    Notes: A study of children's help-seeking behaviour with respect to bullying and parental arguing is reported. A number of specific hypotheses and open-ended questions were investigated with boys and girls aged8-17 years. Female helpers were preferred overall, thought a relationship existed between the child's and helper's gender. Parents, friends, and teachers were all chosen, and parent and peer helpers were seen as complementary rather than competitive sources of help. Reasons for choosing helpers were notably varied, but largely related to perceived qualities of the helper rather than the child's own needs. Children depicted helping as an active response on the part of the helper. These findings, are discussed and implications of practice noted.

    Notes: The temperature dependence of threshold current between 130 and 310 K of 1.65 μm In0.53Ga0.47As–InP bulk lasers grown by chemical beam epitaxy has been measured. Comparison with a calculation of the spontaneous recombination current at threshold allows one to determine the proportion of current loss over this temperature range. It is found that the loss can be described using an Auger recombination component of the form Rauger=C'n3 exp(−Ea/kBT) where n is the carrier population density in the undoped active region. The activation energy Ea is found to be 39±5 meV which is in excellent agreement with the theoretical value for the conduction to heavy hole band/split-off to heavy hole band Auger process. The values obtained for the Auger coefficient C over the temperature range are in close agreement with published values obtained by time resolved photoluminescence. © 1995 American Institute of Physics.

    Notes: Deformation of plant cells by a fine probe can provide a measure of cell wall stiffness which is responsible for many of the mechanical properties of the plant. An example system of potato tuber parenchyma cells is used, and cell wall deformation approximated as the axisymmetric deformation of a circular membrane made from a nonlinear-elastic material with a Mooney strain energy function. A system of differential equations describing the shape of the membrane is derived and solved. The effects of turgor and initial cell wall strain on the force required to produce a given deflection and on the shape of the deformed cell are examined. Values for the elastic constants of the wall material are proposed, guided by statistical data for cell size and stiffness obtained from micropenetration tests.

    Notes: The aim of this study was to locate all ferruginous discharges within the Welsh coalfield areas and to assess their impact upon receiving watercourses. The project was undertaken in two stages. In the first stage, visual assessments were carried out on an area of river bed affected by iron hydroxide deposition. Chemical samples of the discharges and the receiving watercourse were analysed for a suite of physico-chemical determinands. Ninety discharges were located impacting upon 59.4 km of river, and an area of 22 ha was affected by iron hydroxide deposits.A ranking method, incorporating the physical/chemical determinands, was developed to assess the comparative impact which discharges were having on receiving watercourses. A total of 33 of the top ranked discharges- (20 to classified watercourses and 13 to unclassified watercourses) having the highest environmental impact were selected; chemical, biological and fisheries impact assessments were then carried out on these discharges in the second stage of the project. The sites were then ranked on the basis of biological and fisheries impact. A list of the highest impacted sites was produced, and some of these were further investigated for remediation options and associated costs.

    Notes: The structure of the catalytic core of the endoglucanase V (EGV) from Humicola insolens has been determined by the method of multiple isomorphous replacement at 1.5 Å resolution. The final model, refined with X-PLOR and PROLSQ, has a crystallographic R factor of 0.163 (Rfree = 0.240) with deviations from stereochemical target values of 0.012 Å and 0.037° for bonds and angles, respectively. The model was further refined with SHELXL, including anisotropic modelling of the protein-atom temperature factors, to give a final model with an R factor of 0.105 and an Rfree of 0.154. The initial isomorphous replacement electron-density map was poor and uninterpretable but was improved by the use of synchrotron data collected at a wavelength chosen so as to optimize the f′′ contribution of the anomalous scattering from the heavy atoms. The structure of H. insolens EGV consists of a six-stranded β-barrel domain, similar to that found in a family of plant defence proteins, linked by a number of disulfide-bonded loop regions. A long open groove runs across the surface of the enzyme either side of which lie the catalytic aspartate residues. The 9 Å separation of the catalytic carboxylate groups is consistent with the observation that EGV catalyzes the hydrolysis of the cellulose, β(1→4) links with inversion of configuration at the anomeric C1 atom. This structure is the first representative from the glycosyl hydrolase family 45.

    Notes: The laccase from the fungus Coprinus cinereus has been prepared and crystallized in a form suitable for X-ray diffraction analysis. Small plate-like crystals of an enzymatically deglycosylated form of the enzyme have been grown by the hanging-drop method using polyethylene glycol as precipitant. These crystals diffract to at least 2.2 Å. They belong to the space group P212121 with cell dimensions a = 45.4, b = 85.7, c = 143.1 Å with a single molecule of laccase in the asymmetric unit.

    Notes: Summary There is a significant body of evidence which suggests that the α-isoform of the CCAAT/enhancer binding protein (C/EBPα) plays a central regulatory role in energy metabolism in the liver. However, there is little information available regarding regulation of its expression in this tissue. In this study, we examined the effect of hormones and diabetes on its expression in rat H4IIE hepatoma cells and in rat liver. Treatment of H4IIE cells with dexamethasone led to a threefold increase in C/EBPα mRNA within 4 h. Insulin treatment produced a bi-phasic response, initially reducing mRNA levels up to the 4 h time point, but after 8 h a twofold increase in C/EBPα mRNA was observed. Treatment with 8-chlorophenylthio-cAMP produced a twofold induction of C/EBPα mRNA after 8 h. Western analysis indicated that the changes in mRNA in response to hormonal treatment generally resulted in corresponding alterations in C/EBPα protein levels. Finally, we observed an inhibition of C/EBPα gene expression in streptozotocin-diabetic rat liver, reflected by a decrease in both mRNA and protein levels that were partially reversed by insulin treatment. These results indicate that the expression of C/EBPα in liver is under complex control by both hormonal and metabolic signals, which is consistent with its role as a trans -regulator of genes which play a role in energy metabolism.     Notes: Abstract. Objective: We studied changes in cell surface morphology following treatment with secretagogue or trifluoperazine in a mast cell model.¶Materials and Methods: Rat basophilic leukaemia (RBL) cells were treated with antigen and or the calmodulin antagonist, 0–50 μM trifluoperazine (TFP). The release of a secretory granule enzyme, β-hexosaminidase, into the external medium was used as a measure of secretion. Quantitation of cell surface microvilli was determined by using a computer with input from a digitising tablet from scanning electron micrographs. Cytoskeletal proteins present in microvilli were analysed by confocal immunofluorescence.¶Results: When RBL cells are stimulated to secrete with an antigen, the cell surface is transformed from a microvillous morphology to a ruffled one. The cell surface rearrangement preceded β-hexosaminidase secretion: the majority of microvilli disappeared rapidly after stimulation (t1/2 of 39 s) whereas secretion can only be measured after a lag of 47 s. The calmodulin antagonist, TFP did not inhibit antigen-induced secretion or loss of microvilli, however TFP alone caused a similar loss of microvilli but was unable to stimulate or potentiate secretion. The microvilli mostly disappeared within 30 s, and a half-maximal effect occurred at approximately 8 μM TFP. Using immunofluorescence, calmodulin was localized to punctate structures on the dorsal cell surface which presumably correspond to the microvilli, and which also stained for F-actin and myosin I.¶Conclusions: Loss of cell surface microvilli on RBL cells precedes secretion and could reflect a cytoskeletal rearrangement which facilitates fusion of secretory granules with the membrane. It can be mimicked using trifluoperazine and we suggest it may involve calmodulin-binding components of the microvillus cytoskeleton such as myosin I.
