    Notizen: Abstract: The distribution of choline kinase (EC 2.7.1.32) activity was investigated in subcellular fractions of rat striatum. Enzyme activity in the crude mitochondrial fraction, determined after dissolution in Triton X-100, was 5.90 μmol/g initial wet weight/h. When a crude mitochondrial preparation was hypoosmotically shocked and fractionated, followed by the addition of Triton X-100, choline kinase activity in the soluble and particulate fractions was 4.58 and 1.40 μmol/g initial wet weight/h, respectively. Enzyme activity in the paniculate fraction was not detected in the absence of Triton X-100 or in the presence of NaCl (up to 1.5 M). Subcellular enzyme markers indicated that the membrane-associated activity was not attributable to mitochondrial or microsomal contamination. Kinetic analysis of the activity of soluble and membrane-solubilized choline kinase indicated Km values of 0.74 mM and 0.68 mM, respectively. Results indicate that choline kinase activity may be measured in both the soluble and the paniculate fractions of rat striatum, the latter most likely involving enzyme associated with membrane through hydrophobic or covalent interactions. The specific function of the membrane-associated enzyme has not yet been determined.

    Notizen: Psychological screening has become commonplace in a number of occupational groups. With the growing popularity of screening and the relative scarcity of appropriate personality measures, re-screening individuals with the same inventory is frequent. This paper examines the effect of re-screening with the Minnesota Multiphasic Personality Inventory (MMPI) on contract workers (n = 1,997) from the nuclear power industry. Participants completed the Inventory as a part of a routine screening procedure. Each participant provided several profiles as a result of regular employment cycles. Although the differences are generally small, some scales show large enough mean changes to warrant concern when interpreting the profiles of those who have taken the profile multiple times. Although scale reliabilities tend to increase, the usefulness of the Inventory appears to decrease with an increasing number of administrations. The effect is one of increasing normality of test profiles. One interpretation of the results is that candidates for jobs become “test-wise,” and the scores are the result of a combination of responses to the repeated assessment. It becomes increasingly difficult to identify unfit employees with the Inventory. Given these results, further research on retesting effects of the Inventov, as well as other personality measures, is indicated.

    Notizen: Abstract Intracellular element concentrations were measured in rat sympathetic neurones using energy dispersive electron microprobe analysis. The resting intracellular concentrations of sodium potassium and chloride measured in ganglia maintained for about 90 min in vitro at 25° C were 3, 155 and 25 mmol/kg total tissue wet weight respectively. Recalculated in mmol/l cell water, these values are 5, 196 and 32 respectively. There were no significant differences between the nuclear and cytoplasmic values of these ions. Incubation in either carbachol (108 μmol/l, 4 min) or ouabain (1 mmol/l, 60 min) significantly increased the intracellular sodium and decreased the intracellular potassium concentrations. Neither substance materially altered the intracellular chloride concentration. The data obtained are compared and contrasted to those obtained in mammalian sympathetic neurones using chemical analysis and ion-sensitive microelectrodes.

    Notizen: Abstract Intracellular ion concentrations were determined in split skins of Rana pipiens using the technique of electron microprobe analysis. Based on the 1 min Br uptake from the apical bath, two types of mitochondria-rich (MR) cells could be distinguished: active cells which rapidly exchanged their anions with the apical bath and inactive cells which did not. Br uptake and frequency of active MR cells were closely correlated with the skin conductance, g t. Replacing Cl in the apical bath with an impermeant anion significantly lowered g t and the Br uptake and Na concentration of active cells. Even larger reductions were observed after apical amiloride (0.1 mm). The inhibition of the Br uptake was reversible by voltage clamping (100 mV, inside positive). Cl removal and amiloride also led to some shrinkage of active cells. The results suggest that the active cell is responsible for a large part of g t. Inactive MR cells had much lower Br and Na concentrations which were not significantly affected by Cl removal, amiloride, or voltage clamping. Principal cells, which represent the main cell type of the epithelium, showed only a minimal Br uptake from the apical side which was not correlated with g t. Moreover, Cl removal had no effect on the Na, Br, and Cl concentrations of principal cells.

    Notizen: Abstract Two new activators of (1,3)-β-glucan synthase fromSaccharomyces cerevisiae were identified, and a procedure for preparing enriched enzyme fractions by removal of peripheral membrane proteins and entrapped soluble proteins was developed. Microsomal enzyme activity, known to be enhanced by bovine serum albumin (BSA), was stimulated threefold by both β-lactoglobulin and Brij-35. Both apparently substituted for BSA, since no synergistic effects were observed with activators added in combination. Successive washings of microsomal fractions with the detergents Brij-35 and Tergitol NP-40 to remove peripheral and vesicle-entrapped proteins yielded particulate fractions five-fold enriched in glucan synthase activity. GTP, an important effector of glucan synthase, improved purification of the enzyme during detergent extractions. Various membrane fractions were photolabeled with 5[32P]N3UDP-Glc or 8N3[32P]GTP, and potential UDP-Glc and GTP-binding polypeptides were identified. However, further enrichment will be required to determine which of these might represent subunits of the yeast glucan synthase complex.
