    Notes: We developed an advanced Kirkpatrick–Baez (AKB) x-ray microscope which consisted of two hyperbolic mirrors and two elliptic mirrors. The spatial resolution of (approximately-less-than)3 μm was realized over ∼1 mm diam. This AKB microscope was used for x-ray imaging in laser fusion experiments. Laser absorption nonuniformity with a large wave number on a spherical solid target or a plane slab target was estimated by measurements of x-ray emission from the target surface with the microscope. The x-ray images of the imploded core plasmas were also obtained with the AKB microscope, changing the laser focus condition and the laser energy balance. © 1997 American Institute of Physics.

    Notes: We investigated the quantum confined Stark effect in GaAs quantum wires formed in a V-groove structure, demonstrating observation of a blueshift of the photoluminescence peak with the increase of electric fields at 50 K. This blueshift is attributed to the fact that the change in enhanced binding energy of excitons due to the electric field is larger than that in quantized energy levels of electrons and holes. Time-resolved photoluminescence was also measured. The photoluminescence decay time is decreased in small quantum wires of 8 nm width with the increase of electric fields, while the decay time is increased in the quantum wires with a size of 35 nm. These results indicate that the escaping of carriers is more dominant in smaller structures than reduction of the oscillator strength due to the electric fields. © 1997 American Institute of Physics.

    Notes: The mechanism underlying the ontogenetic increase in plasma growth hormone (GH), luteinizing hormone (LH) and follicle stimulating hormone (FSH) concentration during fetal life in mammalian species and the prenatal sex difference in these hormones in some species is not fully understood. To this end anterior pituitaries were collected from German Landrace fetuses and piglets at day (d) 50, 65, 80, 95, 110 pc and d 6 pp and pituitary GH, LHβ, FSHβ and α-subunit mRNA levels were determined by measuring Northern blot hybridization signals. GH mRNA was detected in both sexes as early as d 50 pc. The mRNA level markedly increased with age in both sexes (males〉females, P≤0.05) reaching its maximum at d 95/110 pc. LHβ mRNA signals were first detected at d 50 pc in females and at d 65 pc in males increasing thereafter to a maximum at d 6 pp in both sexes (P≤0.05). In males the augmentation in LHβ mRNA was delayed compared to females (P≤0.01). Before d 80 pc no FSHβ mRNA hybridization signals were apparent. Thereafter the mRNA level continuously increased with age (P≤0.01) in both sexes reaching its maximum at d 6 pp. The FSHβ mRNA level in females was always higher than in males (P≤0.01). As early as d 50 pc the α-subunit mRNA level was high in both sexes and further increased without sex difference to d 6 pp (P≤0.05). In conclusion, the mRNA levels of GH, LHβ and FSHβ are age and sex dependent during fetal development. We suggest that the fetal increase in plasma GH concentration can be accounted for by changes in GH mRNA levels, while the dramatic perinatal decrease in plasma GH concentration seems to be primarily controlled at the posttranslational and/or secretion level. The fetal sex difference and the increase in plasma LH and FSH concentrations seems to be primarily dependent on the cellular concentration of the gonadotropin β-subunit mRNAs and/or number of gonadotrophs.

    Notes: The participation of adhesion molecules in systemic vascular injuries of the generalized Shwartzman reaction was studied. The generalized Shwartzman reaction was induced in mice by two consecutive injections of lipopolysaccharide. Intercellular adhesion molecule-1 (ICAM-1) was expressed on vascular endothelial cells, renal tubular cells and alveolar wall in generalized Shwartzman reaction-induced mice. The preparative injection of lipopolysaccharides induced ICAM-1 expression in those cells, and the provocative injection of lipopolysaccharides for the generalized Shwartzman reaction augmented it further. The simultaneous administration of anti-gamma interferon antibody with the preparative injection of lipopolysaccharides completely inhibited ICAM-1 expression on vascular endothelial cells. The injection of recombinant gamma interferon in replacement of lipopolysaccharides resulted in ICAM-1 expression. The administration of anti-ICAM-1 antibody together with the provocative injection of lipopolysaccharides significantly blocked the apoptosis of vascular endothelial cells in generalized Shwartzman reaction-induced mice. It was suggested that ICAM-1 expression on vascular endothelial cells might be involved in systemic vascular injuries of the generalized Shwartzman reaction, and that it might be regulated by gamma interferon.

