    Notes: Ag–Mn nanoparticles were prepared by advanced gas evaporation. Their composition corresponded to Ag0.89Mn0.11, and the particle size distribution was log normal. The particles demonstrated an unambiguous spin glass behavior with the temperature dependent magnetic susceptibility displaying a plateau at ∼25 K. The magnetic domains were limited by the size of the particles. © 1999 American Institute of Physics.

    Notes: A 2-week training period 2000 meters above sea level performed by 6 male elite Swedish runners influenced neither basal anabolic (total and non-sex hormone-binding globulin (SHBG)-bound testosterone (NST) and insulin-like growth factor-1 (IGF-1) nor catabolic (cortisol) hormones when comparing serum levels prior to and after the training camp. The anabolic vs catabolic hormone balance, expressed as the NST: cortisol ratio, also remained unchanged as well as SHBG and body mass. Thus, training at 2000 meters above sea level, often practised by elite runners to improve performance in competition at sea level, does not result in a catabolic situation after return to sea level, as measured by peripheral hormones. However, the adaptation to high altitude was associated with a slight (NS) decrease in testosterone as well as in anabolic vs catabolic balance as measured the third day at high altitude. Simultaneously, a decrease in subjective performance was claimed by the runners, but could not be shown by objective measurements. From day 3 to day 9 at high altitude, all runners claimed a subjective recuperation of performance. Total and non-SHBG-bound testosterone increased significantly from day 3 at high altitude to the first post-camp sea-level test. The results reflect the necessity of adaptation when travelling to races at different altitudes. The Swedish runners had significantly higher cortisol, total testosterone and NST levels compared with basal values of a group of 17 elite Kenyan runners living and training at high altitude. Since the NST cortisol and IGF-1 values were not lower, a catabolic state or malnutrition was not likely to be present. The results might reflect an adaptation to altitude or ethnic variations.

    Notes: Sixty fasciotomies of the deep posterior compartment were performed on 30 patients with longstanding bilateral medial tibial syndrome (mean duration 25 months). The initial symptoms occurred during sports activities in all cases. Only 2 of the athletes were able to continue sports at an ordinary activity level. The diagnosis was based on a typical history of longstanding exertional pain and clinical findings of tenderness at the postero-medial border of the tibia. Radiographs showed no abnormalities. Scintigrams were performed in 12 patients in whom stress fractures could not be excluded by radiographs and clinical examination. Conservative treatment failed to relieve the symptoms. At follow-up 34 months (range 6–85) after surgery, 95% of the men and 73% of the women were free of symptoms or improved compared with prior to surgery. Whereas 68% of the men were totally free of symptoms, the corresponding figure for the women was 36%. Ninety-five percent of the men and 82% of the women returned to sport after surgery. Seventy-four percent of the men and 54% of the women returned to an activity level similar to that prior to injury. We conclude that fasciotomy of the deep posterior compartment gives a good long-term result in patients with medial tibial syndrome when conservative treatment fails. There was no significant difference between the sexes.

    Notes: Concentric (CON) and eccentric (ECC) isokinetic knee extensor endurance, during 100 repetitive isokinetic contractions, were analysed in 32 consecutive patients with conservatively treated unilateral anterior cruciate ligament ruptured knees. There were 20 men and 12 women (mean age 34 years). The test was performed 6.8 years after the injury (range 4–14). The uninjured leg in each patient served as control. Lysholm subjective score, Tegner activity score and IKDC grading were used to relate the subjective knee function to muscle performance. Knee extensor endurance for concentric as well as for eccentric contractions were significantly lower in the injured side than the uninjured side. The magnitude of decrease in muscle performance was 7.6% for concentric (range-12 to 27%) and 15.3% for eccentric (range -9 to 50%) endurance. The eccentric injured/uninjured endurance ratio correlated significantly to the Lysholm subjective score. We conclude that the knee extensor endurance is decreased in patients with conservatively treated anterior cruciate ligament-ruptured knees. The eccentric endurance test seems to reflect the patients' subjective knee functioning.

