    Notes: We have investigated the electron paramagnetic resonance (EPR) of self-assembled stearic acid films adsorbed on an Al2O3-film. Doping the film with spin labels at different positions of the alkyl chain in order to make the films accessible for EPR spectroscopy provides an opportunity to investigate the rotational motion of the molecule along the alkyl chain. The temperature dependent EPR spectra show a strong dependence of the rotational motion of the molecules with variation of the location of the spin label along the chain. We study the rotational motion by means of the EPR line shape analysis. © 1998 American Institute of Physics.

    Notes: The magnetic properties of noninteracting single-domain particles whose anisotropy is made up of a cubic magnetocrystalline and a uniaxial components were investigated. Various directions of the uniaxial anisotropy were considered and the dependencies of the reduced remanence as a function of the ratio between the two anisotropies were obtained. It was found that for sufficiently strong uniaxial anisotropy and random arrangement of the particle orientations, reduced remanence lower than 0.5 is, in general, an intrinsic property of the system due to the negative remanent magnetization of some of the particles. © 1998 American Institute of Physics.

    Notes: We present experimental magnetoresistance, magnetization, and remanence data for CoCu granular films obtained by electron beam codeposition, subsequently submitted to a thermal treatment. The magnetization measurements show coexistence of superparamagnetic and ferromagnetic (or blocked superparamagnetic) Co grains. By modeling the experimental magnetization curves, the distribution function of the superparamagnetic particle size, the volume fraction of the blocked part of the Co particles, and their saturation magnetization are obtained. δM plots are constructed using the remanence curves. A correlation between the changes of the δM parameters and those of the giant magnetoresistance, caused by the annealing, is discussed. © 1998 American Institute of Physics.

    Notes: We describe the design and performance of a compact ultrahigh vacuum sample holder, which allows (1) sample transfer, (2) liquid-nitrogen cooling, (3) heating by electron bombardment, and (4) temperature measurement in direct proximity to the sample. The device operates from close to liquid-nitrogen temperatures up to temperatures of 800–900 K in the steady-state regime with peak temperatures in excess of 1700 K and allows easy and reliable sample transfer. © 1998 American Institute of Physics.

    Notes: We present experimental and modeling results for the subgap absorption coefficient of intrinsic and doped hydrogenated amorphous silicon (a-Si:H) in order to explore the limitations of the constant photocurrent method (CPM). To properly model the subgap absorption coefficient, we have developed a simulation computer program including all the possible optical and thermal transitions between gap and extended states. Tail states are assumed to be either donor- or acceptorlike and midgap states are assumed to be amphoteric. The defect-pool model is also incorporated in our analysis. We have fitted several experimental dc-CPM spectra by using a standard set of parameters for the density of states. Our analysis demonstrates that in undoped samples the true absorption coefficient and the density of midgap states are underestimated by the CPM measurement. This is due to a nonconstant electron lifetime in the energy range between 0.8 and 1.2 eV. On the other hand, we show that in n-doped or p-doped samples, the CPM provides the correct answer. The energy range for the Fermi level where the CPM fails in a-Si:H is clearly established. © 1998 American Institute of Physics.

    Notes: Abstract The activity of heart interneurons (HN cells) and heart motor neurons in the central nervous system of the medicinal leech was recorded intracellularly from their cell bodies in the third and fourth segmental ganglion (G3 and G4, respectively). Reciprocal inhibitory synaptic transmission between HN cells in the G3 was blocked by photoinactivation of neuropil glial cells in the same ganglion. The block disrupted the alternating rhythmic spike activity of HN cells in the G3 in isolated G3s but not in chains of G3 and G4. In the latter case, the rhythmic spike pattern of HN cells in the G3 appears to originate in the G4 because, for example, severing one connective between the G3 and G4 silenced the ipsilateral heart interneuron in the G3, whereas its contralateral homologue remained rhythmically active. Simultaneous recordings from HN cells in the G3 and G4 suggest that the latter may serve to coordinate the rhythmic activity of HN cells in the G3, when synaptic interaction between HN cells in the G3 is blocked. This study reveals a considerable capacity of the neural network controlling the heart beat to compensate for the impairment of synapses within one ganglion.

