    Notes: Abstract: A simple and efficient transfection technique based on lipopolyamine-coated DNA that can be used for gene transfer in cerebellar granular neurons is described. Gene transfer is achieved by exposure of cells to a DNA/lipid complex obtained by simple mixing of lipopolyamine and plasmid DNA. This procedure may represent a general tool of physiological investigations in primary cells. We show that the promoters of the introduced chimera genes are regulated by their respective trans-acting factors and may be modulated via membrane receptors and second messengers. This procedure has no noticeable toxic effects, nor does it seem to interfere with complex physiological behavior like neuronal differentiation.

    Notes: We show by scanning tunneling microscopy (STM) imaging that native oxide growth in moist air on hydrogen terminated Si(111) 1×1 surfaces begins by continuing formation of small oxide nuclei, 10–20 A(ring) in diameter, in the topmost Si layer. Their statistical distribution on the flat terraces points to a homogeneous nucleation process. Oxidation is extremely slow; after about 800 h only one complete monolayer is oxidized. In addition, a small number of three-dimensional oxide nuclei, several layers deep and 50–100 A(ring) in width, are formed at step edges as a minority species, which may be related to surface defects or contaminations.

    Notes: Postmitotic cerebellar granule cells, maintained for 5–6 days in Dulbecco's modified essential medium supplemented with 25 mM KCl, have been studied in whole-cell recording conditions to characterize calcium currents. With 10 mM Ba2+ as the divalent charge carrier, and using a pipette solution highly buffered for Ca2+ (30 mM EGTA, 100 mM HEPES–Tris, pH 7.2), only a high-threshold voltage-activated barium current was recorded from a holding potential of – 90 mV. The addition of 1 mM ATP to the pipette medium allowed stable recording for an average duration of 10 min, compatible with pharmacological studies of the barium current. Ninety-six per cent of the current was half-inactivated at low negative holding potential (-76 mV). A total block of current was obtained with 1 μM Cd2+. Sixty-three per cent of the mean current was abolished by 3 μM ω-conotoxin (ω-CgTx; Ki= 10 nM for a 15 min application), but individual cells showed either full sensitivity to this toxin or incomplete sensitivity. Seventy-eight per cent of the mean current was also abolished by 10 μM nicardipine but with a higher Ki, of 0.5 μM. After exposure to ω-CgTx, BAY K 8644 had no effect on the remaining current, though it was suppressed by nicardipine. No sensitivity to diltiazem, desmethoxyverapamil or flunarizine could be detected. Our major conclusion is that at least half of the channels have a mixed pharmacology, showing sensitivity to both ω-CgTx and dihydropyridine antagonists.

    Notes: The properties and characteristics of calcium channel openings in cerebellar granule cells were analysed by the cell-attached patch-clamp technique. At depolarized potentials, with 110 mM Ba2+ as the divalent charge carrier, 36% of the patches displayed activity that consisted of elementary events whose amplitude ranged from – 0.3 to –1. 75 pA at 0 mV, giving rise to a high threshold current. In this population of events at least four different types of channel openings were identified by their distinct biophysical and pharmacological properties. Two types of channel openings, with conductances around 24 and 7 pS, had similar characteristics in that both opened following two modes of gating characterized by brief (˜ 2 ms) and longer openings (˜ 8 ms) and both were sensitive to dihydropyridines. A further type of channel opening, with a conductance around 11 pS gated mainly with brief openings (∼1 ms), was shown to be insensitive to dihydropyridines but was undetectable in recordings from the cells that had been treated with ω-conotoxin. The last type of event was revealed after treatment of the cell with nicardipine or nifedipine and ω-conotoxin. The corresponding channel had a conductance of 19 pS and opened in one dominant mode characterized by brief openings (∼1 ms). The data obtained on single-channel activity of cerebellar granule cells are compared with the properties of the total current recorded in whole-cell conditions.

