    Notes: Abstract In the field successive leaves of winter wheat appear at a rate which varies because it depends strongly upon temperature. When plotted against ‘thermal time’, however (temperature accumulated above a fixed base of 0°C), leaf appearance was a strictly linear function of temperature. The mean rate of leaf appearance in thermal time, R′, was faster for a spring sowing than for an autumn sowing. The variation in R′ between sowings was better correlated with the rate at which daylength was changing when the plants emerged than with the mean daylength while leaves were appearing.

    Notes: The effect of tartrazine (T) (yellow dye No. 5) and one of its metabolites an aminopyrazolone analogue (1-sulphophenyl-3-carboxy-5-hydroxypyruzole, SCHP) upon collagen-induced platelet aggregation (C-PA) was investigated in fourteen atopic patients and fourteen normal subjects. Both T and SCHP inhibited C-PA in atopic patients at significantly lower doses than in normal volunteers. The mean inhibitory concentrations of SCHP were similar to aspirin in both atopic and normal individuals. Although the precise mechanism by which these chemicals block C-PA has not been elucidated, this in vitro system may be a useful method of assessing non-immune mechanisms involved in reactions to tartrazine.

    Notes: Summary The dependence of leaf water potential (Ψ), osmotic potential (π) and turgor pressure (P) on relative water content (RWC) was determined for leaves of tall and short growth forms of Spartina alterniflora Loisel. from a site on Canary Creek marsh in Lewes, Delaware. Tall plants (ca. 1.5 m) occured along a drainage ditch where interstitial water salinity was approximately 20‰, and short plants (ca. 0.2 m) were 13 m away near a pan and exposed to 80‰ salinity during the most stressful period. Leaves were collected at dawn and pressure-volume measurements were made as they desiccated in the laboratory. Pressure equilibrium was used to measure Ψ, RWC was determined from weight loss and dry weight, π was determined from the pressure volume curve, and P was calculated as the difference between Ψ and π. Physical properties of the bulk leaf tissue that have a role in regulating water balance of the two growth forms were estimated: relative water content of apoplastic water (RWCa) relative water content at zero turgor (RWC0), the bulk modulus of elasticity (E), and water capacity (C w). There were no detectable temporal trends in any of the parameters measured from Nune through September and no significant differences between the two growth forms when compared on the basis of RWCa, RWC0, E, and C w. There was a clear difference between the two growth forms with respect to π; at RWC0, π was-4.5±0.40 MPa for short form plants and-3.3±0.40 MPa for tall form. Turgor pressure of plants in the field (P′) was lower in leaves from short form than for the tall form plants with average difference of about 0.4 MPa. In July, P′ in short form leaves dropped to zero by mid-morning as expected for leaves experiencing water stress. These results show that S. alterniflora is capable of reducing osmotic potential in response to increased salinity and that turgor pressure was lower in short growth form than in tall forms.

    Notes: Summary Rachitic rats, maintained on diets with low or normal P contents, were given daily intraperitoneal doses of 1,25(OH)2D3 or 25OHD3 at levels of 100 or 200 ng. Plasma chemistry was measured and the ash content and histological appearance of the bones investigated. Using labeled material it was shown that the dosing levels of 1,25(OH)2D3 employed ensured a higher than normal plasma concentration of that metabolite over the period between doses. 1,25(OH)2D3 was not as effective as 25OHD3 in raising bone ash or reducing the amount of osteoid. The difference between the effects of the metabolites was evident at both dietary P levels, but more marked at the higher P level. In contrast, the metabolites reduced the width of the epiphyseal plate to an approximately similar degree, and this is possibly the reason why there are discrepancies between previous reports of the effectiveness of 1,25(OH)2D3 compared with 25OHD3 or vitamin D3. Dosing with 1,25(OH)2D3 failed to maintain a constant plasma Pi value over the period between doses in animals fed the low P diet.

    Notes: Summary Dichloromethylenebisphosphonate (Cl2MBP), a compound structurally related to inorganic pyrophosphate but resistant to hydrolysis of endogenous phosphatase to yield inorganic phosphate, inhibits bone resorption and soft tissue mineralizationin vivo. Previously, we have shown that bone cells isolated from rat calvaria respond profoundly to the exposure of Cl2MBP. To determine whether the cellular effects evoked by Cl2MBP are confined to a particular bone cell type, calvaria from 1 day postnatal rats were subjected to a sequential time-dependent enzyme digestion, yielding five bone cell populations marked by differences in PTH response, alkaline phosphatase activity and collagen, as well as hyaluronic acid synthesis. Culturing these bone cell populations with Cl2MBP revealed that previously observed results found with mixed bone cells (inhibition of cell proliferation, diminution of hyaluronic acid synthesis, and increase in alkaline phosphatase) were limited to cell populations which, according to the isolation scheme, stem from the outer tissue layer(s) of the calvaria. Collagen synthesis, however, was found to be equally increased regardless of cell type. These present results indicate that the action of Cl2MBP on bone may be cell specific.

    Notes: Abstract Three genetically distinct clones of Skeletonema costatum (Grev.) Cleve were grown at 20°C under high (274 μE m-2 s-1) and low (27 μE m-2 s-1) light conditions and their photoadaptive photosynthetic responses compared. When all three clones were grown under low light, pigment analyses and fluorescence excitation spectra demonstrated that the accessory pigments, chlorophyll c and fucoxanthin, became more important in light-harvesting compared to chlorophyll a. Photosynthetic unit sizes increased for Photosystems I and II in low light, but photosynthesis vs irradiance characteristics were not reliable predictors of photosynthetic unit features. Fluorescence excitation spectra and photosynthesis vs irradiance (P-I) relationships indicated that changes in energy transfer occurred independent of changes in pigment content. Large increases in accessory pigment content were not accompanied by large increases in excitation from these pigments. Changes in energy transfer properties were as important as changes in PSU size in governing the photoadaptive responses of S. costatum. When the three clones were grown under identical conditions, each had a separate and distinct pattern of photoadaptation. Significant differences among clones were found for pigment ratios, photosynthetic unit sizes for Photosystems I and II and efficiency of energy transfer between pigments. These strikingly different photoadaptive strategies among clones may partially account for the great ecological success of the diatom species. This is the first quantitative investigation of the importance of both chlorophyll c and fucoxanthin to the adaptive responses of diatoms to light intensity, and represents the most complete characterization of the photoadaptive responses of a single species of marine phytoplankter to differences in light environment.

    Notes: Abstract Human bone cells, maintained in culture, have been subjected to partitioning in an aqueous two-phase system on a countercurrent distribution apparatus. A broad cell distribution was obtained indicating cell-surface heterogeneity. Two major cell populations were identified which appeared to be growing at different rates. The ‘fast’-growing cells had a less hydrophobic cell surface than the ‘slow’-growing cells. Possible relationships of these cell populations with osteoblast differentiation and the potential importance of this technique in studies of osteoblast differentiation are discussed.
