    Notizen: Three levels corresponding to photo energies E0(=1.7 eV), E1(=1.43 eV), and E2 (=energy gap) were observed in differential photocapacitance measurements of Si-doped AlxGa1−xAs grown by metalorganic chemical vapor deposition (0.23〈x〈0.4). On the other hand, E1 and E2 were observed but E0 was not observed in the differential photoconductance measurements. Both photon energies E1 and E2 cause persistent photoconductance (PPC) and the electron concentration coincides with the concentration of Si atoms. Our results support the broken-bond model proposed by D. J. Chadi and K. J. Chang [Phys. Rev. B 39, 10063 (1989)].

    Notizen: We present a new analytical representation of the lowest 3A‘ energy surface for the reaction O(3P)+HCl(X 1Σ+)→OH(X 2Π)+Cl(2P). This surface is derived by fitting ab initio calculations, and adjustments are made to the barrier height so that rate constants derived from accurate quantum scattering calculations match the room temperature thermal rate constants. Contributions to thermal and state resolved rate constants arising from reaction on the lowest 3A' surface are also considered. Comparisons of thermal and state resolved rate constants with experiment are good except at high temperature where the 3A' contributions are significant.

    Notizen: A new optical method is established to measure the crack length and its velocity in transparent crystals by utilizing the total reflection of light beam at the cleavage crack plane. The optical system is constructed by using a laser, a PIN photodiode sensor, and a wave memory. With the apparatus the crack propagation velocity can be measured at any desired position inside the specimen with a time resolution of less than 1 μs. The measurement on a cleavage crack in NaCl reveals that the maximum speed of the crack is about 800 m/s and the fluctuation of the velocity occurs prior to the arrest.

    Notizen: Expression of β-catenin was investigated in normal breast tissue and 66 breast carcinomas in conjunction with expression of epithelial cadherin (E-CD) and α-catenin. In normal mammary ducts and acini, intense β-catenin immunoreactivity was present at the basolateral surfaces of luminal epithelium and weak immunoreactivity was observed at the lateral borders of myoepithelial cells. No β-catenin was revealed at the myoepithelial basal surface. The intercellular expression of β-catenin, as well as of E-CD and α-catenin, was also observed in carcinoma tissues with varying staining intensity. Almost all of 10 intraductal carcinomas and approximately 70% of 41 invasive ductal carcinomas expressed the three molecules at the same level as in normal glands, whereas approximately 80% of 13 invasive lobular carcinomas showed severe deficiency of them. Two lobular carcinomas in situ showed complete absence of all of the proteins. Some of these findings were confirmed biochemically by immunoblotting analysis. In invasive ductal carcinomas, α-catenin was reduced more frequently in diffuse than in solid type tumours, whereas the level of expression of β-catenin and E-CD was unchanged between them. No correlation was present between reduced expression of the adhesion molecules and lymph node metastasis.

    Notizen: Summary The expression of SPRR (small proline-rich protein) was investigated in normal human skin and in diseased skin from patients with psoriasis, squamous cell carcinoma, basal cell epithelioma. Naevus pigmentosus, ichthyosis vulgaris and several inflammatory skin diseases, by immunohistochemical staining. A polyclonal antibody was raised against a synthetic peptide for a C-terminal common region for SPRR l and SPRR 3. In immunoblot analysis, a positive band of 18kDa was detected, which showed the presence of SPRR l in human epidermal keratinocytes. In normal epidermis, positive staining for SPRK was observed in keratinocytes in the granular layer and the uppermost or two spinous cell layers, with no staining of the other spinous or basal layers. The staining was obvious at the cell periphery, weak at the cytoplasm, and absent in the nucleus. Staining was observed in several outer layers of the follicular infundibulum to the isthmus. No staining was detected in the inner root sheath of the hair follicles, hair matrix, sebaceous gland, eccrine gland, eccrine duct, melanocytes. Langerhans cells or fibroblasts. The arrectores pilorum, striated muscles, muscle layers of vessels, and myoepithelia of eccrine gland, were weakly stained. In psoriatic skin, stained keratinocytes were distributed in the spinous cell layers except for the basal layer, in ichthyosis vulgaris. SPRR was barely expressed in the uppermost living cell layers of the epidermis in epidermolytic hyperkeratosis. degenerated squamous cells widely expressed SPRR. In Darier's disease, dyskeratolic cells were clearly stained. In squamous cell carcinoma, staining was observed in keratotic cells around horny pearls. In basal cell epithelioma, naevus pigmentosus, and malignant melanoma, the tumour cells or naevus cells were not stained. The distribution of SPRR was similar to that of involucrin in normal and several diseased skin, except for ichthyosis vulgaris. We conclude that SPRR is expressed in close association with epidermal differentiation in normal skin and skin diseases. The alteration of the expression of the proteins correlated to terminal differentiation, and differs from disease to disease.

