    Notes: Circular dichroism (CD) was used to study changes in conformation of sardine (Sardinops melanosticta) actomyosin after treatment with some organic solvents. Untreated sardine actomyosin showed two negative bands at 208 and 222 nm, typical of proteins possessing α-helix configurations. The water-miscible alcohols methanol, ethanol, isopropanol and n-butanol, and the water-immiscible solvents n-hexane, n-octanol, and dichloromethane altered the native conformation of sardine actomyosin. The degree of alteration of native sardine actomyosin was principally dependent on the alcohol concentration and temperature used for dewatering. The least damage occurred with isopropanol, or with 10 or 90% aqueous ethanol; most damage occurred with 40–50% aqueous ethanol. Dichloromethane and n-butanol increased the α-helical content of the native protein. Treatment with hydrophilic and hydrophobic solvents at 70 and 20°C unfolded the protein to form a random coil, but defatting treatment at 2°C caused little damage.

    Notes: Abstract Antigens, recognized by human monoclonal antibody (HB4C5) generated from a lung cancer patient, were found to occur in porcine pancreas. The antigens-I and -I1 were purified from crude trypsin of porcine pancreas, only by Mono Q column chromatography, and were eluted at 260 and 300 mM NaCl in 10 mM Tris-HCI buffer, pH 7.4, respectively. These antigens differed from trypsin in molecular weight, elution pattern from the Mono Q column, and their reactivity with HB4C5. The molecular weights of the two antigens were almost the same at around 35000. These were used for serodiagnosis with an assay system based on 96-well immunoplates. The reactivities of antigens-I and -II with various sera were similar. When the reactivity of IgG in serum with antigen-II was measured, absorbance at 415 nm in the case of normal and lung cancer patients was 0.178 ± 0.056 and 0.492 ± 0.136 (p 〈 0.005). The rates of positive reaction in ovary, larynx, uterus, lung and liver cancers were more than 50%, but the rates in stomach and breast cancers were less than 30%. Positive reaction was hardly detected in pancreas cancer and normal controls.

    Notes: Summary Four patients with common acute lymphoblastic leukemia antigen (CALLA)-positive myeloma are presented. The subclasses of monoclonal protein were IgDκ (1 case), IgAλ (1 case), and IgAκ (2 cases). Bence Jones proteinuria was seen in all cases. The clinical stages were determined as IIA (2 cases) and IIIA (2 cases). All patients died with a median survival time after diagnosis of 62 days due to rapid development of renal failure (3 cases), and renal insufficiency and pneumonia (1 case). According to light microscopic evaluation, these myelomas corresponded to plasmablastic (1 case), immature (2 cases), and intermediate (1 case) types. Both CALLA and a cytoplasmic immunoglobulin identical with the serum monoclonal protein were simultaneously detected in single cells from all cases using immunofluorescent double labeling. These findings suggest that CALLA-positive and plasmablastic myelomas constitute clinically a subgroup characterized by extremely poor survival but they represent cytologically different subcategories.

    Notes: Abstract Human-human hybridoma SH-76 cells were found to produce a factor that supported the growth of lymphocytic cells at low densities. The factor was purified from serum-free conditioned medium of the hybridoma cells by a successive application of ammonium sulfate precipitation, DEAE-Toyopearl, TSK G3000 SW and DEAE-5PW column chromatograph. The purified factor was a 72K single protein. The factor showed marked growth stimulating effect on lymphocytic cell lines, but had no effect on the growth of human adhesive cancer cell lines. Thus, the factor is a lymphocytic clonal growth factor (LCGF), as found previously in human plasma (Miyata, 1988). The LCGF of SH-76 cells could be produced in growth factor-free RPMI medium and purified easily from the conditioned medium. The factor is inactivated by heating at over 80°C, but is much more stable than the LCGF in human plasma.

    Notes: Abstract Potassium or sodium phosphate was found to stimulate the production of human monoclonal antibody by human-human hybridoma HB4C5. The addition of 15 mM Na-phosphate (pH 7.4) into serum-free culture medium increased the antibody production up to 4-fold, when seeded at cell density of 1×105 cells/ml in dishes. At the higher cell density of 5×105 cells/ml, K-phosphate was more effective than Na-phosphate, at the same concentration. In large-scale continuous culture, the addition of 10 mM Na-phosphate into serum-free culture medium stimulated antibody production by HB4C5 cells 6-fold.

