    Notizen: In Xanthomonas campestris cultures the xanthan formation is reduced by about 40% by the addition of methionine in quantities as small as 1.3 mM. Structural analogues of this amino acid exhibit a similar inhibitory effect on polysaccharide formation. Xanthan formation is also influenced in non-growing cells: cells precultured with methionine synthesize less xanthan than cells precultured without methionine. It therefore follows that apparently the composition of the cell is altered resulting in an altered flow of glucose. In accordance with this, cells grown with methionine excrete a greater portion of carbohydrates with Mrs smaller than xanthan.

    Notizen: Summary The regulation of the synthesis of alcohol oxidase, catalase, formaldehyde dehydrogenase, and formate dehydrogenase was investigated in the methanol-utilizing yeastHansenula polymorpha during growth on different carbon and energy sources. When cells were grown on glucose, the enzymes of the dissimilatory methanol metabolism were not detected during the exponential phase of growth, but were formed in the late stationary phase without addition of methanol. Moreover, the enzymes were synthesized during growth on sorbitol, glycerol, ribose, and xylose. It was shown that the carbon catabolite insensitivity of the synthesis of methanol-specific enzymes is not limited to substrates that are slowly metabolized.

    Notizen: Abstract When Corynebacterium glutamicum ATCC 14310 (leu-) was cultured with 200 mg/l leucine and 150 mM α-hydroxybutyric acid the acetohydroxy acid synthase activity was increased to 0.17 U/mg as compared to 0.03 U/mg in the wildtype. This increase was a combined effect of the limiting amounts of leucine added, together with an apparent additional internal leucine/valine shortage resulting from accumulated α-ketobutyric acid (5 mM) and the kinetic characteristics of the acetohydroxy acid synthase. The increase in the specific AHAS activity by the appropriate amino acid limitation resulted in an increased isoleucine yield of 71 mmol/l as compared to 27 mmol/l obtained under non-limiting conditions.

    Notizen: Abstract Pyruvate decarboxylase (EC 4.1.1.1) from the ethanol producing bacterium Zymomonas mobilis was purified to homogeneity. This enzyme is an acidic protein with an isoelectric point of 4.87 and has an apparent molecular weight of 200,000±10,000. The enzyme showed a single band in sodium dodecylsulfate gel electrophoresis with a molecular weight of 56,500±4,000 which indicated that the enzyme consists of four probably identical subunits. The dissociation of the cofactors Mg2+ and thiamine pyrophosphate at pH 8.9 resulted in a total loss of enzyme activity which could be restored to 99.5% at pH 6.0 in the presence of both cofactors. For the apoenzyme the apparent K m values for Mg2+ and thiamine pyrophosphate were determined to be 24 μM and 1.28 μM. The apparent K m value for the substrate pyruvate was 0.4 mM. Antiserum prepared against this purified pyruvate decarboxylase failed to crossreact with cell extracts of the reportedly pyruvate decarboxylase positive bacteria Sarcina ventriculi, Erwinia amylovora, or Gluconobacter oxydans, or with cell extracts of Saccharomyces cerevisiae.

    Notizen: Summary A new obligate methylotrophic bacterium capable of rapid growth on methanol as its sole carbon and energy source was isolated. The organism grows only on methanol and not on methane or methylamine. It is a gram-negative, motile rod (0.5×1.5 μm) with a single polar flagellum and was therefore classified as a species ofMethylomonas. The doubling time was about 2 hr at pH 7.0, at a temperature of 30 to 36°C and at a methanol concentration of 1% (v/v). Cell suspensions were able to oxidize methanol, formaldehyde, and formate; therefore it seems that methanol-, formaldehyde-, and formate-oxidizing enzymes are present in the bacterium. While no hydroxy pyruvate reductase activity was found, the cell extract contained high hexulose phosphate-synthetase activity, indicating the assimilation of methanol via the ribulose phosphate-pathway. The protein content of the bacterium is 60% and the amino acid pattern indicates that this strain could serve as a good source of single cell protein.

    Notizen: Abstract Recent microbiological findings show how compounds, regarded hitherto as unusual substrates for anaerobic bacteria, are degraded under anaerobic conditions. The complete conversion of halobenzoic acids and halophenolic compounds to methane by lake sediment and sewage sludge microorganisms has been demonstrated. Since haloaromatic compounds are widely used and may be found in such effluents as those from the forest industry, these studies could stimulate a broader interest in anaerobic treatment of industrial waste waters which contain unusual organic compounds.

    Notizen: T. viride ITCC 1433 synthesizes a two component system for the hydrolysis of cellobiose and cellooligodextrins. 80% of the total activity are solubilized during growth. The large protein (A), mol. weight 98 000 d, is glycosylated and slightly acidic (pH = 6.1). The smaller protein (B), mol. weight 70 000 d, is unglycosylated and neutral (pH = 7.2). Both proteins form a two-step system where β-glucosidase A is active at low substrate concentrations (KM = 2.3 × 10-4 M cellobiose) while β-glucosidase B covers the range of 10-fold higher cellobiose concentrations (KM = 1.8 × 10-3 M). The enzymes are fairly stable with a residual activity of 70% at 50°C after 24 h.

    Notizen: The specific growth rate of the ethanol producing bacterium Zymomonas mobilis was lower in the presence of oxygen than under anaerobic conditions. Aerobically, considerable amounts of acetaldehyde and acetic acid were formed in addition to the normal fermentation products, ethanol and carbon dioxide. This bacterium contains considerable amounts of pentacyclic triterpenoids, mainly 1, 2, 3, 4-tetrahydroxypentane-29-hopane. It seems that stability and permeability of the cytoplasmic membrane of this rather ethanol tolerant organism is achieved by the hopanoid content. A continuous culture of Zymomonas mobilis produced 60 g/l ethanol over a test period of 39 days. This strain was used for ethanol production from an enzymatically hydrolyzed wheat starch fraction on an industrial scale of 100 m3.