    Notes: The natural course of 46 stress fractures of the tibia were studied in relation to sports activity, diagnosis and treatment in 28 recreational athletes and 13 elite athletes. Two major types of stress fractures were observed. Thirty athletes sustained a posteriomedial fracture of the tibia and claimed a sudden onset of exertional pain. The initial symptoms occurred during running or orienteering (n= 19), soccer (n= 4), workout or dancing (n= 3), triathlon, basketball, badminton and tennis (one each) and were similarity distributed between left and right leg. Scintigraphs showed Zwas type I/II lesions, while X-rays were negative in most cases. The patients were treated only with restriction in sports activities. The fractures healed within 8–24 weeks with excellent or good results. The remaining 11 athletes had anterior margin stress fractures with a slow onset of exertional pain and a long delay in diagnosis. These injuries occurred in 4 long-distance runners, 2 handball players, 2 dancers, 1 volleyball player, 1 triathloner and 1 decathloner. They were located in the nondominant left leg (in three cases bilateral). The fractures showed Zwas I or II lesions on scintigrams, but differed from the posteriomedial injuries on the typical V-shape appearence on X-ray. The patients had conservative treatment, including immobility and restriction in sports activities. Four of the patients were operated when not symptom-free after 12–34 months. All elite athletes (n= 5) in this group had to interrupt then-elite carreer. At follow up 24–60 months after the initial symptoms, only 1 patient was symptom-free. Thus, posteriomedial tibial stress fractures show a sudden onset and benign natural course, while anterior stress fractures should be treated with caution. The use of bone scintigraphy should be emphasized early when an athlete presents with exertional lower leg pain.

    Notes: The effect of short-term altitude training on sea-level physiological characteristics in elite runners was investigated. Seven middle-distance runners (6 men, 1 woman) belonging to the Swedish national team (mean age 23 years) spent 2 weeks of training at 2000 m above sea level in Kenya. Treadmill tests were performed before and 6 and 12 d after the altitude sojourn. Six other runners (4 men, 2 women) had a corresponding training sojourn at sea level in Portugal (control group). Ro of the runners (1 man, 1 woman) in the Kenya group were omitted from the study because of gastroenteritis. The maximal oxygen uptake (VO2 max; pretravel: Kenya group 212 and control group 188 ml · kg−0.75 - min−1), maximal treadmill time and oxygen cost of running were unchanged in both groups. The maximal oxygen deficit increased in all subjects after the Kenya sojourn (mean 19±6%). Heart rates during running at specified submaximal running velocities were lower post-altitude (Kenya group), but tended to be higher after sea-level training (control group). Maximal heart rate was unchanged in both groups. Perceived exertion (Borg) during submaximal running was lower post-altitude. Submaximal and maximal blood lactate and plasma catecholamine concentrations were not altered in any of the groups. Post-exhaustive plasma ammonia levels were decreased 12 d after altitude descent in the Kenya group. The results suggest an unchanged aerobic capacity in elite middle-distance runners after short-term training at moderate altitude. However, a change in the circulatory regulation during submaximal exercise was observed. Furthermore, anaerobic capacity improved but this bore no clear relation to lactate or ammonia metabolism.

    Notes: The magnetic properties of two-dimensional arrays of Fe particles with well-defined geometry, prepared by electron lithography from epitaxial Fe (001) films of thickness of 50 nm, have been studied. Circular particles with diameters of 0.6 and 0.4 μm and rectangular particles 0.9 μm×0.3 μm and 0.7 μm×0.2 μm, were positioned in square and rectangular lattices, respectively, with lattice constants about twice the particle dimensions. Samples were prepared with the lattices oriented along the 〈100〉 and 〈110〉 directions. Hysteresis curves were obtained in the field range ±2 T at temperatures between 30 and 300 K. The hysteresis curves are characteristic for single particles with a multidomain zero-field state. The magnetization and demagnetization processes are mainly governed by the geometrical shape of the particles and their orientation with respect to the crystal directions and the applied field. This implies that the high quality in the epitaxial Fe films is preserved during the patterning process. Magnetic images, obtained by magnetic force microscopy, support the conclusions drawn from the magnetization measurements. The circular particles showed a fourfold symmetry in their magnetic image, which is compatible with the expected domain structure in a disk with cubic crystalline anisotropy. © 1999 American Institute of Physics.

