Notes: Antibody activity in the major classes and IgG subclasses against antigens in factory humidifier water was quantified by enzyme immunoassay (EIA) in 88 subjects who were exposed at work to the output from these contaminated humidifiers. Those with work-related symptoms had significantly higher mean titres than those who were symptom free, although values overlapped. The individuals with the highest IgG antibody titres also had the highest titres of IgM and IgA antibody, and these parameters did not discriminate between those with and without symptoms any better than the IgG titre. This was also true for the IgG subclasses where activity was predominantly measured in IgG 1. Quantifying the IgG antibody allowed us to demonstrate a significant correlation with years of work exposure (P〈0.001). There was no significant association between antibody and cigarette smoking, as assessed by smoking history and confirmed objectively by serum cotinine levels. There was a significant correlation with total IgG level (P〈0.001) suggesting that a non-specific immune enhancement may accompany the specific response. The antibody titres were followed up to 3 years after modification of the humidification systems, and during this time symptoms resolved and the antilunK levels progressively fell to undetectable levels. The EIA was adapted to measure antigen at nanogram levels thus providing a rapid test for screening of humidifier water as a technique that may help identify the nature of the antigens involved.

Notes: Abstract To investigate the biases associated with manta tow surveys of Acanthaster planci, counts obtained by manta-towed observers were compared with counts made on SCUBA swims under a limited range of conditions. Five 10 m wide strip transects on different parts of two reefs and with different densities of A. planci were surveyed. On average, 22.7% of starfish counted on SCUBA searches were counted on manta tows over the same transect (SD=12.0%, n=69). This proportion is termed sightability. As sightability decreases with increasing transect width, we estimate that, on average, less than 5% of the A. planci present are counted on routine manta tows which are conducted over transect of undefined width. Multiple regression analysis was used to determine which of 33 variables explained most of the variation in sightability in 10 m wide transects. The most influential variables were the proportion of cryptic starfish and an index of the degree of reef complexity. A regression equation designed to improve estimates of the abundance of A. planci on routine manta tows was developed. However, as it explained only 39% of the variation in sightability, this equation is of limited value in stabilising the negative bias associated with manta tow counts. In view of the variability of this bias, the manta tow technique is not suitable for estimating absolute densities of A. planci. Manta tow surveys are appropriate for identifying gross relative differences between densities of A. planci, and thus for determining broad-scale patterns of abundance. As such, they are a cost-effective method of estimating the geographical extent of A. planci outbreaks.

Notes: Abstract We examined the genetic, morphological, and molecular effects of position effect variegation inDrosophila, and the effects of mutations that either suppress [Su(var)] or enhance [E(var)] this phenomenon. All eightSu(var) mutations examined strongly suppress the inactivation of variegating alleles of the genes white [In(l) w m4 ], brown [In (2R)bw VDe2 ] and Stubble [T(2;3)Sb V ]. TheE(var) mutation enhances variegation of these loci. The chromosomal region 3C-E (26 bands) which includes the white locus is usually packaged as heterochromatin in salivary glands of the variegating strainw m4 . Addition of any of theSu(var) mutations restores a more euchromatic morphology to this region. In situ hybridization to polytene chromosomes and DNA blot analyses of gene copy number demonstrate that the DNA of thew + gene is less accessible to its probe in the variegatingw m4 strain than it is in the wildtype or variegation-suppressed strains. Blot analysis of larval salivary gland DNA indicates that the white gene copy number does not vary among the strains. Hence, the differences in binding of thew + gene probe in the variegating and variegation-suppressed strains reflect differences in chromosomal packaging rather than alterations in gene number. The effects of variegation and theSu(var) mutations on chromatin structure were analyzed further by DNAse I digestion and DNA blot hybridization. In contrast to their dramatic effects on chromosomal morphology and gene expression, theSu(var) mutations had negligible effects on nuclease sensitivity of the white gene chromatin. We suggest that the changes in gene expression resulting from position effect variegation and the action of theSu(var) mutations involve alterations in chromosomal packaging.

Notes: Abstract We have measured the resistive upper critical field of overdoped single crystals of Tl2Ba2CuO6 from the zero-field transition temperatureT c (approximately 20 K) to temperatures as low as 12 mK, corresponding to less than 0.001T c. In sharp contrast to the predictions of standard theories of superconductivity, the critical field is found to rise steeply with positive curvature as the temperature is reduced, and no sign of saturation is observed down to the lowest temperatures reached.

Notes: Abstract The influence of low-temperature annealing, at 〈 360 °C, in various reducing and oxidizing atmospheres for a series of BaTiO3 ceramics with a positive temperature coefficient of resistance (PTCR) is discussed. Combined impedance and modulus spectroscopy is used to analyse a.c. impedance data and shows that the total resistance of the sample can be composed of up to three components, dependent on the cooling rate from the sintering temperature. For quickly cooled samples the PTCR response is dominated by an outer shell on individuals grains, whereas for slowly cooled samples the grain boundary resistance dominates. Annealing in reducing atmospheres destroys the grain boundary PTCR effect whereas the outer-shell grain PTCR effect is relatively insensitive to the reducing atmosphere. It is proposed that the acceptor states responsible for the outer-grain and grain-boundary PTCR effects are predominantly intrinsic metal vacancies, i.e. Ba and/or Ti, and adsorbed oxygen, respectively.
