    Notes: Summary The influence of exercise on glycaemia in the post-prandial state was studied for the first time in non-insulin-dependent diabetic (NIDDM) patients. Meal-induced glucose responses were followed for 8 h in 9 diet-treated patients with NIDDM. Subjects consumed a standardized breakfast and 4 h later a standardized lunch. They were studied in the resting state (control day (CD)) and on another day 45 min of bicycle exercise (53 ± 2 % V˙O 2 max (mean ± SEM)) was performed 45 min after breakfast (exercise day (ED)). On day 3 (diet day (DD)), the breakfast meal was reduced corresponding to the extra energy expenditure during the exercise period on ED. Responses were calculated as areas under the plasma concentration curve (AUC) during 4 h after either breakfast (B-AUC) or lunch (L-AUC). B-AUC for glucose was identical on ED (215 ± 63 mmol/l · 240 min) and DD (219 ± 60 mmol/l · 240 min) and on these days lower (p 〈 0.05) than on CD (453 ± 78 mmol/l · 240 min). L-AUC for glucose on CD, ED and DD did not differ significantly. B-AUCs for both insulin and C-peptide were also significantly lower on ED and DD as compared to CD (Insulin: 31337 ± 8682, 26092 ± 6457 and 47649 ± 15046 mmol/l · 240 min, respectively. C-peptide: 99 ± 19, 104 ± 26 and 195 ± 31 pmol/ml · 240 min, respectively). Rate of appearance (Ra) for glucose was unaffected by exercise whereas rate of disappearance (Rd) increased significantly. No differences in Ra or Rd were observed after lunch. In conclusion, postprandial exercise of moderate intensity decreases glycaemia and plasma insulin levels after breakfast in NIDDM patients, but this effect does not persist during and after the following lunch meal. Reduction of breakfast caloric intake has the same effect on postprandial glycaemia and insulin secretion as an equivalent exercise-induced increase in caloric expenditure.     Notes: Abstract Aims/hypothesis. The influence of postprandial high intensity exercise on glycaemia was studied in patients with Type II diabetes mellitus. Methods. Patients who were treated by diet only (n = 8) ate a standardised breakfast and 4 h later a standardised lunch. They were studied in the resting state (control day) and on another day (exercise day) when they did intermittent exercised at high intensity after breakfast) (4 bouts including 3 min at 56.5 ± 3.9 % V˙.O2 max (means ± SEM), 4 min at 98.3 ± 5.1 % V˙.O2 max and 6 min of rest). Responses were calculated as areas under the plasma concentration curve (AUC) during 4 h after either breakfast or lunch. Results. Breakfast-AUCs for glucose, insulin and C peptide were lower (p 〈 0.05) on the exercise day compared with the control day (glucose: 538 ± 94 vs 733 ± 64 mmol · l–1· 240 min; insulin: 16 ± 4 vs 22 ± 3 pmol · ml–1· 240 min; C peptide: 143 ± 22 vs 203 ± 29 pmol · ml–1· 240 min). After breakfast glucose appearance was unaffected by exercise, whereas disappearance and clearance increased (p 〈 0.05). Muscle glycogen was diminished by exercise (p 〈 0.05). After lunch no differences were observed between experiments. Exercise-induced reductions in glucose, insulin and C peptide responses were similar (p 〉 0.05) in this study of intermittent, high intensity exercise and in a previous study of isocaloric but prolonged moderate (45 min at 53 ± 2 % V˙.O2 max) postprandial exercise. Conclusion/interpretation. Postprandial high intensity exercise does not deteriorate glucose homeostasis but reduces both glucose concentrations and insulin secretion. The effect of exercise is related to energy expenditure rather than to peak exercise intensity. Finally, postprandial exercise does not influence glucose homeostasis during a subsequent main meal.     Notes: Abstract  A plant growth system with root-free hyphal compartments was used to examine the interactions between a mycophagous Collembola (Folsomia candida Willem), dry yeast and an arbuscular mycorrhizal (AM) fungus [Glomus caledonium (Nicol. & Gerd.) Trappe and Gerdemann] in terms of Collembola reproduction, AM-hyphal length and AM-hyphal P transport. Collembola reproduction was unaffected by AM mycelium, but a supplement of dry yeast increased the Collembola population size. The addition of dry yeast increased AM-hyphal P transport by increasing hyphal length. Collembola without yeast affected neither AM-hyphal growth nor AM-hyphal P transport, whereas Collembola with yeast decreased AM-hyphal P transport by 75% after 8 weeks. The hyphal density of G. caledonium remained unaffected by Collembola except after 4 weeks in combination with yeast, when a 33% reduction was observed. The results of this experiment show that the interaction between F. candida and the external mycelium of G. caledonium is limited under the conditions imposed.

