    Notes: Abstract: A simple and efficient transfection technique based on lipopolyamine-coated DNA that can be used for gene transfer in cerebellar granular neurons is described. Gene transfer is achieved by exposure of cells to a DNA/lipid complex obtained by simple mixing of lipopolyamine and plasmid DNA. This procedure may represent a general tool of physiological investigations in primary cells. We show that the promoters of the introduced chimera genes are regulated by their respective trans-acting factors and may be modulated via membrane receptors and second messengers. This procedure has no noticeable toxic effects, nor does it seem to interfere with complex physiological behavior like neuronal differentiation.

    Notes: The initial stages of oxide formation on atomically flat, monohydride terminated Si(111) surfaces by oxidation in 30% hydrogen peroxide solution (H2O2) were investigated by scanning tunneling microscopy and x-ray photoelectron spectroscopy. The reaction proceeds via homogeneous nucleation of small oxide clusters on the surface and subsequent lateral growth of these clusters within the surface bilayer. The oxidation of the topmost Si(111) bilayer in H2O2 solution is completed after 30 min, leading to a SiO1.2 average layer composition. For the next bilayer the oxidation rate decreases drastically—after 2 months only 60% of the second bilayer are oxidized. An inverse logarithmic rate of the second bilayer oxidation is consistent with a field assisted growth mechanism. The significant differences in the oxidation rates between the first and the second bilayer allow to produce well defined oxide layers of about 5 A(ring) thickness. © 1995 American Institute of Physics.

    Notes: We show by scanning tunneling microscopy (STM) imaging that native oxide growth in moist air on hydrogen terminated Si(111) 1×1 surfaces begins by continuing formation of small oxide nuclei, 10–20 A(ring) in diameter, in the topmost Si layer. Their statistical distribution on the flat terraces points to a homogeneous nucleation process. Oxidation is extremely slow; after about 800 h only one complete monolayer is oxidized. In addition, a small number of three-dimensional oxide nuclei, several layers deep and 50–100 A(ring) in width, are formed at step edges as a minority species, which may be related to surface defects or contaminations.

    Notes: Postmitotic cerebellar granule cells, maintained for 5–6 days in Dulbecco's modified essential medium supplemented with 25 mM KCl, have been studied in whole-cell recording conditions to characterize calcium currents. With 10 mM Ba2+ as the divalent charge carrier, and using a pipette solution highly buffered for Ca2+ (30 mM EGTA, 100 mM HEPES–Tris, pH 7.2), only a high-threshold voltage-activated barium current was recorded from a holding potential of – 90 mV. The addition of 1 mM ATP to the pipette medium allowed stable recording for an average duration of 10 min, compatible with pharmacological studies of the barium current. Ninety-six per cent of the current was half-inactivated at low negative holding potential (-76 mV). A total block of current was obtained with 1 μM Cd2+. Sixty-three per cent of the mean current was abolished by 3 μM ω-conotoxin (ω-CgTx; Ki= 10 nM for a 15 min application), but individual cells showed either full sensitivity to this toxin or incomplete sensitivity. Seventy-eight per cent of the mean current was also abolished by 10 μM nicardipine but with a higher Ki, of 0.5 μM. After exposure to ω-CgTx, BAY K 8644 had no effect on the remaining current, though it was suppressed by nicardipine. No sensitivity to diltiazem, desmethoxyverapamil or flunarizine could be detected. Our major conclusion is that at least half of the channels have a mixed pharmacology, showing sensitivity to both ω-CgTx and dihydropyridine antagonists.

    Notes: The properties and characteristics of calcium channel openings in cerebellar granule cells were analysed by the cell-attached patch-clamp technique. At depolarized potentials, with 110 mM Ba2+ as the divalent charge carrier, 36% of the patches displayed activity that consisted of elementary events whose amplitude ranged from – 0.3 to –1. 75 pA at 0 mV, giving rise to a high threshold current. In this population of events at least four different types of channel openings were identified by their distinct biophysical and pharmacological properties. Two types of channel openings, with conductances around 24 and 7 pS, had similar characteristics in that both opened following two modes of gating characterized by brief (˜ 2 ms) and longer openings (˜ 8 ms) and both were sensitive to dihydropyridines. A further type of channel opening, with a conductance around 11 pS gated mainly with brief openings (∼1 ms), was shown to be insensitive to dihydropyridines but was undetectable in recordings from the cells that had been treated with ω-conotoxin. The last type of event was revealed after treatment of the cell with nicardipine or nifedipine and ω-conotoxin. The corresponding channel had a conductance of 19 pS and opened in one dominant mode characterized by brief openings (∼1 ms). The data obtained on single-channel activity of cerebellar granule cells are compared with the properties of the total current recorded in whole-cell conditions.