    Notes: The yeast Pityrosporum orbiculare belongs to the normal cutaneous flora but is also considered to be one of the factors that may contribute to atopic dermatitis (AD). In the present study we investigated the possibility that P. orbiculare can act with superantigen activity in AD. P. orbiculare-reactive T-cell lines (TCLs) were obtained after stimulation of peripheral blood mononuclear cells (PBMC) with P. orbiculare extract. T-cell receptor β-chain V-segment (TCRBV) usage was investigated using monoclonal antibodies and flow cytometry. We could not find any difference in TCRBV usage between AD patients (n = 10) and healthy controls (n = 5), either in fresh PBMC or in P. orbiculare-reactive TCLs. Compared with their original PBMCs the P. orbiculare-reactive TCLs showed a decreased usage of several TCRBVs, although increased usage of certain TCRBVs could be seen in some of the individuals. Further analysis of the CDR3-length polymorphism exhibited a shift in CDR3-length distribution, indicating oligoclonal expansion of T cells specific to different antigens in the P. orbiculare extract. In conclusion we have not found any evidence for superantigen activity in P. orbiculare extract, but our data support the importance of classical major histocompatibility complex (MHC)-restricted allergens in P. orbiculare.

    Notes: Background Atopic dermatitis (AD) is associated with increased levels of serum igE. and T-helper (Th) cells are thought to a play role in the pathogenesis. Individuals with AD often develop IgE antibodies against the yeast Pityrosporum orhiculare. a member of the normal cutaneous flora.Objective The role of P. orbiculare in atopic dermatitis was investigated by examining the T-cell reactivity for P. orbiculare.Methods Freshly isolated peripheral blood monoruclear cells (PBMC) were isolated from 10 AD patients with serum IgE antibodies against P. orbiculare, and from six healthy controls. The proliferative response after P. orbiculare stimulation, measured by [3H]thymidine incorporation, was examined in the PBMC and in T-cell clones (TCC) obtained from skin and blood of one patient. The cytokine profile of the TCC was determined by enzyme-linked immunosorhent assay (ELISA). radioimmunoassay (RIA) and reverse transcriptase-polymerase chain reaction (RT-PCR) following challenge with either P. orbiculare extract or anti-CD3 antibodies and phytohaemag-glutinin.Results The PBMC response to P. orbiculare was significantly higher in the AD patients than in the control group (P 〈 0.05). Twenty-nine out of 36 tested TCC derived from one responding patient were reactive for P. orhiculare. The clones were CD2+ and CD4+, except for one CD8+ blood clone. A majority of the TCC derived from lesional skin showed a Th2- or Th2/Th0-like cytokine profile. A co-expression of interIeukin-5 (IL-5) mRNA and II-13 mRNA was detected in five out of six P. orhicularc-reative clones analysed for their cytokinc gene expression with RT-PCR.Conclusion Our data suggest that P. orhiculare can induce a T-cell response in AD patients. The Th2-like profile of P. orbiculare-reactive TCC derived from lesional skin indicates that P. orhiculare may play a role in maintaining IgE-mediated skin inflammation in AD.

    Notes: Background The dust mite Lepidoglyphus destructor is an important cause of allergic reactions to dust, especially in farming environments. Two isoforms, recombinant (r)Lep d 2.01 and rLep d 2.02, of the major allergen Lep d 2, have previously been expressed as recombinant proteins. These isoforms differ 10.4% at the amino acid level. Furthermore, a mutant form of Lep d 2.01 (rLep d 2.6Cys) with a highly reduced IgE reactivity, has also been produced.Objective To investigate the T cell responses to the recombinant isoforms of Lep d 2, the Lep d 2.6Cys mutant and peptides of Lep d 2, in allergic and non-allergic individuals.Methods Peripheral blood mononuclear cells from 18 allergic and 16 non-allergic individuals were stimulated with the different antigens and the proliferative responses were measured. The cytokine production (interleukin (IL)-4, IL-5 and interferon (IFN)-γ) were measured by ELISA.Results Higher T cell proliferation was measured to isoform 01 than to 02 in 28/34 subjects. The responses to rLep d 2.6Cys were lower than to isoform 01 in most subjects, but higher than to Lep d 2.02. Two immuno-dominant peptides, corresponding to amino acid residue 11–25 and 61–75 were identified. The atopic subjects produced significantly lower IFN-γ in response to Lep d 2.01 as compared to the non-atopics.Conclusions There was a significant difference in T cell response between the two isoforms of rLep d 2. The hypoallergenic mutant rLep d 2.6Cys was able to evoke a T cell response with a magnitude which is between the two isoforms. Amino acid residue 11–25 and 61–75 are the most frequently recognized parts of Lep d 2 and are likely to contain the immuno-dominant T cell epitopes.
