    Notizen: Melatonin (MLT) was isolated from water samples using solid phase extraction cartridges and measured by radioimmunoassay. In tanks of rainbow trout Oncorhynchus mykiss, concentrations of MLT in water increased rapidly from 〈0·1 ng l−1 in the light phase to 0·7 ng l−1 in the dark giving calculated release rates of 〈1 ng kg−1 h−1 and 15 ng kg−1 h−1, respectively. This cycle in water MLT values corresponds to reported changes in plasma MLT concentrations. The colour of the tanks and a daytime acute handling stress did not affect concentrations of MLT in either the light or dark phase.

    Notizen: Behavioural and electrophysiological responses of reproductive and non-reproductive female round gobies Neogobius melanostomus to water previously occupied by male round gobies (reproductive male water) were compared. Reproductive females spent more time than non-reproductive females in a tank near the input of water conditioned from reproductive males. Also, reproductive females swam significantly faster than non-reproductive females, suggesting that reproductive male odour may have activated spawning behaviour. Olfactory epithelial field potential measurement (electro-olfactogram, EOG) showed that reproductive male water was a potent olfactory stimulus to reproductive females, but not to non-reproductive females. Reproductive females responded significantly more than non-reproductive females to solid-phase (octadecylsilane) extracts of reproductive male water. Also, when these extracts were separated on reverse phase high performance liquid chromatography (HPLC), reproductive females showed noticeably greater responses than non-reproductive females to the fractions that eluted between 30 and 40 min. The behavioural data support the hypothesis that reproductive male round gobies release compounds into the water that attract potential mates. The EOG data indicate that these compounds can be quantitatively extracted from the water and be partially purified by HPLC. The evidence is not sufficient to indicate whether or not the compounds are steroids. The relatively early elution time on HPLC, however, suggests that if these compounds are steroids, then it is more likely that they will be conjugated rather than free steroids.

    Notizen: In order to gain a better understanding of the reproductive cycles of male and female tench (Tinca tinca), gonadosomatic index, gonad histology and plasma concentrations of estradiol-17β (E2), testosterone, an drostenedione, 11-ketotestosterone (11-KT), 17,20β, 21-trihydroxy-4-pregnen-3-one (17,20β,21-P), 17,20β-dihydroxy-4-pregnen-3-one (17,20β-P) and 17,20α-dihydroxy-4-pregnen-3-one (17,20α-P) were measured at the four seasons of the year, plus a further sampling coincident with the peak of spawning in early July. As expected, in both males and females, the plasma concentrations of androgens (excluding 11-KT in females – undetectable) and C21 steroids were significantly more elevated in the spring and summer (when most gonadal development took place) than in the autumn and winter. The only unexpected finding was that 17,20β-P and 17,20β,21-P, the steroids that are normally associated with oocyte final maturation in females and spermiation in males, were found in substantial amounts in both pre-vitellogenic, pre-spermatogenic and post-spawning fish. This suggests that these steroids may have other as yet unidentified roles in this species.

    Notizen: Levels of sulphated 17α20β-dihydroxy-4-pregnen-3-one (17,20β-P; the oocyte maturation inducing steroid) in blood plasmas of sexually mature male and female rainbow trout Oncorhynchus mykiss, were very low in comparison to those of the free steroid. However, relatively large amounts were found in urine of both sexes.Catheters were inserted into the urinary bladders of unovulated and ovulated females and of ripe-running males, and the fish then placed in spawning channels. Three-hourly urine samples were collected between 09.00 and 18.00 hours and then a 15-h sample between 18.00 and 09.00 hours the next morning. Measurements were made of 17,20β-P-sulphale, testosterone glucuronide (T-G) and 17β-oestradiol glucuronide (E2-G). In females, the highest rates of excretion of E2-G, T-G and 17,20β-P-sulphate were found in unovulated, ovulating and ovulated females, respectively. The rates of excretion of 17,20β-P-sulphate, T-G and E2-G in ovulated females were unaffected by the presence of a male. id males, however, there was a sharp increase in the rate of excretion of 17,20β-P-suiphate and T-G in fish which were paired with an ovutated (nesting) female. A similar increase was found in males injected with male trout pituitary extract.

    Notizen: Urine of reproductively mature female rainbow trout was shown to contain a priming pheromone which raised the levels of 17a,20β-dihydroxy-4-pregnen-3-one (17,20β-P), testosterone and gonadotrophin II in the blood plasma of reproductively mature male rainbow trout. Milt volumes, however, were unaffected.Because it had been established in a previous study that the sulphated form of 17,20β-P is abundant in the urine of spawning rainbow trout, synthetic 17a,20β-dihydroxy-4-pregnen-3-one 20-sulphate was also tested for pheromonal activity. It was found to have only a smalt and inconsistent priming effect on steroid levels and did not alter the orientation or spawning activity of males.It was also shown that, in the majority of experiments, there was a significant drop, over the course of 24 h, in the levels of 17,20β-P and testosterone in the control groups of males.

    Notizen: This paper compares some morphological and endocrinological characteristics of diploid and triploid rainbow trout.Significant differences were found between diploid and triploid females in GSI, condition factor, gut weight, liver weight and percentage dress-out, and between diploid and triploid males in GSI, condition factor and gut weight.Diploid females had large, well-developed ovaries containing yolk-filled secondary oocytes whereas the triploids had only string-like ovaries containing nests of oogonia. No primary oocytes were present.All the diploid males produced copious quantities of milt but it was possible to express a thin, watery milt containing motile spermatozoa from only two of the 12 triploid males. Testes weights in triploids were similar to those of diploids but, while the diploid testes were packed with spermatozoa, those of the triploids consisted mainly of spermatocytes and spermatids with few spermatozoa present. Measurements of the heads of spermatozoa revealed that those from triploids were larger and had a wider size range than those from diploids.Levels of testosterone and 11-ketotestosterone in triploid and diploid males were not significantly different. However, levels of testosterone and 17β-oestradiol in diploid females were considerably higher than those of triploid females.

    Notizen: Earlier work on the factors influencing inhibition of activation of the spermatozoa of Salmo gairdnerii has been confirmed. Diluents containing K+ and of pH less than 7.8 do not activate the spermatozoa. The presence of Na+ in the diluent has a mildly antagonistic effect on this inhibition. It has now been demonstrated that both Ca2+ and Mg2+ markedly antagonize the inhibition due to low pH and K-. It is suggested that inhibition due to potassium salts could account for the inhibition previously attributed to Androgamone I.

    Notizen: 17β-oestradiol, testosterone, 11 -ketotestosterone and calcium were measured in plasma of female rainbow trout over the course of a single spawning season. The patterns of rise and fall of the levels of 17β-oestradiol and calcium during sexual maturation were similar to those demonstrated by other workers. Very high levels of testosterone were found in plasma of sexually mature fish—mean level 211 ng ml-1 in November and 115 ng ml-1 in January. Ovulation occurred from December to February.
