    Notizen: Proper composition and thickness of the InGaAs channel in pseudomorphic high electron mobility transistors (PHEMTs) is critical to assuring good device performance. Typically these characteristics have been measured by high-resolution x-ray diffraction. The results presented in this work show that the subband energy levels obtained from line shape analysis of room temperature photoluminescence spectra on these structures can be correlated very well with thickness and composition obtained from x-ray diffraction. Since the photoluminescence measurement and analysis is quite fast, this technique is suitable for rapid, nondestructive screening of PHEMT epitaxial material. © 1999 American Institute of Physics.

    Notizen: Restoration ecologists use reference information to define restoration goals, determine the restoration potential of sites, and evaluate the success of restoration efforts. Basic to the selection and use of reference information is the need to understand temporal and spatial variation in nature. This is a challenging task: variation is likely to be scale dependent; ecosystems vary in complex ways at several spatial and temporal scales; and there is an important interaction between spatial and temporal variation. The two most common forms of reference information are historical data from the site to be restored and contemporary data from reference sites (sites chosen as good analogs of the site to be restored). Among the problems of historical data are unmeasured factors that confound the interpretation of historical changes observed. Among the problems of individual reference sites is the difficulty of finding or proving a close match in all relevant ecological dimensions. Approximating and understanding ecological variation will require multiple sources of information. Restoration, by its inherently experimental nature, can further the understanding of the distribution, causes, and functions of nature's variation.

    Notizen: Abstract: Previously, we reported a modest but significant reduction in the concentration of neuropeptide Y in frontal cortices from victims of suicide relative to age-matched natural or accidental death control subjects. The reduction in neuropeptide Y appeared to be greatest in a subgroup of victims of suicide for which there was indirect evidence of histories of depression. We pursued these initial findings in the present study by measuring neuropeptide Y concentrations in frontal cortices from natural or accidental death control subjects and from suicide victims in whom a firm diagnosis of major depression was established by psychiatric autopsy. Because several subjects with major depression had a comorbid diagnosis of alcoholism, a group of victims of suicide that had an Axis I diagnosis of alcohol dependence was also studied. No significant differences in neuropeptide Y concentrations were observed between control subjects and victims of suicide with major depression or victims of suicide with alcohol dependence. These findings do not support a role for neuropeptide Y in major depression.

    Notizen: Abstract: β-N-Oxalylamino-l-alanine (l-BOAA), a non-protein neuroexcitatory amino acid present in the seeds of Lathyrus sativus (chickling or grass pea), is known to produce its neurotoxic effects by overstimulation of non-N-methyl-d-aspartate receptors, especially α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors, at micromolar concentrations. It has recently been reported that l-BOAA selectively inhibits mitochondrial enzyme NADH-dehydrogenase (NADH-DH) in brain slices at subpicomolar concentrations. The present study finds that up to 4 mM concentrations of pure l-BOAA fail to inhibit NADH-DH activity in mouse brain homogenate and isolated brain mitochondria. Two known inhibitors (rotenone and 1-methyl-4-phenylpyridinium ion, MPP+) of this mitochondrial enzyme produced significant inhibition under identical conditions. NADH-DH inhibition was also not observed in the homogenate or mitochondria from the brains of animals systemically treated with convulsive doses of l-BOAA. Some inhibition (20–37%) of NADH-DH activity was observed in mouse brain slices incubated with 100–1,000 µM concentrations of l-BOAA for 1 h at 37°C in an atmosphere of 95% O2 and 5% CO2, but the inhibition was nonselective, because the activity of another mitochondrial enzyme, succinic dehydrogenase, was similarly inhibited by l-BOAA. These results are in contrast with the report that l-BOAA inhibits mitochondrial NADH-DH selectively at subpicomolar concentrations. We suggest the observed nonselective NADH-DH inhibition in mouse brain slices treated with l-BOAA is caused by neuronal damage through an excitotoxic mechanism.

