    Notizen: Deoxyhypusine synthase (DHS; EC 2.5.1.46) mediates the first of two enzymatic reactions that convert inactive eukaryotic translation initiation factor-5A (eIF-5A) to an activated form, thought to facilitate translation. A full-length cDNA clone encoding canola (Brassica napus cv. Westar) DHS was isolated from a cDNA-expression library prepared from senescing leaves. Transgenic canola lines with suppressed DHS expression were obtained by introducing a transgene expressing antisense 3′-UTR canola DHS cDNA under the regulation of the constitutive cauliflower mosaic virus 35S (CaMV-35S) promoter. Transformed seed was obtained by vacuum infiltration of canola inflorescences using the protocol developed for Arabidopsis with modifications. The resultant transgenic plants had reduced levels of leaf DHS protein and exhibited delayed natural leaf senescence. Suppression of DHS also increased leaf size by 1.5- to two-fold and resulted in increases in seed yield of up to 65%. Moreover, the enhanced performance of transgenic plants reflected increased tolerance to chronic sublethal stress. When wild-type and transgenic plants were grown in 6-inch pots, the increase in seed yield accruing from suppression of DHS was approximately 4.5-fold greater than when the plants were grown in 12-inch pots. Thus, suppression of DHS appears to ameliorate the effects of sublethal stress engendered by growth in small containers.

    Notizen: Abstract Highly purified plasma membranes were isolated by aqueous two-phase partitioning from rice (Oryza sativa) seedling roots. The effects of lanthanum chloride (LaCl3) on the activities of lipid peroxidation, the redox system and H+-ATPase, Ca2+-ATPase of plasma membranes were studied. The lipid peroxidation of plasma membranes could be depressed by certain low concentrations of LaCl3 and enhanced by high concentrations of LaCl3, while the lipid peroxidation was also dependent on the plasma membrane protein and incubation time. The relative activity of O2 uptake of plasma membranes was inhibited by all tested LaCl3 concentrations. In contrast, the reduction rate of Fe(CN)6 3− by plasma membranes was stimulated below 40 μM of LaCl3, but was reduced above 60 μM of LaCl3. The relative activities of both H+-ATPase and Ca2+-ATPase increased constantly from control to LaCl3 of concentration 60 μM where the activities of both enzymes were the maximum, but decreased remarkably at 80 μM LaCl3 concentrations various LaCl3 were added to culture solutions. In the other measurement case in which various LaCl3 concentrations were added directly to reaction medium and the plasma membrane vesicles only came from the control cultured rice seedling roots, the response of H+-ATPase activity to La3+ was similar to the response in culture solution. However, the La3+ concentration was only 20 μM when the activity of H+-ATPase was the maximum. In contrast to the case of LaCl3 addition to culture solution, Ca2+-ATPase activity was inhibited by all concentrations of La3+ which were added directly to the reaction medium. The above results revealed that REEs inhibited electron transfer from NADH to oxygen in plant plasma membranes, depressed the production of active oxygen radicals, and reduced the formation of lipid peroxides through plasma membrane lipid peroxidation. REEs ions also enhanced the H+ extrusion by both standard redox system and H+-ATPase in plasma membranes at certain concentrations. A possible role for the plant cell wall in REEs effects on plasma membranes was also suggested.

    Notizen: X-ray analysis on iron ores and reduced iron powders revealed that around 60% acidinsolublesubstances were hexagonal and tetragonal quartz, another 40% substances weresillimanite, alumina-silicate, an unnamed zeolite, all contained Si and Al. SEM images displayedthat the particle size of them was in the range of 3~7 μm, which may be the initial source of thecracking in the sintered body. Statistics analysis showed that the Acid-Insoluble Content (AIC) forhigh-grade magnetite powder was (0.130±0.010) % during the latest five months. The predictingvalue for reduced iron powder from ore powders should be 0.179 %. However, the testing value forreduced iron powder was (0.192±0.014) %. The limited difference of 0.013% might imply rarepollution coming from the reduction and milling processes. The most important step for controlAIC should be the separation process of iron ore powders
