    Notes: Human natural killer (NK) cells were tested in the presence of several fatty acid oxygenase inhibitors such as nordihydroguaiaretic acid (NDGA), 5,8,11,14-eicosatetraynoic acid, 3-amino-l-m-(trinuoromethyl)-phenyl-2-pyrazoline (BW 755C). and indomcthacin. All drugs inhibited NK lysis at the post-target cell-binding level at concentrations that also suppressed lipoxygenation of arachidonic acid, suggesting that such reactivity may be required for effector cell triggering. NDGA gave a 50% NK cell inhibition in the range of 10-30 μM and also suppressed antibody-dependent and lectin-dependenl systems. Further evidence of the involvement of arachidonic acid lipoxygenation was found as NK activity could be reconstituled to NDGA-suppressed effector cells with several metabolites such as LTB4. LTB4 analogues, and hydroxyeicosatetraenoic acids lipoxygenated at C-5, C-12, and C-15. Cyclic nucleotides such as cGMP and cAMP could also reconstitute activity with optimal effects at approximately 10−8 M. The combined evidence is compatible with a model for triggering lysis in which lipoxygenation products have a second messenger function. Whether arachidonic acid lipoxygenation is necessary for effector cells at all different activation/differentiation stages and whether the lipoxygenation products activate guanylic cyclase, protein kinase C, or some other target molecule remain to he further investigated.

    Notes: Summary The effects of the antiestrogens tamoxifen and keoxifene on the bone density of intact and ovariectomized female rats were determined after 4 months of therapy. The antiestrogens did not cause a decrease in bone density in intact animals, although uterine wet weight did decrease. Ovariectomy caused an increase in body weight (25%) and a significant decrease in femur density (P〈0.01). Antiestrogens did not further decrease the bone density of ovariectomized rats but rather helped to maintain bone density. Antiestrogens as well as estrogen (oral estradiol benzoate 25µg daily) helped to maintain bone density in the range observed for the intact rats, but inhibited estrogen stimulation of uterine weight. These contrasting pharmacological actions of antiestrogens suggest that patients receiving long-term adjuvant tamoxifen therapy for breast cancer should be evaluated to determine whether tamoxifen can retard the development of osteoporosis.

    Notes: Abstract The Lithium Blanket Module (LBM) is an approximately 80×80×80 cm cubic module, representative of a helium-cooled lithium oxide fusion reactor blanket module, that will be installed on the TFTR (Tokamak Fusion Test Reactor) in late 1986. The principal objective of the LBM Program is to perform a series of neutron transport and tritium-breeding measurements throughout the LBM when it is exposed to the TFTR toroidal fusion neutron source, and to compare these data with the predictions of Monte Carlo (MCNP) neutronics codes. The LBM consists of 920 2.5-cm diameter breeder rods constructed of lithium oxide (Li2O) pellets housed in thin-walled stainless steel tubes. Procedures for mass-producing 25,000 Li2O pellets with satisfactory reproducibility were developed using purified Li2O powder, and fabrication of all the breeder rods was completed in early 1985. Tritium assay methods were investigated experimentally using both small lithium metal samples and LBM-type pellets. This work demonstrated that the thermal extraction method will be satisfactory for accurate evaluation of the minute concentrations of tritium expected in the LBM pellets (0.1–1 nCi/g).

    Notes: The tissue reaction to implanted materials and their wear products is important for the long-term fixation of a total hip replacement (THR). Recent reports indicate a relatively high incidence of aseptic loosening with Christiansen total hip prostheses. We examined tissue reactions to wear products in two groups of prostheses: (1) 16 Christiansen prostheses with polyacetal (Delrin) sockets, and (2) 18 Charnley-Müller total hip prostheses with ultrahigh-molecular-weight polyethylene (UHMWP) sockets. Specimens from these 34 patients were compared in a combined qualitative and semiquantitative morphological study. We saw similar histological changes in specimens of both groups, but specimens from the bone-cement interfaces of the acetabulum and femur shaft in the polyacetal group showed more inflammation and necrosis.

    Notes: Guinea pigs sensitized to nickel, cobalt, or chromium were used to study if impaired fixation occurred as a result of allergy against metal implants in bone. Screws made from stainless-steel (ASTM F55, F138-139) or cobalt chromium (ASTM F75-76) were inserted in the right and left proximal tibiae, and the mechanical strength of the fixation to the bone was evaluated after 4 months. In addition, we studied the histology on decalcified sections of the implant bone interface from the distal femur where the same type of screws had been inserted. To see if changes in bone density occurred in the proximal tibia as a result of allergy, the amount of ash was determined. Although the animals maintained their contact sensitivity throughout the experimental period, there were no differences between allergic and control animals for any of the parameters studied. All screws were well fixed in the tibia at the end of the experiment, and histologically we did not find any significant differences between allergic and control animals by qualitative examination. The experiment supports our earlier results from a 6-week experiment and indicates that contact allergy may be unimportant for the fate of orthopedic implants.