    Notes: Abstract Human plasma was found to contain a macromolecular protein which can grow even a single cell of human lymphocytic cell lines (B-lymphoblastoid cell line HO-323-3 and T-lymphoblastoid cell line CCRF-CEM) and human-human hybridoma clones (SH-9, SU-1 and HB4C5) in a dish, but it has no effect on the growth of epithelial cell lines (lung cancer cell lines PC-8, QG-56 and QG-90). The proliferating activity for lymphocytic cell lines was gradually decreased at 4 or -20°C and dramatically decreased by heating at more than 60°C for 15 min. From human plasma, active fractions were purified by a successive application of Ca2+ treatment, ammonium sulfate fractionation, DEAE-5PW column chromatography (FPLC) at pH 7.6. These active fractions were divided into at least three proteins by DEAE-5PW chromatography at pH 8.5 and chromatofocusing. These purified factors, named lymphocytic clonal growth factors (LCGFs), had similar molecular weights of about 600 K and each factor consisted of a 180 K and two 210 K subunits associated with hydrogen bondings. By the addition of 5 μg/ml of each factor into culture media, incidences of human-human hybridomas and cloning efficiencies of the hybridomas increased several-fold.

    Notes: Summary The purpose of the present study was to contrive a new practical method for estimating total O2 uptake during exercise from total heart beats after individual evaluation of aerobic fitness levels. Twenty healthy male subjects participated in cycle ergometer tests, maximal O2 uptake ( $$\dot V_{{\text{O}}_{{\text{2 max}}} } $$ ) tests and various simple tests including simple endurance tests. From the cycle ergometer results, the following formula for estimating total O2 uptake in exercise was determined: $${\text{TV}}_{{\text{O}}_{\text{2}} } $$ (ml·kg−1)=SR125×(45.8× mean HR+4268)×THB×10−4 where $${\text{TV}}_{{\text{O}}_{\text{2}} } $$ , THB, and mean HR are total O2 uptake, total heart beats, and mean heart rate (beats · min−1) in exercise, respectively, and SR125 is the slope of the regression line between accumulated heart beats and accumulated O2 uptake during exercise at 125 beats · min−1 of mean HR. SR125 had a significant correlation not only with $$\dot V_{{\text{O}}_{{\text{2 max}}} } $$ but also with each score (X) in any simple endurance tests such as, for example, a step test for 2 min. In this case, accordingly, SR125 can be found as; SR125=−0.00118X+0.3478. These formulae indicate that the total O2 uptake of any exercising subject can be estimated from his total heart beats regardless of intensities of exercise when his aerobic fitness level is evaluated by the simple endurance test. The total O2 uptake estimated by our method was compared to that measured by the Douglas bag method, and the discrepancy between the two results was less than the errors of values estimated by traditional methods.

    Notes: Summary In order to estimate muscle endurance during knee extension (KME) and plantar flexion (PME) using a simple method, holding times for the half-squatting posture for KME and for the calf-raising posture for PME were measured in 3,792 males and 3,620 females aged between 6 and 79 years. The regression lines of the aging process for KME and PME in both sexes were determined. Peak values were found at 13 years in males and 12 years in females for KME, and at 15 years in males and 16 years in females for PME. The mean values increased sharply with age up to each peak and then decreased abruptly to a low level at around 20 years, thereafter declining gradually. The values of KME as a percentage of the peak value were 71, 75 and 34 at the ages of 6, 20 and 70–79 years respectively in males, while the corresponding values were 80, 77 and 36 for females. The values of PME at these ages were 34, 93 and 69 in males and 54, 91 and 73 in females. The mean values for males were generally higher than those for females. These results suggest that endurance of the leg muscles differs according to age, sex and the region of the leg muscle. These data will be useful as standard values for evaluating endurance and for prescribing suitable exercise for the promotion of physical fitness.

    Notes: A new finite element for two-layer plates with built-in interlayer slip is developed. The new plate element is based on a new plate theory formulated using a new variational principle due to Reissner. The accuracy of the new element is investigated by applying it to the problem of a two-layer plate with a linear slip law in cylindrical bending for which an exact elasticity solution exists. The comparison of the inplane response for several different values of interlayer shear stiffness and layer thickness ratios shows that the new element gives accurate results. The cylindrical bending of a two-layer plate with bilinear interlayer slip law is investigated with the new element and the results obtained are compared with a plane strain analysis. Good agreement is obtained for in-plane normal stresses and displacements. To further assess the simulation capabilities of the proposed element, a rectangular plate under concentrated load is analysed and the growth of slip failure regions is shown.
