopyroxenite and eclogite exhibit LREE-depleted and LREE-enriched patterns, respectively, but both have flat HREE patterns. Normalized La, Sm and Yb contents indicate that both eclogite and garnet clinopyroxenite formed by high-pressure crystal accumulation (+ variable trapped melt) from melts resulting from two-stage partial melting of a mantle source.Recrystallized textures and P–T estimates of 780–870 °C, 5–7 GPa and a metamorphic age of 231 ± 11 Ma indicate that both mafic and ultramafic protoliths experienced Triassic UHP metamorphism in the P–T forbidden zone with an extremely low thermal gradient (〈 5 °C km−1), and multistage retrograde recrystallization during exhumation. Develop of prehnite veins in clinopyroxenite, eclogite, felsic blocks and country rock gneiss, and replacements of eclogitic minerals by prehnite, albite, white mica, and K-feldspar indicate low-temperature metasomatism.

    Notes: Abstract When in vitro plantlets were cultured in Murashige and Skoog liquid medium supplemented with 8–10% sucrose and 22–44 μM 6-benzylaminopurine, all of the stem explants formed corms. 170–850 μM paclobutrazol increased corm formation, whereas 1700 μM paclobutrazol inhibited corm development. Inclusion of 66 μM 6-benzylaminopurine in 170 μM paclobutrazol treatment resulted in smaller corms, and bigger corms formed in the combination of 1700 μM paclobutrazol and 66 μM 6-benzylaminopurine. No corms formed in 63–630 μM cycocel treatments. In vitro corm growth was also affected by the culture methods. Deep-layer agitated culture yielded corms of up to 2.03 g, with an average fresh weight of 0.7 g, 40 days after induction. In thin layer cultures, corms were up to 1.87 g, with an average fresh weight of 0.5 g. SDS-PAGE analysis of water-soluble proteins revealed changes of polypeptides with corm growth. Compared to smaller ones, corms over 0.2 g had higher dry matter, carbohydrate and anthocyanin content. These corms had a 99–100% survival rate upon transplanting directly to soil after storage at 4 °C for 10 months. This study indicates that the most economic production method of diploid taro seed corm is by thin-layer liquid culture in Murashige and Skoog medium supplemented with 22–44 μM benzylaminopurine and 8–10% sucrose for 6 weeks. The formed corms can be stored at 4 °C up to 10 months and transplanted directly into soil without acclimatization.

    Notes: Conclusions 1. Molybdenum and vanadium improve the strength of alloy 2 (93 Ti, 5% Al and 2% Cr). The hardening of these alloys depends on the content ofβ -phase and the isomorphous stabilizers dissolved in it. 2. The stability of the investigated alloys is a function of the stability of the residualβ -phase. Molybdenum and vanadium, making up a considerable part of theβ -phase, greatly stabilize it. Niobium and tantalum are poor stabilizers. 3. Molybdenum and vanadium improve the heat resistant properties of alloy 2 at 400–500°.
