    Notes: Abstract: Activation of the N-methyl-d-aspartate (NMDA) receptor has been implicated in the events leading to ischemia-induced neuronal cell death. Recent studies have indicated that the properties of the NMDA receptor channel may be regulated by tyrosine phosphorylation. We have therefore examined the effects of transient cerebral ischemia on the tyrosine phosphorylation of NMDA receptor subunits NR2A and NR2B in different regions of the rat brain. Transient (15 min) global ischemia was produced by the four-vessel occlusion procedure. The tyrosine phosphorylation of NR2A and NR2B subunits was examined by immunoprecipitation with anti-tyrosine phosphate antibodies followed by immunoblotting with antibodies specific for NR2A or NR2B, and by immunoprecipitation with subunit-specific antibodies followed by immunoblotting with anti-phosphotyrosine antibodies. Transient ischemia followed by reperfusion induced large (23–29-fold relative to sham-operated controls), rapid (within 15 min of reperfusion), and sustained (for at least 24 h) increases in the tyrosine phosphorylation of NR2A and smaller increases in that of NR2B in the hippocampus. Ischemia-induced tyrosine phosphorylation of NR2 subunits in the hippocampus was higher than that of cortical and striatal NR2 subunits. The enhanced tyrosine phosphorylation of NR2A or NR2B may contribute to alterations in NMDA receptor function or in signaling pathways in the postischemic brain and may be related to pathogenic events leading to neuronal death.

    Notes: Abstract: We investigated the gene expression levels, the immunoreactive protein prevalence, and the functional activity of N-methyl-d-aspartate (NMDA) receptor complexes at early times after severe global ischemia challenge in rats. The mRNA expression levels for the NR2A and NR2B subunits of NMDA receptors changed to different degrees within different subregions of the hippocampus after reperfusion with respect to sham-operated control. No significant change in expression was observed in the vulnerable CA1 subfield at or before 6 h after challenge for either receptor subunit, although changes in expression in other hippocampal subfields were observed. At 12 and 24 h after challenge, significant decreases in expression for both subunits were found in the vulnerable CA1 subfield, as well as in other hippocampal regions. At the protein level, a significant decrease in the amount of NR2A/NR2B immunoreactivity in the total hippocampus was observed at both 6 and 24 h after reperfusion compared with sham control. Electrophysiological assessment of single-channel NMDA receptor activity in the CA1 subfield indicates that the main conductance state of NMDA receptor channels is maintained 6 h after challenge, although by 18–24 h after challenge, this main conductance state is rarely observed. The NMDA receptor component of the excitatory postsynaptic field potential was found to be significantly diminished from sham control 24 h after challenge, such that only ∼10% of the sham response remained, but was not significantly altered from sham control at 6 h after challenge. These results indicate that decreases in the expression levels, the immunoreactive protein prevalence, and that alterations in the functionality of NMDA receptors occur in the hippocampus at early times after severe transient global ischemia.

    Notes: Abstract : We have examined the effects of transient global ischemiaon both the gene expression levels and the functionality of GABABreceptors in rat brain, using antisense in situ hybridization andelectrophysiological evaluations. At the level of gene expression, nosignificant change in GABAB receptor expression was observed in anyhippocampal subfield at either 6 or 12 h after challenge. At 24 hpostchallenge, however, a significant decrease in GABAB receptorexpression was observed in both the CA1 and CA3 subfields, whereas no changewas observed in the dentate granule cell layer. Although expression in boththe vulnerable CA1 and less vulnerable CA3 subfields was diminished at thistime postchallenge, there was no significant difference in the degree of thediminished expression between these subfields. At the functional level, thedose-dependent ability of baclofen (1-100 μM) to inhibit an evokedexcitatory postsynaptic potential (f-EPSP) in the CA1 subfield was evaluatedat 24 h postischemia, in comparison with the dose-response observed insham-operated subjects. No significant differences were observed in theefficacy of GABAB receptor-mediated inhibition of the elicitedf-EPSP at any of the baclofen concentrations examined. These data demonstratethat although the mRNA expression levels for the GABAB receptor arediminished in both vulnerable and less vulnerable neurons of Ammon's horn at24 h following transient global ischemia, the functionality of theGABAB receptor system is maintained at this time postchallenge.

