    Notes: Summary Soluble substances of psoriatic scales (Psoexts) were analysed immunologically to detect immune complex (IC) related to the immune reactions in psoriatic lesions and compared with extracts of scales from a patient with erythroderma due to pityriasis rubra pilaris (PRP-exts) as control. Immunoelectrophoretically IgG, IgA and C3 were detected in Pso-exts, but in PRP-exts only IgG was seen as immunological reactive substances. When analysed by Sephadex G-200 column chromatography, IgG and IgA eluates were found in Pso-exts with a molecular weight of more than 200,000 daltons. By Raji cell assay, the IgG and IgA eluates were shown to have IC characteristics and they were the IC composed of IgG-IgA, which was also confirmed using anti-IgG affinity chromatography. The IC seemed to be binding the epidermal proteins as antigen. Thesefindings suggest that deposits of immunoglobulins and complement in the stratum corneum of psoriatic lesions are not just secondary to the underlying inflammation in the dermis but that the IC detected is related to the immune reactions in psoriatic lesions.

    Notes: We have previously reported the effect of hydrocortisone (HC) on the adenylate cyclase system of pig epidermis. HC had no effect on the basal level of cyclic AMP but there was an increased adrenalineinduced cyclic AMP accumulation when epidermal slices were incubated for more than 6 h with HC. This incubation did not alter the responsiveness to histamine. We are interested in the effects on epidermal cells of HC, which may act through the adenylate cyclase system. It is well documented that adrenaline, histamine and adenosine stimulate adenylate cyciase and cause an accumulation of cyclic AMP in pig epidermis. An increased cyclic AMP results in the inhibition of mitosis and epidermal outgrowth. Employing the rate of epidermal outgrowth and mitotic index as indicators of cyclic AMP effect, we studied the effect of HC on the adenylate cyclase system. The pretrcatment with HC (100 μM) magnified the inhibitory effect of adrenaline on epidermal outgrowth and mitosis, whereas the inhibitory effeet of histamine or adenosine was not affected by HC. It is suggested that HC induced an increased responsiveness of adenylate cyclase to adrenaline, resulting in a magnified inhibitory effect of adrenaline on epidermal outgrowth and mitosis.

    Notes: Using pig skin slices, we investigated the effects of hydrocortisone on the adenylate cyclase system of the skin. In short-term experiments, hydrocortisone, when added singly or in combination with other stimulators of adenylate cyclase in the skin (adrenaline or histamine), had no effect on cyclic AMP accumulation. However, when skin slices were incubated with hydrocortisone for more than 6 h, the response to adrenaline differed, with a greater accumulation of cyclic AMP in the hydrocortisonetreated skin. This effect was seen at a concentration of more than 1 μM hydrocortisone and was most marked 48 h later, while responses to adrenaline in control skin gradually decreased and remained low.Histamine, which is another stimulator of adenylate cyclase of the skin, did not cause a greater cyclic AMP accumulation in response to this hydrocortisone treatment. There was no significant difference in either low Km or high Km cyclic AMP phosphodiesterase activities as a result of this hydrocortisone treatment.Hydrocortisone seems to act by protecting the adrenaline-adenylate cyclase system of the skin.

    Notes: Summary The Kakegawa strain of bovine coronavirus was easily propagated in suckling mice. Infected animals died with nervous symptoms, and serial passage was readily accomplished by intracerebral inoculation with brain emulsions. The 3rd passage viral material from infected mice evoked the same disease in suckling mice, rats and hamsters inoculated by the intracerebral or by the subcutaneous route. Viruses recovered from mice, rats and hamsters could be clearly differentiated from mouse hepatitis virus strain 2 by the neutralization test.

    Notes: Summary A reversed passive hemagglutination (RPHA) method was developed for the detection of bovine coronavirus in fecal specimens. Sheep erythrocytes fixed with glutaraldehyde, and then treated with tannic acid were coated with anti-bovine coronavirus rabbit antibodies purified by affinity chromatography using bovine coronavirus linked to Sepharose 4B. The RPHA test was carried out by a microtiter method. Erythrocytes coated with purified specific antibodies were agglutinated by bovine coronavirus, but not by bovine rotavirus or enterovirus. The reaction was inhibited by antiserum to bovine coronavirus, confirming the specificity of the reaction. The RPHA test detected bovine coronavirus in 13 of 22 fecal specimens (59 per cent), from natural cases of diarrhea, while the positive rates were only 14 per cent (3/22) and 22 per cent (5/22) for immunofluorescent staining of primary cultures of calf kidney cells infected with the specimens, and immune electron microscopy respectively. The advantages of the RPHA method are its simplicity, high sensitivity and rapidity.