    Notes: Abstract Isolated bovine capillary endothelial cells have been examined for voltage-dependent Ca entry. All cells displayed a low threshold activity, with the main characteristics of a T-type transient current, when examined using whole-cell recording for activation and inactivation and cell-attached conditions or inside-out patches for the elementary conductance (8 pS). 25% of the cells displayed an additional sustained current in 5 mM CaCl2 above −40 mV, which was enhanced by application of BAY K 8644, but almost insensitive to superfusion with nicardipine. Two types of channels (2.8 and 21 pS, in 110 mM BaCl2) were shown to have a BAY K 8644 sensitivity. The large conductance channels were L-type channels. The smaller events were elicited at more hyperpolarized potentials (by some 30 mV). Their mean open time was 16 ms in control conditions. In presence of BAY K 8644, additional long open times were observed (up to 100 ms as compared to 7.8 ms for the time constants of the slow mode of the L-type channel). We refer to these channels as SB channels: of small conductance and sensitive to BAY K 8644. In the presence of nicardipine, SB channels are not noticeably modified, in contrast to the L-type openings which are abolished. Also, SB open times are close to control values when nicardipine is added after a BAY K 8644 application. We suggest that, at physiological concentrations of divalent ions, an SB-type activity is elicited above −40 mV which generates the low threshold sustained current.

    Description / Table of Contents: Zusammenfassung Ausgehend von NixMn3−x(C2O4)36H2O Oxalatmischkristallen führt die thermische Zersetzung bei vermindertem partialem Druck von Sauerstoff (po2 = 2%) zur Bildung von NiMn2O4 (x=1) unter metastabilen Bedingungen. Das nur in metastabilem Zustand existierende Ni1.5Mn1.5O4 (x=1.5) wurde ebenfalls hergestellt. Beide Spinellverbindungen sind vom stark umgekehrten Typ. Über einen Sol-Gel Herstellungsweg wurde gefunden, daß Mg2TiO4 ebenfalls in metastabilem Zustand gebildet wird. Tempern ergibt eine Zersetzung der Verbindungen, was zu NiMnO3 und 1/2Mn2O3 oder nur NiMnO3 oder MgTiO3 und MgO führt. Die Reaktionsgeschwindigkeiten sind für die Zersetzung von NiMn2O4 und Ni1.5Mn1.5O4 niedriger als für die von Mg2TiO4. Die Rückreaktion der Bildung von NiMn2O4 oberhalb 730°C zeigt eine endotherme Enthalpie von +61 kJ·mol−1. Für die Bildung von Mg2TiO4 oberhalb 1050°C wurde eine endotherme Enthalpie von +19.3 kJ·mol−1 gefunden. Die Ergebnisse wurden unter Berücksichtigung der Struktureigenschaften der Oxide diskutiert.
    Notes: Abstract Starting from oxalate mixed crystals NixMn3−x(C2O4)3·6H2O thermal decomposition at reduced oxygen partial pressure (po2= 2%) leads to the formation of NiMn2O4 (x = 1) at metastable conditions. Ni1.5Mn1.5O4 (x = 1.5) existing in the metastable state only has been also prepared. The spinel compounds both are of the highly inversed type. Following a sol-gel preparation route Mg2TiO4 has been also found to be formed in the metastable state. Annealing results in decomposition of the compounds providing NiMnO3 and 1/2α-Mn2O3 or NiMnO3 only or MgTiO3 and MgO, respectively. The reaction rates observed are lower for NiMn2O4 and Ni1.5Mn1.5O4 than for Mg2TiO4 decomposition. The reverse reaction of NiMn2O4 formation above 730°C shows an endothermic enthalpy of +61 kJ·mol−1. For Mg2TiO4 formation above 1050°C an endothermic enthalpy of +19.3 kJ·mol−1 is found. The results are discussed in terms of structural features of the oxides.

    Notes: Abstract Xerogel with cordierite composition Mg2Al3[AlSi5O18] from aluminium and magnesium 2-ethoxyethoxides and tetraethylortosilicate have been prepared, the latter being not previously partially hydrolized. Conditions to obtain not only homogeneous but also sinterable gel have been determined. All the powders derived were characterized by DTA, TG, BET and X-ray diffraction. Bulk α-cordierite material of 97% from theoretical density have been obtained by sintering.
