    Notes: Abstract The marine sponge Dysidea fragilis from El Mar Menor, a hypersaline coastal lagoon (Murcia, Spain), contains the furanosesquiterpenoid ent-furodysinin as the major secondary metabolite. D. fragilis emits a defensive white fluid when it is disturbed. Electron micrographs of this fluid revealed intact vesiculated cells together with other amorphous material. Dissociated cells are more rounded in shape but maintain the same ultrastructural features as cells observed in ultra-thin sections of the whole sponge. The defensive secretion is composed mainly of sponge cells with abundant light vesicles. Sometimes these light vesicles appear to open into the intercellular space; this correlates with surface blebs on these cells observed under scanning electron microscopy. The intracellular location of ent-furodysinin was confirmed by Erlich staining. In laboratory assays, we examined the role of ent-furodysinin as a feeding deterrent to generalist fish predators. It was isolated from D. fragilis and incorporated into a carrageenan-based artificial diet. The addition of ent-furodysinin to the artificial diet reduced feeding by the fish Thalassoma pavo. Similarly, fish did not feed on artificial diet above which defensive secretion of D. fragilis had been ejected with a small syringe.

    Notes: Abstract The marine fish pathogen Vibrio sp. 60 has been used as a host for heterologous expression of the Escherichia coli heat-labile enterotoxin B-subunit and derivatives carrying a C-terminal extension. In this study, a chimeric enterotoxin B-subunit with an extension corresponding to the carboxy-terminal nine amino acids -Tyr-Ala-Gly-Ala-Val-Val-Asn-Asp-Leu-cooH from the small subunit of herpes simplex virus type 1-encoded ribonucleotide reductase, is shown to be proteolytically cleaved in the extracellular medium by a single protease that is secreted by the host strain. Such protease behaves as a typical metalloprotease, being inhibited by EDTA but not by a serine protease inhibitor. Purification and amino acid composition analysis of the two proteolysis products revealed a specific cleavage of the peptide bond between amino acids glycine and alanine of the nine amino acid extension with loss of activity. The above observation is relevant for the biotechnological exploitation of Vibrio sp. 60.

    Notes: Depolarized Rayleigh spectra of water have been measured as a function of density along several isotherms in both liquid and supercritical phases. At all the thermodynamical states the spectral line shape can be fitted by a Lorentzian line superimposed to an exponential background. The behavior of the fitting parameters shows that at temperatures lower than ∼470 K the temperature is the most relevant parameter in determining the dynamics of the system, whereas above this temperature both temperature and density play a relevant role. Both the line shape and intensity of the spectra can be consistently interpreted in the framework of the first-order dipole induced dipole approximation to the collision-induced light scattering. The data show a clear evidence for cancellation effects. © 1995 American Institute of Physics.

    Notes: The hydrogen projected OH stretching density of states has been determined by an inelastic neutron scattering experiment in liquid and supercritical water. The results, compared with new measurements of the isotropic Raman spectra at the same state conditions, support the interpretation of the Raman spectra in terms of superposition of the allowed ν1 band with the overtone of the ν2 band. © 1998 American Institute of Physics.

    Notes: Abstract: The puromycin-sensitive aminopeptidase (PSA) is thought to contribute to the degradation of enkephalins. Besides being the most abundant aminopeptidase in the brain, PSA is expressed in other organs as well. From a human fetal brain cDNA library, we have isolated cDNA encoding the human PSA (huPSA) protein. The isolated cDNA gave rise to a protein with a molecular mass of 99 kDa. Compared with mouse PSA, homology at the amino acid and cDNA level was 98 and 93%, respectively. Translation of the huPSA was found to be initiated at the second of two possible start codons, as shown by studies with antibodies directed against peptide sequences of both potential N-terminal regions. Northern blot analysis with RNA isolated from different human organs demonstrated that the huPSA transcript is strongest but not exclusively expressed in the brain. Vesicular stomatitis virus epitope-tagged huPSA protein was expressed in HeLa cells and found to be localized in the cytoplasm, especially in the perinuclear region. By in situ hybridization, huPSA transcript could be identified in cortical and cerebellar neurons, whereas glial cells and blood vessels remained negative.

    Notes: Abstract Jerusalem artichoke is a favorable substrate for inulin or fructose syrup production. The sugar content and the fructose ratio of inulin depend on various factors, particularly on the date of harvest. Incomplete fermentation of extracts by selected yeasts allows the production of inulin with increased fructose content. The yeast strains (Saccharomyces cerevisiae, S. diastaticus...) are chosen for their ability to ferment sucrose and inulin small polymers, but not easily inulin large polymers. A good increase in the fructose ratio and a good yield in residual sugars can be obtained with the better strains. After fermentation and acid or enzymatic hydrolysis, extracts from “early” and “late” harvested tubers lead to syrups of good quality containing up to 95% and 90% of fructose respectively. This fermentative enrichment process is competitive with others (for example, chromatographic enrichment), is appropriate to raw extracts, simplifies the purification steps, and also permits the simultaneous benefit of production of by-products in the form of ethanol and yeast (in addition to the pulps). Unhydrolyzed inulin polymers with high fructose content can be recovered by this selective fermentation.