    Notizen: Abstract: Amphiphysin I and II are nerve terminal-enriched proteins that display src homology 3 domain-mediated interactions with dynamin and synaptojanin. It has been demonstrated that the amphiphysins also bind to clathrin, and we have proposed that this interaction may help to target synaptojanin and dynamin to sites of synaptic vesicle endocytosis. To understand better this potential functional role, we have begun to characterize clathrin-amphiphysin interactions. Using PCR from adult human cortex cDNA, we have cloned a number of amphiphysin II splice variants. In in vitro binding assays, the amphiphysin II splice variants display differential clathrin binding and define a 44-amino acid region mediating the interaction. Amphiphysin II truncation and deletion mutants identify two distinct clathrin-binding domains within this region: one with the sequence LLDLDFDP, the second with the sequence PWDLW. Both domains are conserved in amphiphysin I, and saturation binding analysis demonstrates that both sites bind clathrin with approximately equal affinity. The elucidation of clathrin as a splice-specific binding partner for amphiphysin II begins to address the potential functional role(s) for the multiple amphiphysin II splice variants and further supports an important function for clathrin-amphiphysin interactions in protein targeting during endocytosis.

    Notizen: Abstract : Neuropeptide Y (NPY) is an important regulator of energy balance in mammals through its orexigenic, antithermogenic, and insulin secretagogue actions. We investigated the regulation of endogenous NPY release from rat hypothalamic slices by NPY receptor ligands and calcium channel antagonists. High-potassium stimulation (60 mM) of the slices produced a calcium-dependent threefold increase in NPY release above basal release. The Y2 receptor agonists NPY(13-36) and N-acetyl[Leu28, Leu31]NPY(24-36), the Y4 agonist rat pancreatic polypeptide (rPP), and the Y4/Y5 agonist human pancreatic polypeptide (hPP) significantly reduced both basal and stimulated NPY release. NPY(13-36)-induced reduction of NPY release could be partially prevented in the presence of the weak Y2 antagonist T4-[NPY(33-36)]4, whereas the hPP- and rPP-induced inhibition of release was not affected by the Y5 antagonist CGP71683A or the Y1 antagonist BIBP3226. The selective Y1, Y2, and Y5 antagonists had no effect on either basal or potassium-stimulated release when administered alone. The calcium channel inhibitors ω-conotoxin GVIA (N-type), ω-agatoxin TK (P/Q-type), and ω-conotoxin MVIIC (Q-type) all significantly inhibited potassium-stimulated NPY release, without any effect on basal release, whereas nifedipine had no effect on either basal or stimulated release. Addition of both ω-conotoxin GVIA and ω-agatoxin TK together completely inhibited the potassium-stimulated release. In conclusion, we have demonstrated that NPY release from hypothalamic slices is calcium-dependent, involving N-, P-, and Q-type calcium channels. NPY release is also inhibited by Y2 agonists and rPP/hPP, suggesting that Y2 and Y4 receptors may act as autoreceptors on NPY-containing nerve terminals.

    Notizen: Seismic depth migration may result in false reflector positioning and destructive interference when an incorrect velocity field is used to convert from time to depth. The assumption of isotropy to describe anisotropic rocks is one major source of error in the velocity model, although individual survey images may not be impaired by such an approximation. When different survey types such as VSP and cross-hole reflection seismics have coincident illumination of the subsurface, it is important not only to produce consistent images upon depth migration, but also to determine a consistent velocity model. Using real data sets as examples, both objectives are successfully achieved when anisotropy is incorporated into the velocity model.

    Notizen: We have measured the two-photon absorption coefficent at 0.527 μm to be 1.8±0.4×10−3 cm/GW in a germanium-doped silica fiber. Using absorption and luminescence measurements we estimated the two-photon absorption coefficient at 1.053 μm to be smaller than 6×10−7 cm/GW. Using a capacitive technique we measured the photoconductivity to be approximately 6×10−9 1/Ω cm in the visible. The photoconductivity inreased linearly with intensity. A qualitative discussion of the defect dynamics related to photosensitive phenomena in optical glass fibers is given based on the experimental results. © 1996 American Institute of Physics.
