    Notes: The LipB protein of Pseudomonas glumae is essential for the production of active extracellular lipase encoded by the lipA gene. When lipase is overproduced in P. glumae in the absence of a functional lipB gene, the enzyme accumulates intracellularly in an inactive conformation. Heterologous expression of the lipase in Pseudomonas aeruginosa, Bacillus subtilis and Escherichia coli indicated that LipB is not directly involved in the trans location of the lipase across the inner or outer membrane. However, the presence of LipB was essential for obtaining active lipase and had a profound influence on the stability of the protein to proteolytic degradation. Inactive iipase, produced in the absence of LipB could be activated in vitro by unfolding and refolding, which demonstrates that LipB activity is not responsible for an essential covalent modification of the enzyme. We propose that LipB is a lipase-specific foldase. Furthermore, proper folding of the lipase in the periplasm appears to be essential for Xcp-mediated translocation across the outer membrane.

    Notes: Abstract Propane aromatization (530°C, 1 atm) was used as a reaction model to evaluate the effect of the calcination temperature on the catalytic properties of an as-synthesized [Ga1.3]-ZSM-5 zeolite obtained from alkali-free media and calcined at two different temperatures: 530°C (C-530) and 750°C (C-750). Results show that in spite of its lower acidity, C-750 is more active and selective toward aromatics than C-530. This is probably due to the fact that at higher temperature the decomposition of organic compounds used during the zeolite synthesis is accompanied by a partial degalliation of the zeolitic support leading to the production of a bifunctional x“Ga2O3” /H-[Gay-ZSM-5(2x+y=1.3)catalyst.

    Notes: Abstract Subacute necrotizing encephalomyelopathy (SNE) is a genetic disorder of pyruvate metabolism. Until recently the diagnosis of SNE could only be made at autopsy. However, an antemortem diagnosis can now be suggested by the correlation of clinical and laboratory data with computed tomography (CT) and/or magnetic resonance imaging (MRI). Five children with clinical and laboratory data suggesting the diagnosis of Leigh disease were evaluated by CT and MR. MR was found to be more sensitive than CT in the detection of areas of necrosis in the brain of the five children we studied. The absence of focal lesions detected by either modality in one of our patients did not exclude the diagnosis of SNE since focal lesions were present at autopsy one month following CT and MR.

    Notes: Summary Biofilm development in methanogenic fluidized-bed reactors with sand as the carrier was studied on a laboratory scale. The microorganisms present in consecutive layers of the biofilm of mature sludge granules were preliminarily characterized on the basis of their morphology, element composition and adhesion capacity and were compared to bacteria which take part in the initial colonization of sand. The early phase of biofilm development was monitored with reactors receiving waste-waters containing different mixtures of volatile fatty acids and inoculated with fluidized-bed reactor effluent for different lengths of time. The results obtained indicate that facultative anaerobic bacteria abundantly present in the outermost biofilm layers of mature sludge granules are probably the main primary colonizers of the sand. Methanothrix spp. or other methanogens were rarely observed among the primary colonizers. The course of biofilm formation was comparable under the various start-up conditions employed including variations in waste-water composition, inoculation and anaerobicity. However, omission of waste-water and thus of substrate resulted in rapid wash-out of the attached biomass.

    Notes: Abstract Although experiments in the rat suggest that glomerular hemodynamic alterations following a reduction of renal mass may be implicated in the progression of chronic renal failure, we argue that the deleterious effects of similar adaptations in human renal disease are unproven. In the otherwise normal solitary kidney the supranormal glomerular filtration rate (GFR) remains stable over the longterm, and in early diabetic nephropathy which is also accompanied by hyperfiltration, renal deterioration cannot be dissociated from a rise in systemic blood pressure. In patients with miscellaneous renal diseases and a depressed basal GFR there is indirect evidence that hyperfiltration might occur in some of the remnant glomeruli. However, at present there is little conclusive evidence to indicate that therapies which might normalize glomerular hemodynamics, e.g., dietary protein restriction, have any effect on progression of renal disease, or that angiotensin converting-enzyme inhibitors, which lower glomerular capillary pressure, have any advantage over other antihypertensive agents which are equally efficacious in lowering systemic blood pressure.

These include transgenic technology to introduce novel genes or delete existing genes and methods of gene transfer into the post-natal or adult kidney. The use of such technology has provided insights into renal development and growth and created new animal models of human diseases. Although some of these techniques are of potential use for introducing therapeutically useful gene products into the diseased kidney, many problems remain to be solved before this aim is attained.

    Notes: Abstract This article provides a series of historical reflections (for a period corresponding roughly to the past 20 or 30 years) on the evolving QOL status of older consumers. It uses a two-category QOL framework consisting ofobjective components (e.g., socioeconomic indicators, health and longevity, housing and family living arrangements, and gender differences) andsubjective components (e.g., life satisfaction and cognitive age) to explore the QOL of older consumers. It then examines the strategic reality of “diversity” and speculates on strategic implications of these QOL observations.

    Notes: Abstract To obtain specific DNA probes for the identification of the fish pathogen, Renibacterium salmoninarum, a discriminatory recombinant DNA library was constructed using selective fragments of the bacterial genome. Three renibacterial clones, pMAM29, pMAM46 and pMAM77, containing 149, 73, and 154 bp respectively, were isolated and characterized. The specificity of the probes was confirmed by dot-blot and Southern hybridization analyses. Bacterial hybridization experiments revealed that pMAM29 discriminates the R. salmoninarum genome from that of other fish pathogens such as Aeromonas salmonicida, Yersinia ruckeri, Flexibacter columnaris, Lactobacillus piscicola, Vibrio ordalii, Vibrio anguillarum and Aeromonas hydrophila. Thus, this probe may provide a new means to diagnose bacterial kidney disease in asymptomatic fish and ova.
