    Notizen: We investigated the accelographic train-of-four response evaluated at the great toe at varying stimulating currents. Fifteen adult patients undergoing elective general anaesthesia were studied. The mean current at which a supramaximal T1 value could be elicited was 〉 49 (9) mA [mean (SD)]. Ratios of accelographic T1 values at 50, 40, 30, 20 and 10 mA to accelographic T1 value at 60 mA were 0.90 (0.18), 0.58 (0.37), 0.38 (0.37), 0.19 (0.26) and 0.00 (0.00), respectively [mean (SD), p 〈 0.05 for 50 mA vs. 30, 20 and 10 mA, p 〈 0.05 for 40 mA vs. 20 and 10 mA, and p 〈 0.05 for 30 vs. 10 mA]. Threshold currents for train-of-four (the lowest currents at which any train-of-four response could be elicited) before and after induction of anaesthesia were 30 (10) and 31 (10) mA, respectively. The train-of-four ratio (T4/T1) measured at varying currents did not differ significantly. However, in the patients in whom threshold currents for train-of-four were 40, 30 and 20 mA, the train-of-four ratio recorded at the threshold current was significantly less than at 50 mA. We conclude that at the great toe, the mean current at which a supramaximal response to train-of-four can be yielded is as high as 〉 49 mA. The mean threshold currents for TOF before and after induction of anaesthesia were 30 and 31 mA, respectively. Train-of-four ratio measured at the threshold current is less than that at 50 mA.

    Notizen: 1. Effects of anti-Parkinsonian drugs on neurobehavioural changes induced by bilateral lesions of dopaminergic neurons were investigated in rats.2. Dopaminergic neurons in rats were lesioned bilaterally by injection of 6-hydroxydopamine (6-OHDA; 8 μg) into the medial forebrain bundle at the level of the posterolateral hypothalamus. As a result, a decrease in locomotor activity and marked catalepsy and prolongation of grasping time were observed.3. Levodopa, talipexole, bromocriptine and theophylline dose-dependently antagonized the decrease in locomotor activity induced by bilateral 6-OHDA lesions. These drugs also showed antagonistic effects on the appearance of catalepsy and prolongation of grasping time induced by bilateral 6-OHDA lesions. In contrast, trihexyphenidyl showed no antagonizing effect on the neurobehavioural changes induced by 6-OHDA lesions at any concentration tested.4. Combined treatment with levodopa and talipexole antagonized the neurobehavioural changes induced by bilateral 6-OHDA lesions, whereas no marked changes were observed when either drug was administered separately. The same findings were noted with the simultaneous use of either levodopa (2 mg/kg) and theophylline (2 mg/kg) or talipexole (0.005 mg/kg) and theophylline (2 mg/kg).5. These results indicate that this model may be useful for estimating the effects of drugs in the treatment of Parkinson's disease.

By means of peripheral computed tomography (pQCT), adult cortical bone density and volume was shown to be under a fixed rectilinear relationship regardless of age, sex, and presence or absence of osteoporosis. In children, however, the density-volume regression line followed a clearly different slope from that for adults (P 〈 0.001), indicating a difference in property and composition of the cortical bone between growing bone of children and grown-up bone of adults. Although both relative cortical volume and density increased with age in both boys and girls, no significant increase of trabecular bone was noted in either during the growth period. When the same technique was applied to the bone of rats known to continue growing with indefinite modeling without remodeling, the regression line between cortical bone density and volume was different from that of adult humans and similar to that of growing human children. Growing bone in a constant process of modeling and mineralization thus seems to have cortical bone possibly with less complete mineralization somewhat different from those of grown-up bone only undergoing remodeling. Selective cortical bone measurement by pQCT appears to be useful in characterizing the unique properties of the cortex of the growing bone.

    Notizen: Abstract  A novel ”in vivo cryotechnique” with replica immunoelectron microscopy was developed for detecting caveolin localization on replica membranes prepared directly from living smooth muscle cells. After quick-freezing mouse duodenal walls by our ”in vivo cryotechnique”, the specimens were prepared for freeze-fracture and deep-etch replica membranes. Then they were treated with 5% SDS and 0.5% collagenase to keep some antigens on the replica membranes. The immunogold method could be used to clarify the localization of the caveolin antigen in relation to three-dimensional ultrastructures of living smooth muscle cells. Our new cryotechnique can provide native organization of functional molecules in living cells.

    Notizen: Abstract The synthesis of [γ-32P]ATP with high specific radioactivity was investigated by the enzymatic method. We utilized a part of the glycolytic pathway which consists of three reaction steps. First, we examined the maximum radioactivity of H3 32PO4 without the decrease of labeling yield. Next, we examined the minimum amount of reagent without the decrease of labeling yield, because some reagents contain the phosphorus species as impurity. In this paper, we propose the optimum condition for synthesis of [γ-32P]ATP with high specific radioactivity. As our calculation, we will be able to obtain about 220 TBq/mmol specific radioactivity using 1.5 GBq of H3 32PO4.
