    Notes: Abstract. The aim of the work reported here is the recovery, from a single image taken inside a roughly cylindrical brick sewer pipe of diameter up to one meter, of the pose of the camera relative to the central axis of the pipe. It is shown that the vanishing point associated with the longitudinal mortar lines carries valuable information about the pose. A method for the automatic detection of this point is presented and used to analyse the camera rotations underlying a number of sewer survey videos. It is similarly shown how the angles between the images of the longitudinal lines can be used to recover information about camera pose. The techniques might form an active part of a more comprehensive image understanding system recovering the three-dimensional shape of a surveyed pipe from survey videos and/or be used as an experimental tool during the design of such a system.

    Notes: Abstract The methylation status of 12 CpG sites in three exons of the human factor VIII (F8C) gene was examined by bisulphite genomic sequencing of human sperm DNA from 14 European Caucasians and Asians. Different CpG sites were found to vary in their methylation status both within and between individuals. Strand differences in methylation status were also detected at certain sites, a finding that could reflect hemi-methylation. No evidence for systematic deviations in methylation status were found between the two ethnic groups. Only a limited correlation was observed between the level of methylation of specific CpG sites in sperm DNA and their mutability, a finding that is probably attributable to the pattern of methylation observed in mature spermatocytes not being representative of that of the germline.

    Notes: Abstract Mutation screening in neurofibromatosis type 1 (NF1) families has long been hampered by the complexity of the NF1 gene. By using a novel multi-track screening strategy, 67 NF1 families (54 two-generation, 13 three-generation) with a de novo mutation in the germline of the first generation were studied with two extragenic and 11 intragenic markers. The pathological lesion was identified in 31 cases. Loss of heterozygosity (LOH) in the affected individual revealed a gross gene deletion in 15 of the two-generation families; in 12 (80%) of them, the deletion was maternally derived. Eleven patients with a gross deletion exhibited developmental delay, ten had dysmorphic features and six manifested a learning disability. No gross deletion was apparent in any of the 13 three-generation families, suggesting that such lesions are subject to more intense selection. In these families, the new mutation was of paternal origin in 11 kindreds and the underlying mutational event could be characterised in three of them.

    Notes: Abstract The effect of femoral guide rotation in endoscopic anterior cruciate ligament (ACL) reconstruction is reviewed based on a previous report. The effect of varied offsets of the femoral guide (5.5 and 7.0 mm) are described. This is one of the few isometry studies to evaluate the knee through a practically full range of motion (0°–120°). A 7-mm offset guide rotated to the 12 : 00 position yields the best single fiber and graft excursion patterns (P 〈 0.05). A 5.5-mm offset guide yields inferior single fiber and graft excursion patterns. Single fiber and graft isometry are similar but not identical in endoscopic ACL reconstruction. Centering the single fiber in the tibial tunnel has little effect on isometry patterns, demonstrating that the more posterior tibial positions needed for endoscopic reconstruction are acceptable from an isometry standpoint.