    Notes: Summary Adenosine deaminase, which catalyzes the irreversible hydrolytic deamination of adenosine and deoxyadenosine to inosine and deoxyinosine respectively, plays an important role in the degradation of adenine nucleotide and purine nucleotide salvage pathway metabolism. We investigated human epidermal adenosine deaminase activity using a radiochemical method, which enabled us to measure the adenosine deaminase activity of protein smples as small as several micrograms. We measured adenosine deaminase activity of microdissected pure epidermis of the healthy skin and the psoriatic affected and unaffected skin. It was shown that psoriatic affected epidermis had increased adenosine deaminase activity compared with the healthy epidermis (P〈0.05) and the unaffected epidermis (P〈0.01). There was no difference in enzyme activity between healthy and psoriatic unaffected epidermis. The increased adenosine deaminase activity in the psoriatic affected epidermis may reflect the accelerated salvage pathway of the nucleic acid metabolism probably associated with the hyperproliferative condition of the psoriatic epidermis.

    Notes: Abstract Partially serpentinized dunites containing small amounts of Chlorine (〈 0.5%) from Dumont, Quebec, and Horoman, Hokkaido, Japan, and one containing less than 0.05% Chlorine from Higashi-Akaishi-Yama, Ehime, Japan have been examined using the electron probe microanalyzer and scanning transmission electron microscope with X-ray analytical capabilities. Chlorine was found together with Si, Mg, Ca and Fe in the serpentine minerals of the Dumont and Hokkaido dunites but not in the Ehime dunite. Chlorine is found associated only with the most finely crystalline facies of the serpentine (grain size less than 10 nm). The Ehime dunite contained no such fine grained serpentine, and was thus effectively chlorine-free, as are the coarser grained serpentines of the other samples. The finegrained chlorine-bearing serpentine also has a much higher concentration of Fe, and can contain smaller amounts of Ca, Ni and Mn than the coarse-grained variety as well as minute awaruite (FeNi3) grains. This fine-grained serpentine probably represents an early stage in the transformation of olivine to serpentine, with chlorine from hydrothermal solutions assisting the necessary chemical changes. The Cl increases the reaction rate by lowering the activation barrier to reaction by the introduction of reaction steps.

    Notes: Summary Effects of α-adrenoceptor stimulation by phenylephrine (10−7–10−4 mol/l) in the presence of pindolol (10−7 mol/l) on the action potential and force of contraction were observed in the rabbit left atrium. 1. Phenylephrine reduced resting potential, prolonged action potential duration (APD), decreased maximum rate of rise of action potential ( $$\dot V_{{\text{max}}}$$ ) and increased force of contraction in a concentration-dependent manner. 2. Effects of phenylephrine (10−5 mol/l) were antagonized by prazosin (10−7 mol/l) or phentolamine (10−6 mol/l). 3. APD which had been prolonged by phenylephrine (10−5 mol/l) was slightly shortened by Ni2+ (0.3 mmol/l) at the level of 50% repolarization but almost unaffected at the level of 90% repolarization. TTX (10−6 mol/l) had no effect on ADP which had been prolonged by phenylephrine (10−5 mol/l). 4. Cs+ (10 mmol/l), which inhibits outward current i k 1, depolarized resting potential, but in contrast with phenylephrine Cs+ did not affect APD at 90% repolarization. 5. Phenylephrine (10−5 or 3×10−5 mol/l) restored Ca2+-dependent action potential and force of contraction in 20 mmol/l K+-Tyrode solution. These responses were suppressed by prazosin (10−7 mol/l) or Ni2+ (0.3 mmol/l). 6. Phenylephrine (10−5 or 10−4 mol/l) had no effect on steady-state membrane potential — $$\dot V_{{\text{max}}}$$ relationship. It is concluded that in the rabbit left atrium phenylephrine, via α-adrenoceptors may suppress outward currents perhaps i k 1 and i x, and might increase slow inward current.