    Notes: Abstract: The incidence of fractures and of osteoporosis differs between Oriental and Western Caucasian women. This may depend, at least in part, on nutritional factors, including dissimilarities in dietary intake of phytoestrogens. To investigate this possibility, 2-month-old female rats were ovariectomized (OVX) or sham-operated (SHAM), fed a casein-based diet, injected daily with subcutaneous genistein (GEN), the most abundant and best characterized phytoestrogen, or vehicle (Veh) and killed 21 days after surgery. As expected, ovariectomy resulted in loss of bone mineral density (BMD) and in uterine atrophy. However, administration of 5 mg GEN per gram body weight (b.w.) ameliorated the ovariectomy-induced loss of BMD (189 ± 2 mg/cm2 in OVX and 192 ± 2 in OVX with 5 mg GEN/g b.w. per day; p〈0.05). One microgram GEN per gram body weight did not affect the BMD loss and the effect of the 5 mg and 25 mg GEN per gram body weight were statistically not different. A trend toward reduced uterine atrophy (21% reduction) was noted with the 25 mg GEN dose, but not with the 1 mg and 5 mg doses. A separate experiment with 2 x 2 factorial design was conducted to elucidate the mechanism by which GEN ameliorates ovariectomy-induced bone loss. In this experiment, histomorphometry demonstrated a dramatic reduction in trabecular bone volume after ovariectomy (7.6 ± 0.7% of total bone volume in SHAM-Veh vs 3.3 ± 0.2% in OVX-Veh; p〈0.01) and less bone loss in OVX rats injected with 5 mg GEN per gram per day (3.3 ± 0.2% of total bone volume in OVX-Veh vs 5.2 ± 0.4% in OVX-GEN; p〈0.01). Administration of GEN was associated with higher bone formation rate per tissue volume and with a trend toward a higher number of osteoblasts per bone perimeter. The parameters of bone resorption were not affected by GEN. The concentration of serum osteocalcin and the urinary excretion of deoxypyridinoline provided corroborating results. Since production of proinflammatory cytokines is intimately involved in the pathogenesis of postmenopausal osteoporosis, the effect of GEN on lipopolysaccharide-induced in vitro production of Tumor necrosis factor-alpha (TNFa) was tested in monocytic cells from the same four rat groups. Production of TNFa was markedly elevated in OVX-Veh as compared with the SHAM-Veh rats, but this was blocked by GEN in the OVX rats. This study shows that GEN reduces both trabecular and compact bone loss after ovariectomy and that this protective effect differs from that of estrogen, since it depends on stimulation of bone formation rather than on suppression of bone resorption. Lack of action of GEN on uterine atrophy supports the possibility that this GEN dose affects target tissues via non-estrogenic mechanisms. Modulation of cytokine production may be involved in the effect of GEN on bone.

    Notes: Abstract In the vector Triatoma infestans the human pathogenic flagellate Trypanosoma cruzi colonizes mainly the rectum. The rectal cuticle and associated trypanosomes were examined by electron microscopy. The cuticle of the rectum consisted of a superficial wax layer, which was not retained by conventional preparation, an outer and inner epicuticle, and a procuticle. Epimastigotes of T. cruzi attached to the superficial layer with a specialized area of the flagellum. The composition of the cuticle was analyzed by cytochemistry to determine constituents relevant for parasite attachment. Intense staining with the fluorochrome Nile red indicated the presence of lipids, and measurements of contact angles formed by test fluids with the rectal wall revealed that the luminal surface is hydrophobic. The mechanism of attachment of T. cruzi was found to be based on a hydrophobic interaction. The flagellates bound to lipids extracted from the cuticle and to saturated hydrocarbons. Chitin, which has been presumed to be the natural binding substrate of T. cruzi, was localized using gold-labeled wheat-germ lectin. Chitin occurred in the procuticle but was absent from the superficial and epicuticular layers and, thus, is not accessible for binding by T. cruzi. In addition, it could not be confirmed that galactose-specific lectins or heparin receptors mediate flagellate attachment to the rectum.

    Notes: Human osteoblastic cells were grown in a three-dimensional (3-D) cell culture model and used to test the effects of a 20 Hz sinusoidal electromagnetic field (EMF; 6 mT and 113 mV/cm max) on collagen type I mRNA expression and extracellular matrix formation in comparison with the effects of growth factors. The cells were isolated from trabecular bone of a healthy individual (HO-197) and from a patient presenting with myositis ossificans (MO-192) and grown in a collagenous sponge-like substrate. Maximal enhancement of collagen type I expression after EMF treatment was 3.7-fold in HO-197 cells and 5.4-fold in MO-192 cells. Similar enhancement was found after transforming growth factor-β (TGF-β) and insulin-like growth factor-I (IGF-I) treatment. Combined treatment of the cells with EMF and the two growth factors TGF-β and IGF-I did not act synergistically. MO-192 cells produced an osteoblast-characteristic extracellular matrix containing collagen type I, alkaline phosphatase, and osteocalcin, together with collagen type III, TP-1, and TP-3, two epitopes of an osteoblastic differentiation marker. The data suggest that the effects of EMFs on osteoblastic differentiation are comparable to those of TGF-β and IGF-I. We conclude that EMF effects in the treatment of skeletal disorders and in orthopedic adjuvant therapy are mediated via enhancement of collagen type I mRNA expression, which may lead to extensive extracellular matrix synthesis.
