Geburtstag gewidmet.The synthesis of various dinucleoside monophosphates (6a-e) with thymidine (3a) and 1-(2′-deoxy-β-D-ribofuranosyl)lumazines (3b-d) as building blocks via the phosphotriester approach is described. The completely protected phosphotriesters (4a -e) as well as the partially deblocked intermediates (5a-e) have been isolated and characterized by physical methods. Intramolecular interactions in the free dinucleoside monophosphates 6a-e have been detected by the determination of the hypochromicities and measurement of CD spectra, revealing a small base-stacking effect. Enzymatic hydrolysis of the dinucleoside monophosphates with snake venom and spleen phospodiesterase strongly depends on various structural features and is greatly retarded in presence of the 6,7-diphenyllumazine moiety as an aglycone.
    Notes: Die Synthese verschiedener Dinucleosidmonophosphate (6a-e) mit Thymidin (3a) und 1-(2′-Desoxy-β-D-ribofuranosyl)lumazinen (3b-d) als Bausteine wird mit Hilfe der Phosphotriester-Methode realisiert. Die voll- (4a - e) und partiell-geschützten Phosphotriester (5a - e) werden isoliert und durch physikalische Daten charakterisiert. Intramolekulare Wechselwirkungen in den freien Dinucleosidmonophosphaten 6a-e werden durch Bestimmung der Hypochromizitäten und Aufnahme von CD-Spektren nachgewiesen und deuten auf eine schwache Basen-Stapelung hin. Enzymatische Hydrolysen dieser Dinucleosidmonophosphate mit Milz- und Schlangengift-Phosphodiesterase zeigen eine starke Strukturabhängigkeit und sind bei Gegenwart des 6,7-Diphenyllumazins als Aglykon stark verlangsamt.

Geburtstag gewidmet.A series of new trinucleotide diphosphates (31 - 36, 45 -48) containing lumazine or its 6,7-dimethyl- or 6,7-diphenyl derivative, respectively, as aglycones at the 3′-end have been synthesized by the phosphotriester method. The various intermediates in protected (19 - 24, 37 - 40) and partially deblocked forms (25 - 30, 41 - 44) have been isolated, purified by chromatography and characterized by spectroscopic methods. Intramolecular interactions in the free trinucleoside diphosphates 31-35 and 45 - 48 have been studied by determination of hypochromicities and measurement of CD spectra. Enzymatic cleavages by phosphodiesterases revealed a structure-dependent decrease of the rate of hydrolysis of the internucleotidic linkage on degradation from the 3′-end using snake venom phosphodiesterase. The experimental findings are discussed.
    Notes: Nach der Phosphotriester-Methode werden eine Reihe neuer Trinucleosiddiphosphate (31-36, 45-48) synthetisiert, die alle am 3′-Ende als Aglykon Lumazin bzw. sein 6,7-Dimethyl- oder 6,7-Diphenyl-Derivat tragen. Die verschiedenen Zwischenprodukte in geschützter (19 -24, 37-40) und partiell deblockierter Form (25-30, 41-44) werden isoliert, chromatographisch gereinigt und durch spektroskopische Methoden charakterisiert. In den freien Trinucleosiddiphosphaten 31-36 und 45-48 werden die intramolekularen Wechselwirkungen durch Bestimmung der Hypochromizitäten und Aufnahme der CD-Spektren untersucht. Enzymatische Spaltungen mit Phosphodiesterasen weisen eine strukturbedingte, verlangsamte Hydrolyse der Internucleotidbindung beim Abbau vom 3′-Ende her mit Schlangengift-Phosphodiesterase auf. Die experimentellen Befunde werden diskutiert.
