    Notes: The role of impurities in melting of the charge-ordered (CO) state in half-doped manganese perovskites is discussed based on the systematic study of Cr and Al doping on the CO state as well as on the ferromagnetic–metallic (FM) state. The CO state in Nd0.5Sr0.5MnO3 is destroyed and the FM state is restored by 2% high-spin (S=3/2) Cr3+, whereas the CO state survives doping of spinless Al3+. On the other hand, the doping effects on the FM state in La1−xSrxMnO3 are almost identical between Cr and Al, except that Cr degrades Curie temperature less. The slight resistance of the CO state against Cr doping can be explained by the magnetic interaction between the Cr and the Mn t2g spins. © 2002 American Institute of Physics.

    Notes: Niobium tips for use in scanning tunneling microscope (STM) light emission measurements have been fabricated by an electrochemical etching method. The typical radius of curvature of the tip is less than 100 nm. The Nb tip has been demonstrated to be capable of producing STM images of highly oriented pyrolytic graphite and Au(110)-(2×1) surfaces with atomic spatial resolution. It has been confirmed by STM light emission spectroscopy that the front of the Nb tip becomes superconducting for temperatures below the superconducting transition temperature of Nb. © 2001 American Institute of Physics.

    Notes: Three pathways are recognized in the complement activation cascade. The aim of our study was to elucidate immunohistologically which complement pathway is associated with the activation in IgA glomerulonephritis (GN) and the relation of IgA subclass to the complement activation. Immunohistological staining was performed on biopsied renal specimens from 36 patients with IgA GN, 10 with systemic lupus erythematosus (SLE) and 16 with other glomerulonephritides using polyclonal antibodies of IgG, IgA, IgM, C3c, C4, C1q and monoclonal antibodies of IgA1, IgA2, mannose-binding lectin (MBL) and MBL-associated serine protease-1 (MASP-1). Mesangial deposits of IgA1, IgA2, C3c, C4, MBL and MASP-1 were detected in 19 of the 36 patients with IgA GN, and IgA2 and MBL/MASP-1 were colocalized in the mesangium in these 19 patients. The remaining 17 patients showed mesangial deposition of IgA1 alone. Twelve of these 17 patients presented mesangial deposition of C3c without deposition of C4, MBL and MASP-1. No deposition of C1q was evident in IgA GN patients. Three of the 10 SLE patients showed glomerular deposition of MBL and MASP-1 without deposition of IgA2. No patient with other glomerulonephritides showed glomerular deposition of IgA1, IgA2, MBL and MASP-1. There was no correlation in clinical and pathological indicators between IgA2-positive and IgA2-negative patients with IgA GN. In conclusion, alternative pathway-mediated complement activation is associated in patients with mesangial deposition of IgA1 alone in IgA GN. In those with the deposition of both IgA1 and IgA2, both alternative and lectin pathways are activated, and mesangial deposition of IgA2 is associated with the lectin pathway-mediated complement activation in IgA GN.

    Notes: Aim: To investigate the influence of water extracts of CagA-positive or -negative Helicobacter pylori on healing of chronic gastric ulcers and on connexin formation, proliferation and apoptosis, in acetic acid-induced chronic gastric ulcers in rats. Methods: Acetic acid was used to induce chronic gastric ulcers in rats, and a water extract of H. pylori was given by mouth every day. Connexin 32 formation was assessed using Western blotting as previously described. The frequencies of proliferating cell nuclear antigen staining and of TdT-mediated dUTP-biotin nick end labelling were examined. Results: In untreated rats, acetic acid-induced gastric ulcers healed after 14 days and the electrophoretic band corresponding to connexin 32 appeared 4 days after ulcer induction. Treatment with a water extract of H. pylori delayed ulcer healing, with the ulcers remaining unhealed even on the 14th day; healing was delayed more when treatment was with an extract of CagA-positive rather than CagA-negative H. pylori. Connexin 32 appeared earlier when treatment was with a CagA-negative rather than a CagA-positive extract, but in both cases later than in the untreated control group. Proliferating cell nuclear antigen labelling on the fourth day was seen in 14.5% ± 1.6% of mucosal cells of control group, but in 35.9% ± 1.4% and 36.5% ± 1.4% of mucosal cells treated with either VacA(+)CagA(–) and VacA(+)CagA(+) H. pylori extract, respectively. Furthermore, extracts of both H. pylori strains, especially VacA(+)Cag(+) H. pylori promoted apoptosis. Conclusions: A water extract of H. pylori increased both proliferation and apoptosis, which are related to excerbation and healing of ulcer as well as appearance of connexin 32.

