    Notes: Biosynthetically labelled Qa-2 antigens were isolated from mouse spleen cells by immunoprecipitation with anti-Qa-2 antisera. When newly synthesized Qa-2 molecules from several different inbred strains were analysed by two-dimensional (2D) gel electrophoresis; four different phenotypes were observed that differed in the number of polypeptides present. The ability to distinguish Qa-2+ phenotypes was used to map the recombination points in two congenic strains, B6. Tlaa and A. Tlab. No alternative Qa-2-like polypeptides were detected in B6.K1 (Qa-2−) cells using a polyspecific rabbit antiserum against mouse class I antigens, but a new molecule was detected in BALB/cBy (Qa-2−) cells. Pulse-chase and surface-labelling experiments showed that some, but not all, of the newly synthesized Qa-2 precursor forms are processed to mature cell surface molecules.

    Notes: Abstract Effective diagnosis and treatment of periodontal disease requires accurate evaluation of disease status before and after treatment. This study evaluated relationships among multiple parameters that have been used for periodontal disease evaluation. A total of 61 initially deep (≥6 mm) interproximal pockets from 16 patients examined before and after a 3-week course of root planing therapy were evaluated for probing depth, bleeding on probing, darkfield and cultural characterization of subgingival plaque, and histometric indices of infiltrated connective tissue (ICT) and mature plasma cell infiltrate. 36 sites were evaluated before treatment, and 25 after treatment. A comparison of mean scores for pre-treatment sites versus post-treatment sites indicated that there was an average improvement in most disease-related parameters. However, patterns of significant correlations among the parameters differed markedly. Motile bacteria enumerated by darkfield microscopy were significantly correlated with ICT and plasma-cell populations before, but not after treatment. In contrast, probing depth and populations of black pigmented Bacteroides (BPB's). principally B. gingivalis, were significantly correlated with ICT after, but not before, treatment. Bleeding on probing was not significantly correlated with ICT levels before or after treatment. This suggests that different sets of parameters should be used for evaluating periodontal disease status before or after treatment. Our data further suggest that B. gingivalis populations may be related to delayed healing of deep periodontal lesions after instrumentation.

    Description / Table of Contents: Zusammenfassung Geringfügige Verschiebungen der allgemeinen Zirkulation führen oft zur Entstehung quasi eines Mosaiks feuchter und trockener Flecken, wobei sich im Mesoscale Dürregebiete bilden. Mit Hilfe der bedingten Wahrscheinlichkeit, an Nachbarstationen gleichzeitig Niederschlagsdefizite festzustellen, wurde eine Methode entwickelt, das räumliche Muster dieser Dürre zu beschreiben. Diese Methode wurde anhand jahreszeitlicher Daten für den Osten North Carolinas (USA) erprobt. Die daraus resultierenden Verteilungsmuster können mit der synoptischen Klimatologie des Gebietes in Beziehung gebracht werden. Im Sommer verzeichnet man zahlreiche kleine Gebiete mit Niederschlagsdefiziten, während in den anderen Jahreszeiten größere Trockengebiete auszumachen sind, die die unterschiedlichen Zugbahnen der Tiefdruckgebiete, Küsteneinflüsse und Regenschatteneffekte widerspiegeln. Ein Vergleich mit dem üblicherweise verwendeten „Palmer Drought Severity Index“ zeigt, daß letzterer nur ausgedehnte Trockenheiten im geeigneten Maß darstellt, die Identifikation von Dürre im Mesoscale jedoch nicht zuläßt. Die vorliegende Methode erlaubt eine Identifikation jener Gebiete, die für mesoskale Dürre predästiniert sind, gibt ein Bild der kausalen Zusammenhänge und schafft damit die Grundlage zur Beobachtung der Entstehung solcher Trokkenheiten.
    Notes: Summary Minor shifts in the pattern of the general circulation frequently result in the development of a mosaic of localized wet and dry patches, creating regions of mesoscale drought. A method of identifying the spatial pattern of such droughts is developed, using the conditional probability of adjacent stations experiencing precipitation deficits simultaneously. The method was tested using seasonal data for the eastern portion of North Carolina, U.S.A. The resulting patterns can be related to the synoptic climatology of the area. Summer reveals numerous small areas of precipitation deficit, while the other seasons have larger deficit areas which reflect changing depression tracks, coastal influences, and rainshadow effects. Comparison of the results with the commonly used Palmer Drought Severity Index indicates that the latter is an appropriate measure only when drought is widespread, and cannot be used to identify mesoscale droughts. The present method allows identification of areas prone to mesoscale drought, suggests causal mechanisms, and provides a basis for monitoring the development of such droughts.

    Notes: A procedure is described for the analysis of the estrogens equilin and estrone in human plasma following oral administration of conjugated estrogen preparations. After enzymatic hydrolysis of the sulfate conjugates, plasma proteins are precipitated with methanol and the estrogens extracted into ethyl acetate. Derivatization with the reagent flophemesylamine converts equilin and estrone into volatile pentafluorophenyldimethylsilyl ethers ideally suited to capillary gas chromatography-negative ion chemical ionization mass spectrometry. Using a 15 meter dimethyl silicone bonded phase fused silica capillary column separation of the estrone and equilin derivatives is achieved within 9 minutes. Selected ion monitoring of the intense negative molecular ions enables levels of 1 ng.ml-1 to be measured with coefficients of variation of 9.3 % and 14.2 % for estrone and equilin respectively. Plasma levels of the compounds are reported in two male volunteers up to 24 hours after dosing with 5 milligrams of Premarin™. (™ Ayerst Laboratories Inc., New York, USA.).