    Notes: Abstract Objective: In vitro studies have shown that tacrine is metabolized by cytochrome P4501A2 (CYP1A2). One of the monohydroxy-metabolites has been incriminated with tacrine-induced hepatotoxicity. The aim of this study was to establish whether the potent CYP1A2 inhibitor fluvoxamine in clinically relevant doses could inhibit tacrine metabolism. Methods: Eighteen healthy young men were enrolled in an open, randomized crossover study. In the first study period a single oral dose of tacrine 40 mg was given. In the second period the volunteers were randomized to maintenance doses of fluvoxamine 50 or 100 mg per day, and a single oral dose of tacrine 20 mg was given. Results: Fluvoxamine was found to be a very potent inhibitor of tacrine metabolism. A fractional decrement in tacrine clearance of approximately 85% was found with both fluvoxamine doses, which was in good agreement with a prediction based on in vitro data. The medians of the steady-state concentration of fluvoxamine were 43 nM (range 25–49) and 70 nM (range 44–124) in the 50 mg per day and 100 mg per day groups, respectively. The steady-state concentration of fluvoxamine correlated with the fractional decrement in tacrine clearance (Spearman Rs = 0.53, P 〈 0.05). Modest, but statistically significant, reductions in the formation of the metabolites 1- and 2-hydroxytacrine were found during concomitant fluvoxamine treatment. Conclusion: Fluvoxamine at clinically relevant doses is a potent inhibitor of tacrine metabolism. This interaction is very likely to have clinical relevance. Whether concomitant fluvoxamine treatment reduces tacrine-induced hepatotoxicity needs further study.

    Notes: The influence of Ag doping on the transport properties of Y1Ba2Cu3O7−x thin films prepared by Y, BaF2, and Cu co-evaporation and optimized ex situ post annealing has been investigated. Both undoped and Ag doped films have values of Tc above 90 K, but Jc (77 K) is highly dependent on the nominal thickness (tnom) of the as-deposited film. For undoped films with tnom≤300 nm Jc (77 K) ((very-much-greater-than)106 A/cm2) decreases monotonically with increasing film thickness. Above 300 nm Jc (77 K) decreases rapidly to values below 5×105 A/cm2. Ag doped films with tnom≥200 nm have higher Jc (77 K) values than those of undoped films. Ag doped films have a maximum in Jc (77 K) around 250 nm. As for the undoped films, there is a large decrease in Jc (77 K) for Ag doped films with tnom≥300 nm. It was found that the higher values of Jc (77 K) for the Ag doped films were due to a better epitaxial growth of the YBCO compound. The low values of Jc (77 K) for both undoped and Ag doped single layer films with tnom≥300 nm were found to be due to the absence of 1–2–4 inclusions in these films. Based on these findings high Jc (77 K) films with tnom(approximately-greater-than)300 nm were grown by successive deposition and annealing of films with tnom〈300 nm on top of each other. A 2×150 nm undoped film was found to have a Jc (77 K) value of 4, 1×106 A/cm2. This is almost a doubling of the Jc (77 K) value as compared to the value for the 300 nm single layer undoped film (2,2×106 A/cm2). Ag doped double layer films of 2×150 nm and 2×215 nm had comparable Jc (77 K) values (5,8×106 and 5,6×106 A/cm2, respectively). In comparison with the undoped 2×150 nm film Jc (77 K) is thus further increased (by about 50%) when doping with Ag. © 1996 American Institute of Physics.

