    Notes: Summary Glutathione-insulin transhydrogenase catalyzes the inactivation of insulin by splitting the hormone into A and B chains. We have localized this enzyme immunocytochemically by light microscopy in the pancreas, kidney and liver of both lean and obese (ob/ob) mice and similarly in normal and streptozotocin-diabetic rats. Localization was achieved by an antibody to glutathione-insulin transhydrogenase using a peroxidase-antiperoxidase technique. In comparison with tissues from control animals, positive immunostaining for glutathione-insulin transhydrogenase was increased in the obese mouse but reduced in the diabetic rat. Different tissues showed considerable variation in the amount of glutathioneinsulin transhydrogenase which could be detected. In the pancreatic islets there was little or no evidence for the presence of the enzyme in peripheral cells. In the kidney, immunocytochemical staining was found only in the proximal tubules. In the liver there was a generalised distribution of the enzyme, but the greatest concentration was in the periportal region. These observations parallel the biochemical data relating to glutathione-insulin transhydrogenase, indicating that different amounts of insulin degrading activity exist in different regions of tissues.

These include: (1) small mouth (intercommissural distance 〈 3rd percentile); (2) tented upper lip; (3) small nose (interalar distance –2 SD below mean); (4) fullness of cheeks giving a ’jowly' appearance; (5) overhanging outer thirds of upper eyelids. These features may reflect prolonged tissue oedema and protein malnutrition in utero and early life. Conclusion Characteristic facial features in congenital nephrotic syndrome may reflect intra-uterine malnutrition.

    Notes: Objective To study the pathological features of fetuses dying because of parvovirus B19 infection, with particular reference to the presence of hydrops; to assess the usefulness of immunochemistry as a screening method for the detection of parvovirus infection at post-mortem examination.Design Review of clinical, sonographic, serological and pathological data; immunohistochemical staining of post-mortem tissue.Sample Cases of intrauterine fetal death occurring during the 18-month period January 1993 to June 1994 inclusive, referred for post-mortem examination to the Pathology Department, Royal Victoria Infirmary, Newcastle upon Tyne.Results Eleven cases of fetal death due to parvovirus infection were identified. Seven fetuses were less than 18-week size. Three fetuses showed conspicuous hydropic change. One of the 11 cases was detected for the first time by retrospective immunochemical screening. Of cases originating from the Newcastle district, parvovirus infection was responsible for about 10YO of all non-malformed fetal deaths occurring between 10 and 24 weeks of gestation referred for pathological examination.Conclusions During the period of study, parvovirus infection was a relatively common cause of mid-trimester fetal death. Many fetuses dying because of this infection are not noticeably hydropic, and the possibility of parvovirus infection should be considered in any case of intrauterine fetal death. Immunochemistry can be used to confirm the histopathological diagnosis, and may be of particular help where there is advanced autolysis; immunohistochemical screening may detect occasional cases not initially identified by examination of routinely stained tissue sections.

    Notes: Methods used to track the free surface in numerical flow simulations, typically during the casting of metals into moulds are examined in this paper. The algorithm employed makes use of a mixed interpolation formulation to approximate the discretised governing equations for elimination on a Langrangian type moving mesh. Significant savings in CPU time are realised by virtue of the the air domain not being considered in the finite element analysis. The advantages and drawbacks of commonly used methods are discussed, and a method proposed for the numerical simulation of flow where the free surface boundary conditions are important. The effectiveness of the proposed algorithm in solving typical industrial flow problems is demonstrated using numerical examples. The results obtained are compared with analytical and numerical data with a view to validating the algorithm.

    Notes: Immunoperoxidase techniques provide the pathologist with the capability for staining a wide range of antigens in tissue sections. More than 100 different antigens have been successfully demonstrated in fixed paraffin sections; other antigens can only lie visualized in frozen sections. This latter group particularly includes lymphocyte surface antigens detectable by monoclonal antibodies. This review describes the current state of the art and provides several illustrations of the use of monoclonal antibodies for the identification of T-lymphocyte phenotypes in fro/en section from cases of leprosy, mycosis fungoides, halo nevus, Kaposi's sarcoma, lichen planus and atopic dermatitis. Technical details and potential applications arc discussed. The growing availability of commercial immunostaining kits makes these techniques more accessible to the surgical pathologist; indeed a whole new range of truly specific, special stains are available, as pathologists we must simply learn to use them.
