    Notes: The antibody response to human gamma globulin (HGG) and Aeromonas salmonicida (AS) in control and long-term thymectomized rainbow trout was individually monitored in 120 fish. The fish responded poorly to HGG and no memory response was observed. There was no difference in response between control fish and fish which had been thymectomized for 5 months before immunization. However, 9-month thymectomized fish had lower titres than controls. The response to AS was vigorous and significant memory was demonstrated after a second injection. There was no difference in response between control and thymectomized fish. The individual variation in response was not related to body size, antigen dose, sex or reproductive state. The results are discussed in terms of the thymus dependency or independency of antigens in fish, and the role and life span of T cell subsets, such as ‘helper’ and ‘;suppressor’ cells.

    Notes: The time to safe return of neuromuscular function after atracurium 0.3 mg/kg intravenously was assessed in 24 patients in whom anaesthesia was maintained with halothane 0.5%. Safe reversal (recovery to a T4 ratio 〉 05), after this dose of atracurium, could only be reliably and rapidly (〈 2 minutes) achieved with edrophonium if a period greater than 30 minutes elapsed since administration of the relaxant. This coincides with the appearance of four recognisable twitches if a train-of-four pattern of nerve stimulation is used. Thus, if no monitoring equipment is available, at least half-an-hour should elapse after administration of atracurium in a moderate dose (e.g. 0.3 mg/kg) before rapid and reliable reversal can be anticipated. Four twitches in a train-of-four should be recognisable, if a nerve stimulator is available.

    Notes: Percutaneous tracheostomy is a well established technique used primarily to assist weaning from mechanical ventilation on many intensive care units. We report our experiences of a total of 36 procedures performed with the new Blue Rhino™ Percutaneous Tracheostomy Introducer Set developed by Ciaglia. The technique was successful in all cases and was simpler and quicker to perform than with the earlier Ciaglia percutaneous tracheostomy set. Difficulties were encountered when using Shiley™ tracheostomy tubes. Significant complications included one posterior wall tear and one tracheal cartilage ring fracture.

    Notes: Tetracapsuloides bryosalmonae is the myxozoan parasite that causes the commercially important proliferative kidney disease (PKD) in salmonid aquaculture. Previous studies on the binding of lectins to T. bryosalmonae identified Griffonia simplificola agglutinin I (GS I) as useful for parasite identification. This lectin was also implicated as recognizing antigenic structures on the parasite. Here, we examine the histochemical staining and ultrastructural localization of a panel of 21 lectins on the extrasporogonic stage of T. bryosalmonae. The histochemical staining studies indicated that the majority of lectins bound to the renal stages of T. bryosalmonae, however not all of these lectins could be successfully localized using immunogold electron microscopy. Of the lectins that were localized many, including GS I, bound to membranes associated with the lysosomal pathway within the extrasporogonic primary cell of the parasite, indicating that these organelles are rich in glycoconjugates. The histochemical staining of Erythrina cristagalli ECL was unique and highlighted a different distribution of glycoconjugates in the periphery of some extrasporogonic parasites within the renal sinuses when compared with stages in the interstitium, suggesting the presence of distinct blood forms of T. bryosalmonae.

    Notes: Photobacterium damsela ssp. piscicida (Phdp) isolates were grown in various bacteriological media, in eukaryotic cell culture media and in the presence of fish cells (resembling some aspects of in vivo growth environments). Bacterial cells, extracellular products (ECPs) and crude capsular polysaccharide were isolated and analysed by electrophoresis and Western blot using sea bass sera. Growth in bacteriological media conserved the synthesis of cell and extracellular components when these were compared with those prepared under near-in vivo growth conditions. In fact, synthesis of a larger range of cell components was induced after growth in bacteriological media. Certain media based on yeast extract and peptones from various sources and a specific salt formulation induced the synthesis of novel cell components at approximately 21.3 and 14 kDa. These antigens were recognized by sea bass sera collected after natural pasteurellosis outbreaks and other sea bass sera raised against live or inactivated Phdp cells. The ECPs of the pathogen were not good immunogens in their soluble form despite various treatments prior to immunization. The results are discussed with respect to vaccine development.

    Notes: The production of macrophage activation factor (MAF) by rainbow trout, Oncorhynchus mykiss (Walbaum), head kidney leucocytes was examined after culturing in vitro with extracellular products (ECP) collected from Mycobacterium sp. Cultures of leucocytes were prepared from naive fish, or fish previously vaccinated with either the ECP or with formalin killed whole cell preparations (WC) of the bacterium. The cells were then incubated with the ECP in vitro and the ability of their supernatants to activate macrophages assessed. Macrophages from control fish were incubated with the supernatants, and their ability to reduce nitroblue tetrazolium (NBT) measured as an indicator of macrophage activation. Incubation of head kidney macrophages from naive fish directly with 1, 10 or 100 μg mL–1 of ECP for 48 h significantly enhanced macrophage activation compared with control macrophages. Vaccination of fish with either ECP or WC had no significant effect on the respiratory burst of control macrophages 4 weeks post-vaccination. By the eighth week, however, absorbance levels of respiratory burst reflecting both the primary (cells from vaccinated fish cultured in vitro with PBS) and the secondary (cells cultured in vitro with ECP) MAF responses of fish vaccinated with ECP and WC, had peaked and these were significantly different from the non-vaccinated controls. This activity had fallen to levels similar to control fish by week 12 for fish vaccinated with WC.

    Notes: About 10% of the large (L) protein gene of Strawberry crinkle virus (SCV) was sequenced after amplification with degenerate primers designed to conserved regions of the rhabdovirus L protein. The virus sequence was extended to 1362 nucleotides through rapid amplification of cDNA ends. One pair of degenerate L gene primers amplified a 683-bp fragment from four different isolates of SCV cultured in the experimental host Physalis pubescens; the nucleotide sequences of these fragments differed by 〈 1% to 10% indicating the suitability of this region as a diagnostic target. This information enabled the development of a reverse transcription polymerase chain reaction (RT-PCR) detection method for SCV using primers designed to the L gene sequence. SCV was amplified from infected P. pubescens (573 bp fragment) and from infected Chaetosiphon fragaefolii aphids (770 bp fragment). SCV was also detected by RT-PCR in total RNA extracts from three strawberry plants showing symptoms typical of SCV infection but failed when the intensity of the disease symptoms decreased. However, both SCV positive-sense RNA, and negative-sense genomic RNA, were detected by nested PCR in chronically infected strawberry plants sampled in September.