    Notes: In Sakhalin taimen Hucho perryi two choriogenins have been identified in serum, designated high- and low-molecular-weight vitelline envelope-related protein (hVERP and lVERP). Two immunoassays were developed using specific antibodies; single radial immunodiffusion (SRID) and enzyme-linked immunosorbent assay (ELISA), for measurement of the precursors to vitelline envelope proteins. ELISA measures hVERP and lVERP in serum at concentrations as low as 16 and 47 ng ml−1, respectively. Using these immunoassays, changes in serum levels of VERP were measured after treatment with E2 and during oocyte growth. When immature 2-year-old taimen were injected with 2 mg E2 per kg body weight, both VERPs were induced in serum within 24 h. In the course of the first ovulation, serum VERPs levels increased in September (8 months before ovulation), reached maximum in January, and decreased sharply prior to ovulation in May. These changes generally paralleled that of vitellogenin except for the drastic decrease of VERPs in May.

    Notes: Enhanced expression of decay-accelerating factor and CD59/homologous restriction factor 20 in intestinal metaplasia, gastric adenomas and intestinal-type gastric carcinomas but not in diffuse-type carcinomas Aims: Variable expression of the complement regulatory proteins, decay-accelerating factor, CD59/homologous restriction factor 20 (HRF20) and membrane cofactor protein has been shown in human gastrointestinal malignancies, but their expression in gastric cancer has not been fully described. Thus, we immunohistochemically defined the distribution of these proteins in human normal gastric mucosa, intestinal metaplasia, adenomas and gastric cancers. Methods and results: Gastric tissues were obtained by endoscopic biopsy or surgical resection and stained with mouse monoclonal antibodies to decay-accelerating factor, CD59/HRF20, and membrane cofactor protein. In the normal gastric mucosa, membrane cofactor protein was diffusely stained on the basolateral surface of epithelial cells, whereas the expression of decay-accelerating factor and CD59/HRF20 was inconspicuous. In intestinal metaplasia, adenoma and intestinal-type gastric carcinoma cells, decay-accelerating factor and HRF20 were intensely stained on the apical surface; membrane cofactor protein retained its location on the basolateral surface. In diffuse-type gastric carcinomas, the expression of decay-accelerating factor, CD59/HRF20 was lost, but membrane cofactor protein was present on the tumour cell surface. Conclusions: These findings suggest that membrane cofactor protein plays a primary role in the regulation of complement activation in normal and neoplastic gastric cells and that the expression pattern of the complement regulatory proteins is closely related to gastric carcinoma development.

