    Notizen: A cybernetic framework is presented which views microbial response in multiple substrate environments as a judicious investment of cellular resources in synthesizing different key proteins according to an optimal regulatory strategy. A mathematical model is developed within the cybernetic framework for the diauxic growth of Klebsiella pneumoniae on a mixture of D-glucose and D-xylose. The “bang-bang” optimal policy describes well the experimental observations obtained using a fermenter coupled to an Apple II microcomputer. Striking variations in respiratory levels are observed experimentally during the switching of the cell's adaptive machinery for the utilization of the less preferred substrate.

    Notizen: Plasmid loss kinetics for Saccharomyces cerevisiae transformed with the 2-μm DNA-based-plasmid pUCKm8 were measured in nonselective and selective media. The plasmid pUCKm8 gives the organism two new phenotypes: resistance to the wide spectrum antibiotic G418 sulfate, and the ability to produce the enzyme, β-lactamase. Plasmid stability was determined using the production of β-lactamase as a marker. The effect of G418 on the growth rates of all organisms present in the culture and on plasmid stability was also determined. Mathematical models describing plasmid loss kinetics during exponential growth for both nonselective and selective conditions are used to simulate the experimental data. In nonselective medium, over 80% of the cells still exhibited the desired phenotype after 50 doublings. In medium containing G418, improvements in plasmid stability were only marginal due to the appearance of antibiotic-resistant cells.

    Notizen: Summary Porous glass fiber has a very high surface area and good mechanical properties that make it an excellent support for biocatalyst immobilization. By packing aligned glass fibers in a tubular reactor such that the fibers are all parallel to the axis of the tube, the resulting pressure drop is considerably smaller than for a similar bed of packed beads. The utility of this support was demonstrated by immobilizing α-glucoamylase by silane-glutaraldehyde coupling, and measuring its activity toward converting maltose to glucose. Using optimized immobilization conditions, an enzyme loading of 1.5 mg protein perm 2 surface area was obtained, with an activity of 370 units/g glass at 50°C. The half-life of the immobilized glucoamylase was more than twice as long as that of the free enzyme.

    Notizen: Summary The main product of fermentation byKlebsiella oxytoca is 2,3-butanediol. This organism also produces acetic acid, ethanol, and acetoin. In this report, product inhibition due to 2,3-butanediol and acetic acid is considered. Although the acetate ion has little effect on growth, acetic acid is a strong inhibitor. Acetic acid inhibits growth more strongly than it inhibits respiration. The neutral product 2,3-butanediol is not a strong inhibitor; its effect on growth is no more than is expected by the decrease in water activity it causes. The effect of 2,3-butanediol on respiration can also be explained by a decreased water activity. It appears that it is possible to accumulate as much as 130 g/L butanediol while as little as 0.45 g/L acetic acid completely inhibits growth.
