    Description / Table of Contents: Zusammenfassung Kürzlich berichteten die Autoren über toxikologische Untersuchungen nach Aufnahme Guaifenesin-haltiger Arzneimittel. Dabei waren bei der Dünnschichtchromatographie von hydrolysierten Urinproben nach Detektion mit Marquis-Reagenz zwei intensiv violette Flecken aufgefallen. Es wird über den Gang der Identifizierung vonO-Desmethylguaifenesin undHydroxyguaifenesin mit Hilfe der Gaschromatographie-Massenspektrometrie berichtet. Über den Metabolismus von Guaifenesin beim Menschen wurde bisher nur die Oxidation zuβ-(2-Methoxyphenoxy)milchsäure mitgeteilt. Die Demethylierung von Guaifenesin wird durch die in den Lebermikrosomen enthaltene O-Demethylase durchgeführt und diese scheint Hauptmetabolisierungsenzym für Guaifenesin zu sein. Es wird auf eine pharmakologisch interessante Strukturbeziehung zwischen Guaifenesin und Codein hingewiesen.
    Notes: Abstract Recently the authors reported about toxicological investigations after uptake of guaiacol glyceryl ether containing drugs. There had been detected two intensive pink spots on the TLC from urine samples after detection with Marquisreagent. It is reported about the identification ofCatechol glyceryl ether andHydroxyguaifenesine via Gas Chromatography Mass Spectrometry. Until today only the oxidation of guaiacol glyceryl ether toβ-(2-methoxyphenoxy)lactic acid is described. The demethylation of G. is performed by O-Demethylase localized in liver microsomes. This enzyme seems to be the main enzyme for the metabolism of G. It is pointed out to a pharmacologically interesting relationship between the chemical structure of guaiacol glyceryl ether and codeine.

    Cell-specific synthesis and glycosylation of secretory proteins in larval salivary glands of Chironomus thummi (1981)

    Notes: Abstract Synthesis and glycosylation of larval salivary gland secretory proteins of Chironomus thummi were analyzed with respect to cell specific differences in the Balbiani ring (BR) pattern and glycoprotein composition of secretion formerly detected by histochemical staining procedures. In the secretion of a special cell type in salivary glands, which is characterized by the appearance of an additional BR, an additional polypeptide with a relative molecular weight (Mr) of 160 kD was found differing in its antigenic properties and tryptic fingerprint pattern from main cell secretion proteins. This so-called ssp-160 component is preferentially synthesized and glycosylated in the special cells. In the same cells, both the synthesis and glycosylation of all other major secretory proteins was found to be diminished or even repressed. In contrast to the conspicuous cell-specific differences at the level of protein synthesis, RNA analyses show the prominent synthesis of 75 S RNA in both main and special cells and gave no clear indication of the synthesis of a smaller RNA fraction as expected from the size of ssp-160 component. — These and further data on synthesis and properties of secretory proteins as well as expression of BR DNA are discussed with regard to the assumption that at least some of the eight major secretory polypeptides are coded for by BR DNA. The BR gene(s) might have originated by manifold duplications and modifications of short repetitive prototype DNA sequences, which are coordinatively expressed.

    Notes: Abstract The salivary gland cells of Chironomus larvae synthesize almost exclusively secretory proteins, for constructing the tubes in which they live. The secretory proteins of C. thummi contain a high percentage of basic amino acids, chiefly lysine (about 20%), and consist of two main giant secretory proteins of about 1,000 kd, called sp-Ia and sp-Ib, and a number of low-molecular-weight fractions, sp-180, sp-150, sp-36, sp-18, and sp-15. Galactose treatment shows that the synthesis of sp-I fractions is correlated with the puffing of Balbiani rings, BRb and BRc, and with the relative abundance of Balbiani ring RNA transcripts in the cytoplasm. The sp-I fractions, which appear to represent real polypeptides, display individual- and strain-specific size variations. Cleavage of sp-I proteins with cyanogen bromide (CNBr) yields peptides of about 17 kd and peptide ladders composed of 8–10 multimers of this 17kd “basic sequence”. These properties are typical for proteins encoded by the giant, internally repeated Balbiani ring genes.

    Notes: Zusammenfassung Organkonzentrationsmessungen an zwei Feten bestätigen die mit Lidocain im Tierversuch gemessenen hohen Leberkonzentrationen beim Menschen mit Bupivacain. Die in der Literatur bekannte und bisher nicht erklärbare Häufung von Hyperbilirubinämien beim Neugeborenen nach Epiduralanalgesie wäre hiermit erklärbar. Die durch Mehrfachuntersuchungen gesicherte exzessiv hohe Lungenkonzentration von Bupivacain in einem Fall ist bislang nicht eindeutig erklärbar. Es handelt sich um ein vorläufiges Ergebnis, weitere Untersuchungen hierüber werden folgen. Unsere Untersuchungsergebnisse sollen ein differenzierteres Verständnis des Klinikers für den Stoffwechsel bei der Epiduralanalgesie fördern.Keinesfalls soll hieraus eine Empfehlung zur Aufgabe der von vielen Aspekten her für Mutter und Fet positiven Wirkung der Epiduralanalgesie abgeleitet werden.

    Notes: Summary The knowledge of serum levels of azathioprine or its metabolite 6-mercaptopurine is important for the monitoring of the immunosuppressive therapy after kidney transplantation, especially when kidney function is low. For this reason, the work of Bailey et al. was modified. 6-Mercaptopurine was extracted from the serum with a mixture of isopropanol-ethylacetate (1:1 v:v), derivatized with trimethylaniliniumhydroxyde and measured gaschromatographically. Sulfhydryl-protecting reagents were added to prevent loss of substance. Oxyethyltheophylline was used as the internal standard. The extraction and clean-up method recovered 80% of the 6-mercaptopurine present, the lower limit of sensitivity was 20 ng/ml, i.e., ten times lower than that of Bailey et al. The method could be realized on eight patients treated with azathioprine, the half life of the drug was about 20–40 min within the first hour after i.v. application.

    Notes: Abstract It is theoretically possible that lesions generated by a Q-switched Nd-YAG laser could lead to chemical changes in polymethylmethacrylate (PMMA) intraocular lenses. These changes could be of clinical relevance. Therefore, we measured the residual monomer content of intraocular lenses before and after generation of lesions by means of a short-pulsed Nd-YAG laser. Despite the explosive destruction of the lens surface caused by this type of laser, there is no measurable release of monomer in the sense of a depolymerization of the PMMA material, even when over 100 lesions are generated. The lesions generated on intraocular PMMA lenses with the Nd-YAG laser are predominantly of a mechanical nature and include the chipping away of small particles from the intraocular lens.

    Notes: Summary Downflow fixed bed reactors operated at 35°C, were sucessfully used for the anaerobic digestion of a petrochemical effluent. COD reductions of 93–95% were found at an optimum retention time of 2.3 days and a loading rate of 4.7 kg COD/m3/d. The amount of biogas produced was 0.88 m3/m3/d (STP), with a methane content of 90–96%.
