    Notes: . Using the polymerase chain reaction and arbitrarily selected oligonucleotide primers of 10 or 11 bases, we have amplified DNA sequences from Trypanosoma vivax parasites isolated from South America and Africa. On the basis of polymorphisms in the DNA fingerprints generated by three of the primers, the parasites could be separated into two major groups, one comprising T. vivax isolates from Kenya and the second including all the other T. vivax parasites (from Colombia, The Gambia, Nigeria and Uganda). One of these three primers (ILo 525) also gave isolate-specific DNA fingerprints for the parasites for the parasites tested, which will allow the use of this technique both in the species identification and discrimination of T. vivax parasites.

    Notes: Abstract This paper presents a conventionalist or modified contractarian perspective on constitutional and legal theory as a platform from which to address five important questions about the connections between critical morality and constitutional order. It finds that natural law and critical morality are inappropriately linked to constitutions and laws, but that there is nonetheless a clear moral dimension to all law. Furthermore even though law is explained as a function of human agreements, the very process of agreement commits law to an inherent set of standards which distunguish laws based on agreement from those which rest on coercion. These standards are more familiarly known as the principles of the rule of law.

    Notes: Summary Following a previous collaborative study (Fraccaro et al. 1980), 20 new cases of 11q;22q translocation are described. Twelve families were ascertained through an unbalanced carrier of the translocation and eight cases were ascertained as balanced carriers. A segregation analysis was performed on the 110 families so far published. It was concluded that the 11q;22q translocation is a relatively frequent event, and that all the cases thus far reported might have the same breakpoints at 11q23.3 and 22q11.2. The translocation seems to be independent of environmental factors and it seems to have a low rate of mutation as indicated by the scarcity of de novo cases. The new data confirmed that only one type of unbalanced karyotype (47,XX or XY+der(22)t(11;22)(q23.3;q11.2)) is found among the offspring of the translocation carriers. The minimal overall recurrence risk for an unbalanced translocation was estimated to 2%. There was no difference between the recurrence risks for male and female balanced carriers, while the trend was confirmed of an excess of female balanced carriers among the phenotypically normal offspring of the t(11;22) female carriers.

    Notes: Summary Pachytene analysis was undertaken in a male patient heterozygous for a 14q22q Robertsonian translocation. The relatively low rate of XY autosome association led us to examine the relationships existing between the chromosomes involved in the translocation, the rate of XY-autosome association and the degree of spermatogenic failure. Cytogenetic investigations in infertile men and the results of the meiotic studies suggest a direct correlation between the frequency of XY-autosome association at pachytene and the degree of spermatogenic failure. Whether associations arise as a consequence or cause of germ cell failure is still not certain.

    Notes: Summary Richard Levins introduced fitness sets as a tool for investigating evolution within heterogeneous environments. Evolutionary game theory permits a synthesis and generalization of this approach by considering the evolutionary response of organisms to any scale of habitat heterogeneity. As scales of heterogeneity increase from fine to coarse, the evolutionary stable strategy (ESS) switches from a single generalist species to several species that become increasingly specialized on distinct habitats. Depending upon the organisms' ecology, the switch from one to two species may occur at high migration rates (relatively fine-grained environment), or may only occur at very low migration rates (coarse-grained environment). At the ESS, the evolutionary context of a species is the entire landscape, while its ecological context may be a single habitat. Evolution towards the ESS can be represented with adaptive landscapes. In the absence of frequency-dependence, shifting from a single strategy ESS to a two strategy ESS poses the problem of evolving across valleys in the adaptive surface to occupy new peaks (hence, Sewell Wright's shifting balance theory). Frequency-dependent processes facilitate evolution across valleys. If a system with a two strategy ESS is constrained to possess a single strategy, the population may actually evolve a strategy that minimizes fitness. Because the population now rests at the bottom of a valley, evolution by natural selection can drive populations to occupy both peaks.

    Notes: Abstract Low density lipoprotein (LDL) processing has been investigated in the subcloned human colonic carcinoma cell line HT29-18. LDL binding at 4°C was a saturable process in relation to time and LDL concentration. The Kd for LDL binding was 11 μg/ml. ApoE-free HDL3 or acetylated LDL did not significantly compete with125I-LDL binding, up to 500 μg/ml.125I-LDL binding was decreased by 70% in HT29-18 cells preincubated for 24 hours in culture medium containing 100 μg/ml unlabelled LDL. Ligand blotting studies performed on HT29-18 homogenates using colloidal gold labelled LDL indicated the presence of one autoradiographic band corresponding to an apparent molecular weight of 130 kDa, which is consistent with the previously reported molecular weight of the LDL receptor in human fibroblasts. At 37°C,125I-LDL was actively internalized by HT29-18 cells and lysosomal degradation occurred as demonstrated by the inhibitory effect of chloroquine. LDL uptake and degradation by HT29-18 cells also resulted in a marked decrease in endogenous sterol synthesis. These data demonstrate that the HT29-18 human cancerous intestinal cells are able to specifically bind and internalize LDL, and that LDL processing results in down-regulation of sterol biosynthesis. Thus, intestinal epithelial cells possess specific LDL receptors that can be exploited to accomplish drug delivery and gene transfer via the receptor-mediated endocytosis pathway.