    Description / Table of Contents: Résumé Une spondylectomie totale en bloc a été effectuée sur 22 patients atteints de tumeurs vertébrales métastatiques. Des lésions ostéolytiques ont été décelées sur 15 sujets, tandis que les 7 autres présentaient des lésions ostéoblastiques ou mixtes. Nous avons comparé les dimensions des tumeurs vertébrales métastatiques mesurées avant l’opération par IRM et TDM aux dimensions mesurées immédiatement après l’opération lors de l’examen histologique de prélèvements vertébraux. Pour le groupe souffrant de lésions ostéolytiques, les diamètre dorsoventral transversal relatif de la tumeur métastatique mesuré par le IRM était plus élevé que celui déterminé au cours de l’examen histologique. D’un autre côté, ces mêmes paramètres mesurés à l’aide des scanners TDM affichaient aussi une différence importante. Bien que les différences entre les mesures des tumeurs ostéoblastiques et ostéolytiques n’aient pas été très grandes, le TDM a sous-estimé les dimensions des tumeurs ostéoblastiques ou mixtes plus que les tumeurs ostéolytiques. Nos données suggèrent une interprétation prudente de l’estimation réalisée par le IRM et le TDM concernant les dimensions de tumeurs ainsi que l’invasion locale.
    Notes: Abstract  Total en bloc spondylectomy was performed in 22 patients with metastatic vertebral tumors. Osteolytic lesions were present in 15 cases while 7 cases had osteoblastic or mixed lesions. We measured the dimensions of the tumors preoperatively using magnetic resonance imaging (MRI) and computerized tomography (CT), and postoperatively by histological examination of the resected specimens. In the osteolytic group, the dorsoventral and transverse diameters of the metastatic tumor as measured on MRI were significantly greater compared to those determined by the histological examination. On the other hand, the same parameters measured by CT were smaller than the histologically determined dimensions. Although there were no significant differences between measurements of osteoblastic and osteolytic tumors, CT underestimated dimensions in osteoblastic or mixed tumors more than in those of osteolytic tumors. Estimation of tumor dimensions and local invasion using MRI and CT should be interpreted carefully.

    Notes: Abstract. It is widely known that glucocorticoids induce and accelerate osteoporosis. High-dose glucocorticoids are administrated daily to patients in the acute phase of nephrotic syndrome. It could be inferred that high-dose glucocorticoids rapidly decrease patients' basal bone mineral density (BMD) and this accelerates the natural progress of osteoporosis associated with aging or menopause. Nine nephrotic patients (male/female: 5/4) without previous prednisolone administration were chosen to measure BMD and the level of the markers for bone turnover before and after treatment for 3 months (total prednisolone administration: 4.5 ± 0.0 g). Twenty-three patients under remission with prednisolone administration (male/female: 14/9) were included in the long-term treatment group. Patients in this group whose %YAM in the lateral lumbar spine was less than 89% were classified into a low BMD group (n = 10, male/female: 3/7). They were administered etidronate disodium at 200 mg/day for 14 days. BMD and % of young adult mean (YAM) in the lumbar spine (L2-L4 in lateral objection) and other regions were measured by dual-energy X-ray absorptiometry. As markers of bone metabolism, the urinary level of deoxypyridinoline (Dpd) was determined to evaluate osteogenesis, and serum osteocalcin was measured to evaluate bone resorption. BMD of the lumbar spine significantly decreased in the 3-month treatment group (752 ± 96 mg/cm2, 7 ± 4% reduction) compared with the pretreatment group (810 ± 85 mg/cm2). BMD in the long-term treatment group decreased continuously (683 ± 135 mg/cm2). No significant differences were noted in other measurement sites. BMD in the etidronate treatment group increased significantly (597 ± 55 mg/cm2) compared with the pretreatment group (549 ± 76 mg/cm2). Etidronate did not change BMD at the sites with a normal BMD. Among the biochemical markers (BM) examined, the urinary level of Dpd (nMol/liter · Cr) significantly increased in the 3-month treatment group (8.6 ± 5.1 nMol/liter·Cr) compared with the pretreatment group (5.8 ± 2.0 nMol/liter · Cr). No significant differences were seen in the BMs measured in the long-term treatment group. The urinary Dpd level of the etidronate treatment group decreased (3.9 ± 1.4 nMol/liter · Cr) compared with the pretreatment group. These data indicate that etidronate could improve the accelerated bone resorption. In conclusion, high-dose glucocorticoid therapy causes rapid bone resorption and accelerates the natural progress of osteoporosis associated with aging or menopause. Etidronate administration prevents the progress of osteoporosis in nephrotic patients. Preventive treatment should be performed when the estimated BMD in 3 months falls below the baseline by more than 7 ± 4%, reaching the therapeutic range.
