    Notes: Abstract. Growth and cytology of root apical meristems of Chinese Spring wheat were studied in relation to temperature. The maximum rate of growth increased with temperature, a marked rise occurring between 10°C and 12°C. At all temperatures studied nucleolar volume increased to a maximum and then declined. The maximum nucleolar size achieved showed particular temperature sensitivity, higher volumes being attained at lower temperatures. The peak at 5°C was 70% higher than at 20°C. However, in comparison, cell and nuclear volumes were only 38% and 47% larger, respectively. Ultrastructural analysis of the nucleoli revealed a temperature-dependent relationship between the proportion of granular component and dense fibrillar component. The results are discussed in relation to the regulation of ribosome synthesis and function during growth and development at different temperatures.

    Notes: Crosses were made between the hexaploid wheat ‘Chinese Spring’ (2n = 42) and the diploid grain sorghum ‘S9B’ (2n = 20). Sixty-nine out of 100 florets fixed 48 h after pollination contained an embryo, an endosperm, or both, a remarkably high frequency in view of the taxonomic distance spanned by the cross. Percentages of single or double fertilization ranged from 50 % to 91 % for individual spikes. The hybrid origin of the embryos was confirmed by examining zygotes from spikes fixed 25 to 27 h after pollination. Seven of the 8 zygotes in which chromosomes were sufficiently contracted to be counted contained 21 large wheat chromosomes and 10 much smaller sorghum chromosomes. The eighth contained 21 chromosomes from wheat and 20 from sorghum. Sorghum chromosomes did not appear to be attached to the spindle in zygote nietaphases and showed no evidence of movement towards the spindle poles in the single zygote anaphase found. Embryos with two or more cells invariably contained one or more micronuclei and metaphases in embryos with three or more cells contained only 21 wheat chromosomes showing that sorghum chromosomes were rapidly eliminated. Endosperm, when present, was always highly abnormal.

    Notes: Abstract Serial sectioning of wheat roots prepared for electron microscopy was used to count the number of fibrillar centres per nucleus and nucleolus and to calculate their sizes. After growth at 35 ° C for two days the nucleoli became segregated and a one to one relationship was evident between chromosomal nucleolar organizers and fibrillar centres. This was confirmed using an aneuploid line carrying an additional pair of organizers. Quantitative studies showed that the fibrillar centres occupied a volume 0.24% of the total chromatin reticulum. From this it was calculated that only about 1/3 of the fibrillar centre material was likely to be nucleolar organizer chromatin. The other material was considered to be the protein revealed in silver staining studies. The importance of this was shown by its constant ratio to the size of all nucleoli in a given nucleus. — Evidence was found for the movement and fusion of organizer flanking regions during growth at 35 ° C. The number of junctions between chromatin and nucleolar organizers drops by about half giving one per organizer after segregation, and serial sectioning demonstrated such junctions in close proximity, an arrangement suggestive of incipient fusion.

    Notes: Abstract The positions of the genomes originating from each parent were analysed in root-tip nuclei of the mature, sexual F1 hybrid plant Hordeum vulgare (barley) x Secale africanum (a wild rye). The two genomes of the hybrid were identified in both spread and sectioned material by non-radioactive DNA:DNA in situ hybridization using labelled total genomic DNA from one parent as a proble and unlabelled total genomic DNA from the other parent to block non-specific hybridization. Complete three-dimensional reconstructions of sets of labelled sections enabled detailed analysis of the position of the genomes at interphase. The parental genomes lay in various non-intermixed configurations, including lateral and concentric arrangements. On spread preparations, the two parental genomes were found to be spatially separated throughout the cell cycle; the genome originating from H. vulgare tended to be located more centrally than that from S. africanum. This results show that the nucleus is spatially organized above the level of the DNA and chromosome at the genome level.

    Notes: Abstract The natural variation in chromosome size in root-tip and shoot apex meristems of Vicia faba has been studied. Chromosomes in main root-tips of one week old plants are 2–3 times larger than those in small lateral root-tips of 3 week old plants. While the nuclear DNA content remains constant, the nuclear RNA and nuclear histone contents show a positive linear correlation with chromosome volume. The DNA: histone ratio varies and is lowest in cells with large chromosomes. Nuclear volume and chromosome volume are not linearly related and changes in nuclear density therefore seem likely. A positive linear relationship between chromosome volume and mitotic index in colchicined squashes is demonstrated. The results strongly suggest that variation in chromosome size indicates a corresponding change in the rate of cell metabolism and may well reflect change in genetic activity.

    Notes: Abstract In monosomic additions of Aegilops sharonensis to Chinese Spring wheat (2n=42 wheat chromosomes + 1 homoeologous group 4 Aegilops chromosome known as 4S1), probably all functional gametes carried one Aegilops chromosome. Such preferential transmission is unusual in monosomic alien additions. Male and female meiosis seemed usually normal, but about 75% of embryo sacs and 28% of pollen grains were visibly abnormal near anthesis. Before the first gametophyte mitosis, up to about 13% of megaspores and pollen grains showed abnormalities usual in wheat aneuploids. However, in first mitosis, 50% of megaspores at metaphase and anaphase and 41% of pollen grains at anaphase and telophase contained acentric chromosome (or chromatid) segments of various sizes, up to about 32 in a cell, which were separated from the rest of the chromosome (or chromatid) by a gap or a thin Feulgen-positive thread. Such separated segments (SSs) are not normally seen in wheat and its aneuploids. The data indicat that alien and wheat chromosomes interact in meiocytes so that meiospores with the alien chromosome develop into normal gametophytes, but meiospores lacking the alien chromosome have SSs at first mitosis by whose loss or unequal distribution between daughter nuclei sterilizing deficiencies arise. Thus only gametophytes with the alien chromosome are competent.