    Notes: In this study the CH4 dissociation probability on Ru(0001) is found for various translational and vibrational energies. The absolute sticking values are determined from King and Wells experiments and carbon uptake curves. The carbon amount is determined from the recombination signal of carbon with oxygen obtained after the beam exposure when heating in an oxygen atmosphere. The measured sticking coefficient of CH4 is strongly enhanced both by increasing the translational and the vibrational energy of the CH4 molecule. A model is applied to the data and an estimate of the thermal activation energy for CH4 dissociation is found to be in good agreement with previous bulb experiments. © 1999 American Institute of Physics.

    Notes: Alloying Au into the outermost atomic layer of Ni(111) significantly alters the physical and chemical properties of the surface. The reactivity is investigated by the use of seeded supersonic molecular beams of CH4 and it is found that the nobleness of Au reduces the overall reactivity toward CH4. This is accounted for in an ensemble model resolving the sticking probability on Ni atoms having different nearest neighbor surroundings. Although a mean field description of site distributions is found to be a very good approximation it is improved by using experimentally determined ensemble statistics from STM images. The strong influence of the vibrational temperature on the sticking coefficients of CH4 vs translational energy on the pure Ni(111) is also demonstrated. Desorption energies of CO and D2 is observed to decrease approximately 25–30 kJ/mole as the coverage of Au is increased from 0.0 to 0.7 ML. In TPD spectra of deuterium saturated surface alloys a new clearly resolved desorption state is observed at 180–220 K with maximum intensity around θAu=0.3–0.4 ML. This state is clearly related to chemisorption sites involving both Au and Ni. A site model based on mean field statistics adequately accounts for the appearance of this state. The effect of Au is also evident in the TPD spectra of CO saturated Au/Ni(111) surface alloys where the saturation coverage decreases and new structure develops. © 1996 American Institute of Physics.

    Notes: Background Allergen-specific T lymphocytes play an important role in the pathophysiology of atopic disease. Detailed studies of their epitope-specificity and crossreactivity are required for the development of novel approaches for specific immunotherapy.Objectives The aim of the study was to characterize the fine specificity of Bet v 1-specific T cells from allergic donors.Methods Polyclonal T-cell lines (TCL) and T-cell clones (TCC), specific for Bet v 1, the major birch (Betula verrucosa) pollen allergen, were isolated from the peripheral blood of three birch-allergic patients. Their epitope-specificity was studied using overlapping synthetic peptides, and crossreactivity with other tree pollen allergens of the Fagales order was evaluated. In addition, the Bet v 1-specific TCC were studied for their phenotype and cytokine production.Results All isolated Bet v 1-specific TCC (19/21 CD4+, 2/21 CD8+) reacted with affinity purified Bet v 1, but showed different reactivities with recombinant Bet v 1 (rBet v 1), and with group 1 allergens from other Fagales species. Epitope mapping of rBet v 1-reactive TCC with synthetic peptides of Bet v 1 showed the presence of four T-cell epitopes. Polyclonal T-cell lines reacted with 13 different peptides, and displayed even broader crossreactivity with group 1 pollen allergens from other Fagales members.Conclusion This study demonstrates that apart from T-cell epitopes of rBet v 1, many other crossreactive or Bet v 1 isoallergen-specific epitopes exist. This indicates that isoallergenic variation plays an important role in the induction of Bet v 1-specific and crossreactive T-cell responsiveness to allergens.

    Notes: SODA which stands for Système d'Organisation des Données et d'Analyse, is a general database for TdeV. SODA has the following goals: to produce a database of a reduced set of physical data; to ensure that these data are validated; to record all the parameters relevant to tokamak operation and experiments; to facilitate the retrieval of data using given selection criteria; and to improve data accessibility and analysis. The relational database ORACLE™ has been chosen to provide flexibility and to accommodate the increasing expectations of the TdeV researchers. In-house expertise allows custom-made tables and centralized data management. In the process of creating SODA several new interfaces for the scientific coordinator, machine operator, and diagnosticians have been added to provide a better definition of the experiment for the archiving system. The database includes the more relevant machine and diagnostic parameters, plasma perturbations (rf, biasing, gas...), mean and standard deviation of physical signals, plasma profiles, and code results (equilibrium...) for selected time windows in a discharge. Users of the X-window interface of SODA are not required to know the database structure or the SQL language. SODA has been operating successfully for over a year and its capabilities are continuously expanding. © 1997 American Institute of Physics.
