    Notes: Determination of HER-2 status is an essential prerequisite in considering a patient's eligibility for anti-HER-2 therapy; few studies have focused on its evaluation on cytological material. We present a new method of assessing HER-2 status on cytological samples, by fluorescence in situ hybridization (FISH) with an automated detection system, using fresh frozen (FF) and May–Grünwald–Giemsa (MGG) stained smears, and we evaluate the reliability of HER-2 determination on fine needle aspiration cytology (FNAC). The pre-treatment protocol for FF smears is easier and faster than for MGG stained slides. However, with the described procedure, FISH is also feasible on archival MGG stained slides. We conclude that with this method, cytological samples obtained by FNAC, either FF or MGG, are a reliable option for assessing HER-2 status.

    Notes: Contribution of carrier recombination from the p/i interface regions and the bulk to the dark current–voltage (JD–V) and short-circuit current–open-circuit voltage (Jsc–Voc) characteristics of hydrogenated amorphous-silicon (a-Si:H) p–i–n and n–i–p solar cells have been separated, identified, and quantified. Results are presented and discussed here which show that a maximum 1 sun Voc for a given bulk material can be validly extrapolated from bulk dominated Jsc–Voc characteristics at low illumination intensities. © 2000 American Institute of Physics.

    Notes: Hypertrophic scars and keloids are two forms of excessive cutaneous scarring. Considering the importance of extracellular matrix elements in tissue repair, a morphological and quantitative analysis of the elastic system components (fibrillin-1 and elastin) was performed in normal skin, normal scars, hypertrophic scars, and keloids. In superficial and deep dermis, fibrillin-1 volume density was significantly higher in normal skin compared with normal scars, hypertrophic scars, and keloids. The fibrillin-1 volume density did not show differences between hypertrophic scars and keloids in superficial or deep dermis. In superficial dermis, elastin volume density was higher in normal skin compared with normal scars, hypertrophic scars, and keloids. In deep dermis, the elastin volume density was higher in keloids compared with normal skins, normal scars, and hypertrophic scars. We showed that the distribution of fibrillin-1 and elastin is disrupted in all kinds of scars analyzed, but there are two patterns: one for normal scars and another for excessive scars.

    Notes: Multipotent stem cells (source for interfollicular epidermis, hairs and sebaceous glands) are localized in the bulge region of the outer root sheath of hair follicles, while stem cells giving rise to interfollicular epidermis reside in its basal. Using the multifunctional lectin galectin-1 as a marker to localize accessible binding sites in situ as a step to figure out galectin functionality in stem cells, we studied hair follicle-derived keratinocytes. Specific nuclear binding of galectin-1 associated with expression of ΔNp63α, a potential marker of epidermal stem cells, was detected. Binding of chimera-type galectin-3 to a nuclear site was not found in parallel assays. During the process of ageing in culture when cells acquire properties of senescence, disappearance of the nuclear signal for galectin-1 binding was accompanied by a similar decrease of nuclear ΔNp63α expression and increased binding of galectin-3 to the cell membrane, namely in regions of intercellular contacts. Expression of cytokeratin 10, a marker of the terminal differentiation was seen only in a small fraction of the cell population. These data extend the evidence for nuclear sites with galectin-1 reactivity in squamous epithelial cells, the expression of which is modulated upon senescence. Moreover, the results document the divergence of galectin-1 and -3 on the level of ligand selection in this cell type, underscoring the importance of the technical aspect to employ tissue lectins as probe and to perform a fingerprinting with several markers of the galectin family in parallel.

    Notes: Abstract Female Coelopa nebularum attempt to avoid mating with males by adopting up to three behavioral responses. Kicking and shaking are designed to dislodge mounted males, whereas downward abdominal curling prevents engagement of genitalia. We find that some females are more willing to mate than others, but their choice of rejection strategy is inconsistent. Mating does not affect behavior in immediately subsequent encounters. Male size influences the choice of rejection strategy and is positively associated with mating success. Different strategies have similar success rates. The response to a mount is not influenced by the size of a male encountered immediately beforehand. We propose that these results are inconsistent with adaptive mate choice or mate assessment and suggest that females are simply attempting to avoid the costs of mating with all males.

    Notes: Abstract Initial ligand selection and the intermolecular spatial arrangement ofglycan-lectin complexes are assumed to be essential to induce formationof stable cell aggregates by a lectin. To distinguish effects of thesetwo processes, the tetrameric mistletoe lectin and its isolated B-chainwere used. A reduced impact of multivalency for Ehrlich ascites tumorcells in contrast to rat thymocytes was revealed. Signaling is thusinitiated in a cell-type-dependent manner. Using selective metabolicinhibitors to reduce signal transfer for aggregate stability, decreasein cellular SH-group level was shown to be a common effect accompanyingsuppression of lectin-dependent aggregate stability. The resultsunderscore an intrinsic variability in the relative importance oflectin-dependent glycan aggregation on the cell surface for triggeringpost-binding lectin effects.

    Notes: The total synthesis of a neobetanidine derivative (3b) is described.Preliminary experiments led to the synthesis of neobetenamine (4), which presents the ring system of neobetanidine (3). A general method for the synthesis of several compounds containing the essential neobetanidine chromophore (a 1, 7-diazaheptamethine system incorporating a pyridine ring) consisted of Vilsmeier-Haack condensations involving the active (enolizable) methyl group of γ-picoline. Neobetenamine (4) resulted from this reaction with N-formyl-indoline, and also by an amine exchange between indoline and the Vilsmeier-Haack product from γ-picoline and N-methyl-formanilide.The methyl group of γ-picoline-2, 6-dicarboxylic ester 9, however, was resistant to the Vilsmeier-Haack condensation, but could be activated by introduction of a carboxyl into it: 4-chloropyridine-2, 6-dicarboxylic ester (11a) (from chelidamic ester) was used to alkylate malonic ester. The product (12a) lost only one carboxyl group when saponified. The resulting 2, 6-dicarboxy-pyridine-4-acetic acid (13a) readily underwent a novel decarboxylative condensation with the Meerwein acetal of dimethyl formamide to 4-(2-dimethylamino-vinyl)-2, 6-dimethoxycarbonyl-pyridine (14b), the first synthetic derivative of a neobetalaine. The enamine 14b was subjected to amine exchange reactions with indoline to 2-decarboxy-5, 6-dideoxy-neobetanidine dimethyl ester (15), and with (S)-cyclodopa (16) 5, 6-di-O-methyl-neobetanidine trimethyl ester (3b). The latter was identical with the diazomethane transformation product of betanidine (1), the aglucone of the pigment of the red beet, betanine.A few proton resonance and electron spectral properties, as well as the basicities of several of the synthesized compounds, are tabulated and discussed as far as they express special structural and electronic features of the common 1, 7-diazaheptamethine chromophore.
