    Notes: A novel ridge waveguide laser diode has been developed in which the ridge is buried in a newly developed polyimide with a low thermal expansion coefficient close to that of AlGaAs which reduces the thermal stress to the junction and simplifies the wafer processing. A planar configuration, which is suitable for optoelectronic integration and an episide down mount for high-power operation, has been achieved by an etch-back process. Under cw operation, a low threshold current of 15 mA at 25 °C, a characteristic temperature T0 of 145 K, and maximum power output higher than 30 mW/facet have been obtained in a molecular-beam-epitaxial-grown graded-index separate confinement heterostructure single-quantum-well laser emitting at 780 nm.

    Notes: A new electron-beam annealing technique, an amplitude modulated pseudoline electron beam, has been proposed for recrystallization of large-area silicon layers on insulating materials (SOI). The technique utilizes an amplitude modulated sinusoidal wave for high-frequency beam oscillation. Through computer simulation of the temperature distribution for the sample surface, precise control of the position probability density profile of the line beam proved to be essential in realizing wide and uniform annealing. An optimum oscillation waveform was determined from the simulation. A large-area SOI, 4 mm (D'Alembertian), was successfully recrystallized.

    Notes: A new seed structure, partially thickened silicon films on insulator (PTS), is proposed for multilevel seeded-laterally epitaxial silicon films on insulator (SOI). Two-level seeded-laterally epitaxial silicon films on thick SiO2 (2.5 μm) were successfully fabricated, using the PTS structure in conjunction with the electron-beam technique. The SOI surface flatness after recrystallization was also improved by the PTS. Through computer simulation of the electron-beam recrystallization process, it is determined that the PTS structure enhances the lateral heat conduction within the SOI. This is important in smoothly melting the seed and field SOI regions.

    Notes: Hair treatment chemicals induce sudden and severe hair damage. In this study, we examined cuticles from untreated, permed, and bleached hair that were mechanically discriminated by shaking in water. Both perming and bleaching treatments are prone to easily delaminate cuticles. Confocal microscopy revealed that the cuticles of permed hair were delaminated with larger pieces than untreated ones. On the other hand, the cuticles of bleached hair tend to fragment into small peptides. At the minimum concentration of thioglycolate required to elute S100A3 protein from the endocuticle into the reductive permanent waving lotion, enlarged delaminated cuticle fragments were observed. Although S100A3 is retained in bleached hair, S100A3 is irreversibly oxidized upon bleaching treatment. It is likely that the oxidative cleavage of disulfide bonds between cuticle-constituting proteins, including S100A3, results in the fragile property of cuticles. Here we present a more comprehensive model of hair damage based on a diverse mechanism of cuticle delamination.

    Notes: Background:  Goblet cell metaplasia is a pathologic characteristic of asthma, associated with excess mucus secretion. Interleukin (IL)-4 and IL-13 plays an important role in mucus hypersecretion. Suplatast tosilate (suplatast), an antiallergic agent, is a Th2 cytokine inhibitor that suppresses the synthesis of IL-4, IL-5, IL-13, and eosinophilic airway inflammation.Objective:  We examined the effects of suplatast on mucus production in bronchial biopsy specimens taken from asthmatic subjects.Methods:  Oral suplatast 300 mg daily, or placebo was administered for 3 months in a double-blind, parallel-group study in 25 patients with asthma. Biopsy specimens were evaluated at before and after treatment for alcian blue/period acid-Schiff (AB/PAS), MUC5AC staining in bronchial epithelium and IL-4+, IL-13+ cells as well as inflammatory cells in lamina propria.Results:  There were significant decreases in the percentage of AB/PAS (P 〈 0.01) and MUC5AC (P 〈 0.01) stained area in the suplatast group. These changes were accompanied by significant decreases in IL-4+ and IL-13+ cells in suplatast-treated subjects. Additionally, we have observed that the number of infiltrating eosinophils and CD4+ T cells significantly decreased.Conclusions:  These findings suggest that suplatast prevents goblet cell metaplasia through modulation of Th2 cytokine production and the recruitment of eosinophils and CD4+ T cells in the asthmatic airways.

