    Notes: Summary Extracts from homogenates of rat and human parotid glands contained insulin-like immunoreactivity. The values were 5.6±2.8 ng/g wet tissue in six groups of rat parotid glands and 23.8 and 39.7 ng/g wet tissue in two human extracts. Upon gel filtration immunoreactive insulin of rat origin was eluted in a peak corresponding to the elution volume of isotopically labelled insulin. The material obtained from the two peak fractions showed an immunoassay dilution curve identical with that of rat insulin. Furthermore, biosynthesis of insulin-like immunoreactivity in rat and human parotid glands was confirmed in vitro by a specific separation method using anti-insulin antibody. These findings suggest that the parotid gland may be a further extrapancreatic source of insulin, and that insulin biosynthesis does occur in extrapancreatic tissues.

    Notes: Abstract A new association scheme which can still recall appropriate data when some key elements are missing (blank) is presented. The traditional associative memory models are designed to deal with complete (memorized) keys, but in the real world, key elements are often missing due to error, equipment failure, observation difficulty, etc. The traditional models, in this case, can not have an optimal association except for special cases. When an incomplete key containing blanks is given, we wish to get the same data, as nearly as possible, as would be obtained with the complete key. In this paper, the optimal associative memory model which operates with partly missing keys is proposed. The model is constructed on the basis of the theory of the pseudoinverse of matrices. Even from the incomplete keys which contain a large percentage of blanks, the model recalls the appropriate data optimally under the MSE criterion. From the results of computer simulations, we can show that the model has the expected ability.

    Notes: Abstract A recombinant CHO cell line producing human prorenin was cultivated on microcarriers in serum-free medium. Best growth was obtained when the cells were cultivated on a collagen coated microcarrier (Cytodex 3) using a serum-free medium (SF-02) supplemented with fibronectin. It was possible to reduce the necessary concentration of fibronectin in the feed medium from 10 μg/cm3 to 2 μg/cm3 during perfusion cultures in a spinner reactor equipped with an UF-membrane. Also in this system, the prorenin concentration increased up to 8 times higher compared to that in a conventional repeated-batch culture. The cells grew in multilayers on the microcarriers during the perfusion culture. The specific prorenin productivity was not significantly affected by the cell growth rate, and the secretion of prorenin continued even after the cells had ceased to grow.

    Notes: The presence of neutral and alkaline proteinases in the extract from Antarctic krill, Euphausia superba was confirmed. In addition to these enzymes, acid proteinases were found in the extract from E. superba and the two major acid proteinases, A and B, were purified 80–140 fold. The molecular weights of acid proteinases A and B were estimated by gel filtration on Sephadex G-100 to be approximately 45,000 and 64,000, respectively. The optimal pH of enzymes A and B was 3.0 toward hemoglobin. They were partially inhibited by acid proteinase specific reagents: diazoacethyl-DL-norleucine methylester (DAN) and 1,2epoxy-3-(p-nitrophenoxy) propane (EPNP). However, enzyme B was less sensitive to pepstatin while enzyme A was inhibited by pepstatin like almost all other acid proteinases.

    Notes: Abstract: The polypeptide elongation factors (EF-1L, EF-1H, and EF-2) of the developing chick brain were separated and purified by means of a combination of gel chromatographic methods. The molecular weight of EF-1H of the chick brain ranged from 5 to 10 × 105, and was different from that of the chick liver (about 7 × 105). The molecular weight of other purified factors was about 5 × 104 for EF-1L. and 9.4 × 104 for EF-2. High activities of polyphenylalanine (poly-Phe) synthesis per mg protein in the developing chick brain were observed between the 3rd embryonic week and the 1st post-hatch week and declined afterwards. On the other hand, the levels of both EF-1 and EF-2 per mg protein in the brain were observed to be high in an early embryonic stage, gradually declining afterwards to the adult level. The brain EF-1L was a major component of EF-1 in an early embryonic stage, while EF-1H became recognizable in the 3rd embryonic week. Moreover, the EF-1H activities were found to be more than double with regard to the binding reaction and to be more than 10-fold as active in respect to poly-Phe synthesis in comparison with the activities of EF-1L. It is proposed that the brain EF-1H could be due to aggregates consisting of EF-1L, a stimulatory factor, and other components.