    Notes: Syringomas may be at least partially under estrogen and/or progesterone influence, as they are more common in women and are known to proliferate at puberty. During pregnancy and the premenstrual period an increase in tumor size has also been described. We examined nine syringomas using immunohistochemical markers for estrogen (ER) and progesterone (PR) receptors. Scattered tumor cells displaying nuclear and cytoplasmic staining for ER were noted in one of the nine cases. Intense nuclear and cytoplasmic staining for PR was noted in most (〉80%) of the neoplastic cells in 8/9 syringoma cases. Current immunohistochemical evidence supports the theory that syringomas are under hormonal control.

    Notes: Abstract  The callosal connections of ferret auditory cortex were studied by making multiple injections of wheat germ agglutinin-horseradish peroxidase into the middle ectosylvian gyrus or by packing crystals of horseradish peroxidase into the transected corpus callosum. The primary area (AI) had strong callosal connections that arose from somata mainly located in layer III. Other layers contained sparsely distributed cells that projected across the midline. The projecting cells occurred over the whole extent of AI but were not homogeneously distributed in layer III. The axons from these cells terminated mainly in the upper layers of the contralateral cortex, where they converged onto three discrete bands. The three elongated bands lay in a dorsoventral orientation, parallel to the tonotopic axis. They were slightly curved and had a fairly uniform width. The posterior band had a width of about 200 μm, while the anterior and middle bands were more variable and had widths of 300–800 μm. The centre-to-centre distance between the posterior and middle bands was 520 ± 60 μm and for the anterior to middle bands was 620 ± 210 μm. The retrograde labelling produced by the same injections showed that the cell bodies had a higher density in the terminal bands than in the intervening spaces. The bands of dense callosal connections appear to correspond to the binaural summation columns, which have been clearly demonstrated in the ferret, but direct evidence of this will need to be sought in a future study. The discrete nature of the callosal bands in the ferret appears to make it a suitable species for studying the relationship between callosal terminals and those arising in other areas of the brain and for clarifying the possible existence of separate functional systems within the auditory cortex.

    Notes: Abstract  Histological methods and 2-deoxyglucose autoradiography were used in an attempt at finding distinguishing characteristics that would permit the clear definition of different auditory areas on the ectosylvian gyrus. This region was studied in both coronal and flattened tangential sections. In tangential sections a crescent-shaped region of high deoxyglucose uptake was identified. The centre of this crescent was in the position of the primary auditory area on the middle ectosylvian gyrus. The ventro-anterior arm of the crescent was on the surface of the anterior ectosylvian gyrus and the ventro-posterior arm on the posterior ectosylvian gyrus. All three parts of the crescent appear to have an auditory function, because ablating the inferior colliculus or inserting a contralateral earplug reduced their deoxyglucose uptake. This was shown by using two separately distinguishable forms of 2-deoxyglucose, incorporating the 18F and 14C isotopes. In addition, another area of high deoxyglucose activity was identified in the ventral wall of the suprasylvian sulcus, which seems to correspond to the anterior auditory field. These four areas with high deoxyglucose uptake also have high levels of succinate dehydrogenase activity and moderately high densities of myelinated fibres. Succinate dehydrogenase histochemistry provides a simple method for identifying auditory cortical areas and should be of use in future physiological studies. These results provide evidence that the ferret has four separate auditory areas with relatively high metabolic and functional activity.

    Notes: Abstract Almost all patients with familial adenomatous polyposis develop duodenal polyps most of which occur in a cluster around the ampulla of Vater. Between 5 and 10% of FAP patients will die from upper gastrointestinal cancer, usually periampullary in origin. In an attempt to prevent cancer a screening programme has been developed using a well defined staging system to detect those patients most at risk of developing the disease. Treatment options are limited, with endoscopic clearance being contraindicated in most cases. The only certain method of preventing duodenal cancer is prophylactic radical surgery which has its own associated morbidity and mortality. Future developments may include new drug treatments or even gene therapy. Until then patients with FAP should all be considered for clinical trials as research continues.
