    Notes: Summary Ultrastructural immunostaining of carbonic anhydrase in gastric parietal cells was accomplished with the immunoglobulin-peroxidase bridge procedure applied to cryostat sections of fixed guinea-pig stomach prior to dehydration and embedment. Of a variety of fixatives tested, only freshly prepared paraformaldehyde buffered with calcium acetate provided both immunostaining and adequate preservation of ultrastructural morphology. Delipidization or exposure of specimens to detergent prior to staining enhanced the intensity of the immunostaining and increased the sensitivity of the method. Increased diaminobenzidine concentration in the peroxidase substrate appeared also to intensify the densification at the reactive site. Carbonic anhydrase was localized ultrastructurally with this pre-embedment immunobridge procedure in the hyaloplasm of gastric parietal cells and less consistently in the superficial surface epithelium. The basal portion of the parietal cells stained more intensely than the apical region and immunoreactivity appeared concentrated at the plasmalemma and around mitochondria.

    Notes: Summary The ultrastructural densification of mitochondria by the high iron diamine (HID) method has been investigated by staining gastric parietal cells with each component of the HID reagent, alone or in combination, or with an otherwise modified HID solution. The effect of certain chemical treatments prior to staining has also been assessed. These tests provided evidence for at east three cytochemically distinguishable constituents in the mitochondrial matrix. In addition, the results from these tests and observations obtained with a newly introduced diaminobenzidine (DAB)-FeCl3 staining reagent indicated that the probable mode of action of the HID method in imparting density to mitochondrial matrix entails combination between a complex of iron with polymerized or aggregatedm- andp-diamine and some matrix constituent. Assessment of the cytochemical staining in varied fixation condition revealed that the lucent population of mitochondria recently described in HID-stained parietal cells reflects a failure to stain with the method because of inadequate fixation. The possible nature of the HID-reactive substance in the mitochondrial matrix is discussed in the light of these observations and other cytochemical reactivities.

    Notes: Summary The ultrastructural localization of peanut lectin-binding sites in the gastric surface epithelial cell has been studied using a horseradish peroxidase-labelled peanut lectin (PL-HRP) conjugate and other cytochemical techniques. The PL-HRP procedure has visualized glycoprotein with presumed terminal galactose residues in the apical plasmalemma and secreted mucins and has localized such glycoprotein selectively in the intermediate Golgi cisternaé situated between the saccules of the maturing face and those of the forming face of the Golgi stacks. Other cytoplasmic organelles, including the forming and stored secretory granules, did not reveal glycoprotein with terminal galactosyl residues. These results demonstrate the applicability of the PL-HRP labelling technique at the electron microscopical level to localize not only extra but also intracellular peanut lectin-reactive sites. The observations afford information concerning the possible site in the Golgi apparatus where galactose residues are added to the growing oligosaccharide side-chain of mucous glycoprotein.

    Notes: The distribution of complex carbohydrates has been investigated cytochemically at the light and electron microscope levels in collecting ducts of the guinea pig kidney. The dialyzed iron method demonstrated acidic complex carbohydrate ultrastructurally on the outer surface of the apical and the basolateral plasmalemma of the principal cells and in their maturing Golgi cisternae and secretory granules. Glycoconjugate in these sites stained for sulfate esters with the high iron diamine method but lacked reactivity toward the periodic acid-thiocarbohydrazide-silver proteinate (PA-T-SP) sequence for visualizing vicglycol-containing glycoprotein. Lability to testicular hyaluronidase and resistance to sialidase identified the Glycosaminoglycan (GAG) in principal cell granules and the plasmalemmae as a chondroitin sulfate. In contrast, intercalated cells of the collecting ducts failed to stain with the cationic reagents, but showed light PA-T-SP reactivity demonstrative of neutral glycoprotein in the glycocalyx of the apical plasmalemma. Immunostaining with the immunoglobulin-enzyme bridge procedure localized carbonic anhydrase selectively to the intercalated cells. The ultrastructural and cytochemical observations on the guinea pig collecting ducts implicate intercalated cells in fluid and electrolyte transport and principal cells in secretion of a chondroitin sulfate to the tubule lumen and intercellular space.

    Notes: Ultrastructural and complex-carbohydrate cytochemical studies were carried out on guinea pig gastric mucosa to assess the histochemical properties of the secretions of different gastric epithelial cells and to investigate the differentiation, origin, and renewal of certain cell types. The observations disclosed heterogeneity or variability of the secretory granules within individual mucigenic cells and zymogen cells. The cytochemical methods also served in characterizing and distinguishing five cell types in the gastric glands, including the isthmus cell, a mucous cell considered comparable to the mucous neck cell, the chief cell, and forms transitional between the isthmus and mucous cells and the mucous and zymogenic cells. The several cell types differed widely in the cytochemical properties of the secretory granules and the apical plasmalemma, and each had a distinctive distribution in the gastric gland. Cytochemical staining observed here provided evidence on synthesis and intracellular metamorphosis of mucous droplets and on formation of glycocalyx.