    Notes: Abstract: The higher-molecular-weight elongation factor-1 (EF-1H) of the chick brain was observed to contain three subunits (denominated α, β, and γ), contrary to a previous report that the brain EF-1H consisted of aggregates of low-molecular-weight elongation factor- 1 (EF-1L). Crude EF-1H, obtained from 20-day embryonic brain, was treated with 0.4 M ammonium chloride and 0.1 mM GTP, and EF-1βγ, was obtained using a DEAE-Sephadex column equilibrated in 0.025 mM GTP. Both EF-1β, and EF-1γ, were isolated by means of a DE-52 column equilibrated in 6 M urea and were found to have molecular weights of 2.8 and 4.8 × 104, respectively. EF-1β and EF-1γ were also obtained from young rat and calf brains by the same procedures. The molecular weight of the isolated EF-1α was 5 × 104. It was found that EF-1β stimulated the two EF-1α-dependent reactions, i.e., phenylalanyl-tRNA binding (reaction 1) and polyphenylalanine synthesis (reaction 2), and also stimulated the nucleotide exchange reaction in the EF- 1α-guanine nucleotide binary complex (reaction 3). The degrees of stimulation of reactions 1, 2, and 3 by the addition of EF-1β were 2 to 3 times, about 18 times, and 2 to 3 times as much as with EF-1α alone, respectively. The amino acid compositions of EF-1α -1β, and -1γ and EF-2 were very similar to those of other eukaryotic tissues. Thus the constituents and properties of EFs of the brain were found to be basically similar to those of other tissues of eukaryotes, although EF-1β, and EF-1, had not been reported in the brain. A possible physiological significance of EF-1β during brain development is also discussed.

    Notes: Total discectomy was performed experimentally in the rabbit temporomandibular joint (TMJ). In order to evaluate the injury and repair of the articular cartilage of the mandibular condyle after discectomy, both histologic and immunohistochemical observations were made. Immunohistochemical observation by using bromodeoxyuridine (BrdU) and its monoclonal antibody in vivo labelling method was used.The present study demonstrated that the cartilage on the articular surface of the condyle disappeared at 1 week post-operatively. This defect in the injured articular cartilage was not repaired by the cartilage, but was covered by a fibrous connective tissue at 6 weeks post-operatively, where the condylar recontouring by the adjacent articular cartilage, and flattening and hyperplasia of the condyle were observed. Thus, the cartilaginous repair following discectomy was characterized by the proliferation of the adjacent cartilage to the injured site and osteoblastic formation in the bone marrow at the exposed subchondral bone.

    Notes: The ion beams are produced by a cylindrical thermal contact-ionization plasma source with an electrostatic lens system. The source plasma is produced at the inside surface of a 5-mm-diam 20-mm-long Re foil cylinder protected with a W cylinder. The surface is heated up to 3000 K by electron bombardment. The material to be ionized is fed to the surface through a guide pipe from a reservoir. Generation of all alkaline metal ions have been tested and a beam current of hundreds of μA is obtained. The efficiency for Li+ production is 30%, while that for Cs+ is nearly 100%. Alkaline earth ion beams, except Mg, are generated. A current of Ba+ up to 200 μA is obtained, while a Tl+ beam of over 200 μA is achieved. Halogen negative ion beams, except F−, of the order of 10 μA are also obtained when alkali halides are used. Attempts to extract Li− from the source are also being made.

    Notes: Both renin and cathepsin B were co-localized in identical granules of adrenocortical cells. At day 16 of gestation, many renin-containing granules were observed and gold particles showed homogeneous intragranular distribution; whereas, those for cathepsin B was distributed heterogene-ously. At day 18 of gestation, renin immunoreactivity was decreased or undetectable, whereas cathepsin B was still demonstrated at the same level as on day 16 of gestation.
