    Notes: Electron cyclotron emission measured vertically through the center of a tokamak plasma yields detailed information about the electron velocity distribution. A diagnostic developed for this purpose on Alcator C tokamak uses specialized focusing optics to obtain a well-collimated viewing chord, a compact viewing dump made of Pyrex or Macor to reduce the effects of wall reflection and depolarization, and a rapid-scan polarizing Michelson interferometer InSb detector system for the spectrum measurement; all constrained by the limited access and the compact size of Alcator C. Results of diffraction analysis are used to evaluate the theoretical performance of the optical system.

    Notes: Compact, vacuum-compatible submillimeter viewing dumps, whose design parameters were based on the results of simple ray calculations, have been fabricated from Pyrex, Macor, and alumina. The measured performance of the dumps agrees qualitatively with calculated results. The absorption is somewhat lower than expected, but still acceptable (≥99% absorption for Pyrex and Macor). In an electron cyclotron plasma emission experiment on Alcator C tokamak, comparison of the emission spectra with and without the dump shows that the dump in this configuration is effective in suppressing the unwanted radiation by approximately 90%. The additional reflection is believed to be due to a portion of the antenna pattern missing the dump.

    Notes: Nonthermal electron distribution functions, arising during lower hybrid heating and current drive, have been measured on Alcator C tokamak using vertical electron cyclotron emission and also bremsstrahlung x-ray emission. The methods used for deconvolving the distribution from the radiation measurements are summarized and examples are given of results from different types of plasma, illustrating the present capabilities of the techniques. The cyclotron emission results are consistent with the x-ray analysis but provide additional information, particularly at lower energies (30–100 keV). The x rays are less ambiguous at high energy and give good forward/backward asymmetry diagnosis. The quality of information appears to be already adequate for a quantitative comparison with theory, but further experimental improvements seem possible.

    Notes: Summary Light and electron microscopic localization of cathepsin H in rat kidney was studied using post-embedding immunocytochemical techniques. For ligh microscopy, Epon sections of the kidney were stained by immunoenzyme method after removal of Epon and for electron microscopy, ultrathin sections of the Lowicryl K4M-embedded material were labeled by protein A-gold (pAg) technique. By light microscopy, fine granular staining was found in throughout the nephron, but the staining intensity considerably varied. The strongest staining was noted in the S1 segment of the proximal tubules followed by the S2 and S3 segments and the medullary collecting tubules. The glomeruli, the distal tubules, and the cortical collecting tubules were weakly stained. By electron microscopy, a gold label was found exclusively in lysosomes, which showed various sizes and labeling intensity. The results were quite consistent with the light microscopic results. The labeling intensity tended to increase as the matrix of lysosomes was condensed. Quantitative analysis of the labeling density of lysosomes demonstrated that the highest labeling density is found in the S1 segment of the proximal tubules and the labeling density of other renal segments is significantly low levels. The results indicate that a main site for cathepsin H in rat kidney is the S1 segment of the proximal tubules.

    Notes: Summary The localization of 5′-nucleotidase in rat parotid and submandibular glands was investigated at the electron microscope level by an immunohistochemical technique using a highly specific antibody, and the results were compared with those obtained using the newley developed cerium method for enzyme histochemistry. Both methods demonstrated that 5′-nucleotidase is located on the external surface of the luminal plasma membranes of acinar cells as well as on intercalated and striated ductal cells. In the basolateral membranes of these cells, the portions adjacent to myoepithelial cells exhibited intense reaction products, but the other areas of plasma membranes contained only trace amounts of the reaction products. Both cerium-based enzyme histochemistry and immunohistochemistry showed that myoepithelial cells retain the enzyme on their plasma membranes. Neither method produced reaction products in the intracytoplasmic structure of constitutive cells of the salivary glands. We discuss the usefulness of the cerium-ion method for the demonstration of 5′-nucleotidase activity and compare it with the traditional lead-ion method.

    Notes: Abstract An emulsion polymerization of styrene in the presence of an amphoteric surfactant; N,N-dimethyl-n-laurylbetaine (LNB) was carried out at pH 7.0. The polymerization rate and the concentration of the latex particle were proportional to the LNB concentration of 0.6 power. The latex particle sizes became smaller with increasing concentrations of LNB. The molecular weights of polystyrene latices increased with the concentration of LNB. The zeta-potentials of latex particles were significantly dependent on the pH, and showed the existence of an isoelectric point.

    Notes: Summary In peripheral blood and synovial fluid of patients with rheumatoid arthritis (RA), increased levels of Ia antigen-positive (Ia+) T cells have been demonstrated. Therefore, we examined these Ia+ T cells in vitro to identify their role in the production of rheumatoid factor (RF) and to study the immunologic abnormalities of RA. When Ia+ T cells from peripheral blood of RA patients were added to pokeweed mitogen (PWM)-non-stimulated autologous B cells, the amount of IgM-RF production was 25.8±6.4 (mean±SE) (p〈0.001) as compared to 16.0±4.6 ng/ml (mean±SE) in the presence of Ia− T cells. When Ia− OKT4+ cell fractions, obtained by excluding Ia+ T cells from OKT4+ cells, were added to B cells, the increase in IgM-RF production was markedly lower than that obtained with the OKT4+ cell fraction. These results indicate that the helper T cells which induce the production of IgM-RF may derive from the Ia+ OKT4+ cell fraction. B cells from rheumatoid synovial fluid produced IgM-RF levels as high as 102.7±19.2 ng/ml (mean±SE) even without stimulation. When T cells from autologous synovial fluid were added, IgM-RF production was not increased. These data suggest that B cells from RA synovial fluid had already been activated. When synovial fluid T cells were added to B cells from autologous peripheral blood, larger amounts of IgM-RF were produced as compared to experiments in which T cells from peripheral blood were added, suggesting that T cells from synovial fluid induce an enhanced IgM-RF production by B cells. The presence or absence of Ia antigen on the surface of synovial fluid T cells did not affect the level of IgM-RF production. Our results indicate that Ia+ T cells from the peripheral blood of RA patients induce the production of IgM-RF by autologous B cells.
