    Notizen: Abstract.— Filial cannibalism has been identified as a constraint to the intensive production of swordtails Xiphophorus helleri. The objective of this study was to quantify the effect of refuge availability and quantity, broodstock population density, and adult sex-ratio on the rate of cannibalism under culture conditions. The availability of shelter had a significant effect on the number of harvestable juveniles, while the quantity of shelter did not affect harvest size. At a constant sex-ratio of 1:1 and broodstock population densities of four, ten, and 16 fish per 300-L tank the amount of juveniles harvested was lowest at the stocking density of four fish per tank, but did not increase significantly when stocking density was raised from ten to 16 fish per tank. Rate of cannibalism was lowest at the lowest population density. At a constant stocking density of ten fish per tank and sex-ratios of 1:1, 1:4, and 4:1 (male: female), the highest number of juveniles was obtained at a social structure of two males and eight females. Rate of cannibalism was independent of sex-ratio, indicating that males and females are equally cannibalistic. These results suggest that the 300-L broodstock tanks should be stocked with a maximum of two males and between five and eight females to obtain the greatest number of harvestable juveniles per tank.

    Notizen: Recombinant Sulfolobus solfataricus glyceraldehyde-3-phosphate dehydrogenase has been purified and found to be a tetramer of 148 kDa. The enzyme shows dual cofactor specificity and uses NADP+ in preference to NAD+. The sequence has been compared with other GAPDH proteins including those from other archaeal sources. The purified protein has been crystallized from ammonium sulfate to produce crystals that diffract to 2.4 Å with a space group of P43212 or P41212. A native data set has been collected to 2.4 Å using synchrotron radiation and cryocooling.

    Notizen: Abstract The MUC7 gene encodes the protein core of the low molecular weight human salivary mucin (MG2, mucin glycoprotein 2) and is expressed in a tissue-specific manner in salivary glands. The purpose of this study was to examine MUC7 expression by transgenic mouse technology. A 16 kb DNA fragment, containing the MUC7 gene (10 kb) and 3 kb of the upstream and 3 kb of the downstream sequences, was used to generate transgenic mice. We have identified five transgenic founder mice which were propagated as individual transgenic lines and analysed. Tissues of transgenic offspring from each line were analysed by RT-PCR to determine the sites of the MUC7 expression. The results indicated that only line 3 and line 5 expressed the MUC7 gene in salivary glands. The level of MUC7 expression in selected tissues was then determined by northern blot analyses. The results showed that line 3 mice contained high levels of MUC7 transcripts in the sublingual glands of both males and females and indicated low levels of MUC7 transcripts in the submandibular glands of females. No MUC7 expression was detected in this line by northern blot analysis in any other tissue tested. On the other hand, no expression of MUC7 was detected in any tissues of line 5 mice examined by northern blot analysis. A Southern blot analysis of human and mouse genomic DNA demonstrated multiple copies of the MUC7 transgene in line 3 and a single copy in line 5. Collectively, these results indicate that the regulatory sequences required for the tissue-specific expression of MUC7 are within the MUC7 transgene. However, the sequences necessary for expression comparable to that of MUC7 in human salivary glands may be missing from this construct. Western blot analysis of protein extracts from different tissues of transgenic mice line 3 showed that MUC7 gene product was produced in the submandibular-sublingual gland complex of both male and female mice and not in the other tissues examined

    Notizen: Abstract Mechanisms that regulate cytokine-mediated inflammation in the lungs of preterm infants, including factors which regulate production of the chemokine IL-8, remain poorly defined. Sequential bronchoalveolar lavage samples were obtained from preterm newborns with hyaline membrane disease over a 28-day period. Bronchoalveolar lavage cell cytokine relationships were evaluated and the differential regulation of IL-8 by IL-1β and TNFα was studied in a short-term culture system. In vivo, IL-8 and IL-lβ protein levels correlated closely with each other and with macrophage counts. In cell culture, exogenous anti-IL-1β antibody led to a 40% maximum inhibition (approximately) of IL-8 production by lipopolysaccharide stimulated lung inflammatory cells. Comparable amounts of exogenous anti-TNFα antibodies achieved a 15% maximum inhibition (approximately) of IL-8 production. Anti-IL-1β and anti-TNFα antibodies in combination did not inhibit IL-8 production beyond that achieved by anti-IL-lβ antibody alone. These results, in preterm newborns, support the concept of lung inflammation mediated in part by a macrophage, IL-1β, and IL-8 cell cytokine pathway. The results also suggest that factors other than IL-1β and TNFα regulate IL-8 expression in the lungs of preterm infants.

    Notizen: Abstract The most fundamental difficulty in the construction of an Earth-like dynamo model is associated with the constraint caused by the rapid rotation of the Earth. To stabilise numerical codes, many workers have introduced hyperviscosity into the governing equations. One of the major effects introduced by hyperviscosity is to offset the rotational constraint, and, consequently, to alter the key dynamics of an Earth-like dynamo. In this paper, an Earth-like convection model with or without the presence of an imposed magnetic field is investigated with or without the effect of hyperviscosity. A nonlinear dynamo model with the mean field approximation is also used to examine the dynamical effect of hyperviscosity. The results suggest that great care should be taken when hyperviscosity is employed in geodynamo models.
