    Notizen: Summary Thirty-eight biopsy specimens from 18 cases of bullous pemphigoid (BP) were observed using direct immunofluorescence (IF) techniques with fluorescein isothiocyanate (FITC)-labelled antisera against human serum factors. In addition to deposits of immunoglobulins and serum components at the basement-membrane zone (BMZ), 15 specimens from eight cases displayed homogeneous and globular fluorescent bodies in the uppermost dermis and/or the blisters when FITC-labelled antisera to human IgM and other serum factors were used. Using immunoperoxidase staining, haematoxylin/eosin (HE) and periodic acid-Schiff (PAS) staining, these immunoglobulin and/or complement-positive cell-sized bodies were shown to be slightly eosinophilic and PAS positive. Electron microscopy revealed entangled networks of microfilaments approximately 7–8 nm in diameter. These homogeneous, fibrillar bodies were histologically, immunohistologically and ultrastructurally indistinguishable from the colloid bodies found in lesional skins of lichen planus, lupus erythematosus, dermatomyositis and several other dermatoses. In BP, degenerated keratinocytes adjacent to the blister roof, may, after undergoing a filamentous change, drop off into the dermis and subsequently form homogeneous, fibrillar bodies in the uppermost dermis when reepithelization is completed.

    Notizen: Summary The developmental changes of enzymes involved in prostaglandin (PG) synthesis were investigated in rat skin from birth to 1.5 years old. In all stages of development, the activities of PG-synthesizing enzymes were found in 100,000xg supernatants of homogenates of rat skin, and PGD2 was the major PG among those formed from PGH2 in the presence of 1 mδ glutathione (GSH). The PGD synthetase activity in rat skin at birth was 2.14 nmol/min per mg protein, increasing to the highest level (3.69 nmol/min per mg protein) at 3 weeks after birth and then gradually decreasing up to 1.5 years old. The activities of PGE2 and PGF2α synthetases in rat skin were almost unchanged during development and aging. In contrast, the activity of GSH-S-transferase was at its lowest level at birth and gradually increased, reaching a plateau at 3 weeks after birth and remaining relatively constant during the development. The increase of PGD synthetase activity in 3-week-old rats was mainly due to the increase of specific activity of PGD synthetase in the epidermis, which was separated from the dermis by heat treatment (55° C, 30s). Immunohis-tochemical study, using (rat spleen PGD synthetase)-specific antibody, revealed that the number of immunopositive cells, which were identified as Langerhans cells, increased in the epidermis in 3-week-old rats. These results suggest that a change of PGD2 synthetase activity during aging of the skin is closely related to the development of ATPase+ Langerhans cells in the epidermis.

    Notizen: Summary The biochemical characteristics of cyclic AMP-dependent protein kinase in calf-snout epidermis were investigated. The activity of cyclic AMP-dependent protein kinase was higher in the lower layer than the upper layer of epidermis. The supernatant of homogenates of the lower layer of calf-snout epidermis was fractionated by DEAE-cellulose chromatography and contained two major peaks of protein kinase activity stimulated by cyclic AMP. This chromtographic pattern is similar to that referred to as Type I and Type II of cyclic AMP-dependent protein kinase in bovine muscle. Both peaks of cyclic AMP-dependent protein kinase in calf-snout epidermis could phosphorylate keratin polypeptides in vitro. The phosphorylation reaction was activated by cyclic AMP and inhibited by a heat-stable inhibitor of cyclic AMP-dependent protein kinase. When Type II enzyme of cyclic AMP-dependent protein kinase was incubated with [γ-32P]ATP in the absence of substrates, such as histone or keratin polypeptides, the 54,000 dalton protein was phosphorylated and this autophosphorylation was inhibited by the addition of 10 μM cyclic AMP. These results suggest that cyclic AMP-dependent protein kinase in calf-snout epidermis has properties similar to those in bovine muscle and plays an important role in the phosphorylation of keratin polypeptides in calf-snout epidermis.

    Notizen: Summary The skin response to UVB irradiation in mice was evaluated by means of ear swelling. ICR albino mice were irradiated with 500 mJ/cm2 UVB and the effect of various drugs on ear swelling was examined 24 h after irradiation. Intravenous injections of betamethasone (0.8 μg/g body wt.) or indomethacin (24 μg/g) remarkably inhibited ear swelling, whereas intraperitoneal injections of diphenhydramine (20 μg/g), cimetidine (10 μg/g), or a combination of both these antihistamines did not. In contrast, the number of sunburn cells counted 24 h after UVB irradiation (200 mJ/cm2) in mouse ears was not affected by these drugs. The amount of prostaglandin D2 (PGD2) in mice ears at various intervals after irradiation with 500 mJ/cm2 UVB was determined by radioimmunoassay. Compared with the values before irradiation, the PGD2 levels were significantly higher 3 and 6 h after irradiation and gradually decreased and returned to the basal level by 12 h, although ear swelling continued after 12 h. These results suggest that prostaglandins, are responsible at least in part for the development of ear swelling, but not for sunburn cell formation induced by UVB, and that ear swelling represents a simple and useful response model for the rapid in vivo screening of nonsteroidal or steroidal anti-inflammatory agents.