    Notes: Kimura's disease, eosinophilic granuloma of soft tissues, is a chronic inflammatory condition and appears as subcutaneous tumor-like nodules in the head and neck regions. Histopathologically, it is characterized by the presence of lymphoid follicles, marked infiltration of eosinophils, fibrosis and vascular proliferation. This disease occurring in the orofacial region is relatively rare, only 14 cases having been previously reported. We describe 3 additional cases of Kimura's disease and discuss these in relation to angiolymphoid hyperplasia with eosinophilia.

    Notes: The capacity of a roll of dentin demineralized by either 0.6N HCl, pH 1 for 3 minutes or 3 hours, or by 3M (9N) citric acid, pH 1, for 3 minutes, to induce cartilage and bone when implanted in muscle, was investigated. Serial sections of specimens were examined 7, 10, 14. 17, and 21 days after implantation, and randomly selected sections analyzed histomorphometrically. Cartilage was induced on the internal aspect of the citric acid-demineralized dentin roll, but significantly less than that induced after demineralization with HCl. The quantity of bone deposited subsequently did not significantly exceed the amount of cartilage that preceded it in relation to any of the preparations used. The results suggest that citric acid-demineralized dentin induces chondrogenesis and osteogenesis when implanted in muscle, but does so less effectively than does HCl-demineralized dentin, and only within the confines of a microenvironment. It is therefore unlikely that citric acid demineralization of root surface results in induction of cementum by this mechanism.

    Notes: Background  Dapsone (4,4′-diaminodiphenyl sulphone) is a powerful therapeutic tool in many skin diseases including neutrophilic dermatoses. The drug has an outstanding therapeutic efficacy against many skin diseases characterized by neutrophil-rich infiltrates; however, mechanisms of its action are poorly understood.Objectives  We investigated the effects of dapsone on respiratory and secretory functions of human neutrophils triggered by the chemotactic peptide N-formyl-L-methionyl-L-leucyl-L-phenylalanine (fMLP), the physiological agonist C5a, and phorbol myristate acetate (PMA).Methods  Human neutrophils were isolated from venous blood obtained from healthy donors. We detected extracellular production of superoxide (O2–) by cytochrome C reduction assay, and intracellular production of O2– by flow cytometry. Neutrophil elastase release was measured by the cleavage of the specific elastase substrate N-methoxysuccinyl-Ala-Ala-Pro-Val-p-nitroanilide. Measurement of cytosolic free calcium concentration was performed using the calcium-reactive fluorescence probe, Fluo-3.Results  Dapsone suppressed intra- and extracellular production of O2– and elastase release triggered by fMLP and C5a, but not by PMA. Both fMLP and C5a signalled the above pathways by inducing calcium influx, but PMA functions bypassed calcium influx. Dapsone was capable of antagonizing the induction of calcium influx.Conclusions  These findings suggest that one mechanism of the anti-inflammatory action of dapsone is inhibition of calcium-dependent functions of neutrophils including release of tissue-damaging oxidants and proteases in the affected skin.

    Notes: Abstract Using an antibody raised against d-glucose dehydrogenase (EC 1.1.99.17) purified from Pseudomonas fluorescens, immuno-cross-reactivity with the enzymes from several bacterial strains and localization of the enzyme in Escherichia coli were examined. The antibody cross-reacted with glucose dehydrogenases from various Gram-negative bacteria examined. As a result, it became apparent that the enzymes from Gluconobacter, Acetobacter, Pseudomonas and Acinetobacter, which existed as holoenzymes in the membranes, had lower molecular weights than those from E. coli and Klebsiella, which were apoenzymes.Treatment with trypsin of right-side out and inside-out membrane vesicles from E. coli clearly demonstrated that d-glucose dehydrogenase was located on the outer surface of the cytoplasmic membrane of E. coli, as had been suggested for Pseudomonas.

    Notes: Abstract This report describes a dot enzyme-linked immunosorbent assay (Dot-ELISA) for detecting circulating antigens in the sera of dogs infected with Dirofilaria immitis (D. immitis). Circulating D. immitis antigens could be detected in 24 of 25 infected dogs. The remaining animal had two immature worms. However, non-infected dogs and dogs infected with other parasites were all negative, Few cross-reactions to different parasite antigens were observed. The advantages of the Dot-ELISA include; 1) there is no need for pretreatment and dilution of sera and samples could be immediately bound to nitrocellulose paper set into microfiltration apparatus, 2) this assay could be carried out within 2 h at room temperature, 3) the resulting enzyme-reaction could be measured by both visual observation and densitometric reading.