    Notes: 4f-Electron density in single crystals of CeB6, cerium hexaboride, was measured at 165 K by X-ray diffractometry. Significant peaks 2.0 e Å−3 high were found along the \langle100\rangle directions at 0.41 Å from Ce on the deformation density map. Analysis based on the crystal field theory removed the peaks, confirming that they were due to the Ce 4f-electrons on the t1u-orbital. The deformation density in the B6 octahedron was also markedly improved by the analysis and coincides qualitatively with a theoretical molecular orbital (MO) calculation. It also coincides with the model deformation density map of CaB6 composed of only light atoms. These facts guarantee the accuracy of the intensity measurement for the present study. Since Ce3+ has only one 4f-electron, a highly accurate intensity measurement is necessary to detect its 4f-electron density distribution. A ψ-scan technique was therefore employed to avoid multiple diffraction (MD) and to measure the intensities at ω and χ angles with minimum fluctuation of temperature at the sample position. Relativistic radial functions for orbitals of Ce3+ and the corresponding scattering factors, which take the aspherical electron density distribution of 4f-electrons into account, were used for the analysis. CeB6 is a typical dense Kondo material. The Kondo effect occurs in CeB6 from low temperature to above room temperature. X-ray analysis of the f-electron density based on atomic orbitals (AO) revealed that 1.5 (2) electrons are donated from B6 to Ce and a total of 2.5 (2) electrons localize on the 4f-orbital. κ values are consistent with the 4f-orbitals being highly contracted and thus stabilized. These may be related to the Kondo effect.

    Notes: Abstract SulA is induced in Escherichia coli by the SOS response and inhibits cell division through interaction with FtsZ. To determine which region of SulA is essential for the inhibition of cell division, we constructed a series of N-terminal and C-terminal deletions of SulA and a series of alanine substitution mutants. Arginine at position 62, leucine at 67, tryptophan at 77 and lysine at 87, in the central region of SulA, were all essential for the inhibitory activity. Residues 3–27 and the C-terminal 21 residues were dispensable for the activity. The mutant protein lacking N-terminal residues 3–47 was inactive, as was that lacking the C-terminal 34 residues. C-terminal deletions of 8 and 21 residues increased the growth-inhibiting activity in lon + cells, but not in lon − cells. The wild-type and mutant SulA proteins were isolated in a form fused to E. coli maltose-binding protein, and tested in vitro for sensitivity to Lon protease. Lon degraded wild-type SulA and a deletion mutant lacking the N-terminal 93 amino acids, but did not degrade the derivative lacking 21 residues at the C-terminus. Futhermore, the wild-type SulA and the N-terminal deletion mutant formed a stable complex with Lon, while the C-terminal deletion did not. MBP fused to the C-terminal 20 residues of SulA formed a stable complex with, but was not degraded by Lon. When LacZ protein was fused at its C-terminus to 8 or 20 amino acid residues from the C-terminal region of SulA the protein was stable in lon + cells. These results indicate that the C-terminal 20 residues of SulA permit recognition by, and complex formation with, Lon, and are necessary, but not sufficient, for degradation by Lon.

    Notes: Abstract Immunohistochemical localization of the large proteoglycan, PG-M/versican, was studied in 36 breast tumours, including infiltrating ductal carcinomas, benign tumours and fibrocystic diseases. The relation between the proteoglycan and the other extracellular matrix components was also investigated. In the carcinoma tissues, the interstitial elements of the ‘specific stroma’, consisting of fibroblastic cells and fine fibrils, were reactive to antibody 2B1, which specifically recognizes the large proteoglycan, PG-M/versican. In the peripheral invasive areas of infiltrating ductal carcinoma, the most intense 2B1-positive reaction was visualized in mesenchymal tissues between carcinoma cell clumps and the surrounding tissues, where hyaluronic acid could be demonstrated histochemically. The 2B1-positive elements were not reactive to antibody 6B6, which specifically recognizes small proteoglycan. In the central sclerotic areas, where antibody 6B6 was reactive, a 2B1-positive reaction was detected only in elastosis masses, which also bound antibodies to type IV collagen and laminin, and to some extent antibody raised against chondroitin 6-sulphate proteoglycan. Elastic tissues of blood vessel walls and perivascular elements became reactive to antibody 2B1 when they were involved in carcinoma invasion. The present results have shown that PG-M/versican was localized in the proliferating interstitial tissues, in particular in hyaluronic acid-rich portions, in association with carcinoma cell growth, and also that PG-M/versican accumulated in vascular and perivascular elastic tissues involved in carcinoma invasion. The biological significance of PG-M/versican was briefly discussed
