    Notes: A combination of propranolol and hydrallazine was administered to 13 patients with longstanding hypertension during 15 pregnancies. Hydrallazine was continued through labour and delivery in all patients, while in eight patients propranolol was discontinued 2 to 15 days before delivery. Blood pressure control was uniformly good and superimposed pre-eclampsia did not occur during combined therapy. There were 14 livebirths and one unexplained stillbirth. Except for two cases of mild hypoglycemia, there were no neonatal complications.

    Notes: Summary: The functional significance of B7 co-stimulation in T-cell activation was described first in the context of preventing the induction of anergy. The functions of this pathway are far more complex than initially appreciated in view of the existence of two B7 molecules which have specificities for both CD28 and CTLA-4, which serve to amplify and terminate T-cell responses respectively Mice lacking B7 co-stimulators and CD28 and CTLA-4 co-stimulatory receptors are helping to clarify the functions of this key immunoregulatory pathway. In this review we will focus on the role of B7 co-stimulation in the activation and differentiation of CD4+ helper cells and CD8+ cytotoxic cells. The contribution of B7 co-stimulation to CD+ responses depends upon the activation history of the T-cell and the strength of the T-cell antigen receptor signal. B7 co-stimulation contributes to in Cerleukin (IL)-2 production by both naive and previously activated CD4+ T cells. B7 co-stimulation is most critical for the differentiation of naive CD4+ T cells to IL-4 producers, but predominately influences IL-2 production by previously activated CD4+ cells. B7 co-stimulation is important in development of cytotoxic T cells through both effects on T-helper cells and by direct co-stimulation of CDS+ cells.

    Notes: Abstract The distribution of three radioactively labelled salicylate derivatives with low ulcerogenic activity was compared with that of acetylsalicylic acid (ASA) and salicylic acid using whole body autoradiography and liquid scintillation counting techniques in rats. The methyl ester of ASA (AME) was distributed in vivo very similarly to that observed with ASA and salicylic acid. AME is rapidly demethylated following absorption from the stomach and is subsequently converted to ASA and salicylate. Salicylate is the main metabolite produced from both AME and ASA, which specifically accumulates in inflamed tissues. The 3-methyl-and 6-methyl-substituted salicylic acids are not as rapidly absorbed as either ASA or salicylic acid and do not pass readily into the brain or bone marrow. These results show that the methyl (ester) group of AME (which adequately protects the gastric mucosa from damage caused by ASA itself) does not impair the quantity of pharmacologically active form of drug (salicylate and ASA) generated in vivo. However, insertion of the methyl group at the 3- and 6-position of salicylic acid markedly affects both absorption, distribution and pharmaco-activity of these acids.

    Description / Table of Contents: Zusammenfassung Salben mit Griseofulvin respektive Proquazon wurden aus Monoglyceriden mittlerer Kettenlänge und einem aprotischen Lösungsmittel, Glycerinformal, hergestellt und auf dem Rücken von Gallefistelratten appliziert. Die percutan resorbierten Mengen sowie die Permeabilitätskonstanten sind für beide Wirkstoffe wesentlich höher nach Applikation der Salben als nach Applikation von einfachen Wirkstofflösungen ohne Monoglyceride. Die Verteilung der markierten Wirkstoffe in Rattenhaut wurde durch Mikroautoradiographie kontrolliert. Die Konzentrationen der Wirkstoffe in den einzelnen Schichten von menschlicher Haut sowie die mittleren Penetrationsraten wurden 16 h nach Applikation der Salben auf isolierte menschliche Haut bestimmt. Monoglyceride mittlerer Kettenlänge erhöhen die Permeabilität des Stratum corneum für gelöste Wirkstoffe wesentlich.
    Notes: Summary Ointments containing griseofulvin and proquazone, respectively, were made up of monoglycerides of medium chain length and an aprotic solvent, glycerinformal. The ointments were applied topically on the back of bile cannulated rats. The total amount absorbed percutaneously and the permeability constants of both drugs were considerably higher for the ointments than for simple solutions of the drugs without monoglycerides. Distribution of the labeled drugs in rat skin has been demonstrated by microautoradiography. Concentrations of the drugs in the different layers of human skin together with the medium flow rates have been determined 16h after administration of the ointments onto isolated human skin. Monoglycerides of medium chain length enhance significantly the permeability of the stratum corneum for solutes.

Das „Toxin”, ein Lipid-Protein-Komplex, das als Vorstufe in der normalen Haut vorkommt, wirkt im Tierversuch bei i.p. Applikation letal, setzt die Infektionsresistenz herab, wird in allen Organen angereichert und schädigt die Zellen, wie Untersuchungen der Ultrastruktur und des Stoffwechsels der Leber nachwiesen. Ein Antitoxin wurde entwickelt, das im Tierversuch eine signifikante Schutzwirkung zeigte.
    Notes: Summary In high temperature burns a specific substance is formed in the skin that has been isolated from in vitro burned animal and human skin as well as from serum of serverely burned patients. This toxic lipid-protein complex derived from a naturally occurring precursor has a lethal effect when injected i.p. into recepient mice. It accumulates in all organs and reduces the animals' resistance to infection. Cell destruction was shown by ultrastructural and metabolic changes in the liver. An antitoxin was developed that protects against the lethal effect as well as the superimposed sepsis.