    Notizen: As a part of the study on soil carbon flow in a deglaciated area in Ny-Ålesund, Svalbard (79°N), we estimated the contribution of the belowground respiration of vascular plants to total soil respiration in August 1996. Four study sites were set up along a primary successional series, ranging from newly deglaciated moraine to older moraine with well-developed vegetation cover. Respiratory activity of the belowground parts (roots + belowground stems) of three dominant species. Salix polaris, Saxifraga oppositifolia and Luzula confusa, was determined under laboratory conditions. The respiratory activity and the Q10 value of the respiration were higher in S. polaris than in the other two species. Total soil respiration rates measured in the field varied widely. The areas with dense vegetation cover tended to show high respiration rates. Belowground respiration of vascular plants was estimated based on the respiratory activity and biomass of the belowground parts at each study site. The contribution to the belowground respiration to total soil respiration was negligible in the early stages of succession. On the other hand, the respiration of the belowground parts contributed to a significant proportion (∼29%) of the total soil respiration in the latter stages of succession.

    Notizen: Histiocytic necrotizing lymphadenitis, also called Kikuchi-Fujimoto (KF) disease, is a benign disorder characterized histologically by paracortical necrotic foci surrounded by histiocytic aggregates. We analysed affected lymph node tissues from 34 patients with the disease in an attempt to elucidate its histogenesis. The ‘necrotizing’ cells showed typical apoptotic changes, including cell shrinkage and condensed and fragmented nuclei. Apoptotic bodies with a peculiar ultrastructure were demonstrated, and DNA fragmentation was detected in these cells by in situ end labelling. Immunostaining for the apoptosis-regulating proteins bcl-2, bax, c-myc and p53 failed to show their involvement in KF disease. However, perforin, a killer cell-specific cytolytic protein essential for provoking apoptosis in target cells, was found to be expressed abundantly by the infiltrating cells, which were thought to be cytotoxic T-lymphocytes. Perforin-expressing cells were present in the apoptotic foci of 28 of the 34 patients (82.4%). Virtually no cells containing perforin granules were present in non-pathological regions, lymph node tissues from control subjects with reactive or tuberculous lymphadenitis or those from patients with KF disease with negligible apoptosis. Therefore, the ‘necrosis’ associated with KF disease appears to be attributable to trans apoptotic death of the killer cell target in the affected nodes. We propose that KF disease should be called apoptotic lymphadenitis.

    Notizen: We report a case of β2-microglobulin-induced amyloidosis. The patient was a 40-year-old man suffering from non-amyloid nephropathy, who had been treated by haemodialysis for 20 years. Lichenoid skin lesions, consisting of groups of pin-head-sized shiny papules, were present on the arms and trunk. On histological examination, amyloid deposits were present, principally in the dermal papillae, hut also around the sweat ducts and hair follicles. The amyloid displayed potassium-permanganate-resistant Congo red affinity, and green birefringence under polarized light, Immuno-histochemically, β2-microglobulin was demonstrated in the lesions, confirming that they were a manifestation of β2-microglobulin-associated amyloidosis. Skin lesions of this type have not been reported previously in β2-microglobulin-associated amyloidosis.

    Notizen: Substance P is a neuropeptide present in, and released from, peripheral C nerve endings. The presence of substance P-positive nerve fibres in the epidermis has been reported. We investigated the effect of substance P on the transmembrane signalling system of pig epidermal keratinocytes. Treatment of pig epidermis with substance P resulted in an increase in inositol 1,4,5-trisphosphate (IP3), and in intracellular free calcium. The treatment also resulted in translocation of protein kinase C from a cytosol to a membrane fraction. Substance P, however, did not affect the β-adrenergic- or histamine (H2)- adenylate cyclase responses of the epidermis. Neither forskolin-induced, nor cholera toxin-induced cyclic AMP accumulation were affected by substance P treatment. These results are consistent with the view that substance P stimulates phosphatidylinositol-4,5-bisphosphate (PIP2) hydrolysis of keratinocytes, resulting in IP3-Ca2+ and diacylglycerol-protein kinase C signal activation. Although protein kinase C is known to affect the epidermal adenylate cyclase system, no evidence for such ‘cross-talk regulation’ was detected in keratinocytes by substance P treatment.
